Pilzinfektionen sind geworden ein wichtiges medizinisches Anliegen in den letzten Jahrzehnten haben erheblich zugenommen, vor allem auf einen Anstieg der Zahl von immungeschwächten Personen wie diejenigen in der Krebsbehandlung und Menschen mit HIV/AIDS oder transplantierten Organe1,2. Jedoch beitragen eine sehr begrenzte Auswahl an verfügbaren Antimykotika und die zunehmende Zahl von Berichten über Pilzresistenz dazu zu den großen Problemen bezüglich der Therapie der systemischen Mykosen3.
Eine potenzielle Quelle für neue antimykotische Substanzen sind antimikrobielle Peptide (AMPs), kleine kationischen Peptide, die von vielen Organismen als Bestandteil ihrer angeborenen Immunantwort auf eine Infektion4produziert. Die Screening-Methode, diese Verbindungen gegen Pilzerreger zu testen ist jedoch nicht standardisiert. Verschiedene Verfahren wurden zur antimykotische Aktivität des AMPs, manchmal für das gleiche Modell Mikroorganismus5,6,7zu beurteilen. Diese Unterschiede und den Mangel an Details in einigen Protokollen erschweren Vergleiche zwischen Verbindungen und behindert Reproduzierbarkeit.
Eine Möglichkeit, die Prüfung von neuen Medikamenten-Kandidaten zu standardisieren soll pilzbefallverhütende Anfälligkeit in klinischen Umgebungen, wie z. B. die klinische Definition verwendet und Laboratory Standards Institute (CLSI) M27-A3 Leitlinien folgen. Jedoch diese antimykotische Sensitivitätstests sind zu restriktiv, und nehmen nicht in Betracht-Variante im Stoffwechsel über Arten, da sie nur für wenige ausgewählte Agenten errichtet wurden. Zum Beispiel nehmen sie nicht zu berücksichtigen die metabolischen Bedürfnisse nicht gärenden Hefen.
Dieses Protokoll ermöglicht die Beurteilung der Tätigkeit des Interessenten antimykotische Substanzen und hier für die Suche nach antimykotische Peptide eingeführt. Es basiert auf der Brühe Microdilution Empfindlichkeitstest aus der CLSI M27-A3-Richtlinien mit Modifikationen, die das Screening von neuen Verbindungen8,9zu optimieren. Diese Änderungen ermöglichen den Einsatz von geringen Mengen an Substanz, Temperatur- oder erste Inokulum und verschiedene Medien für ein optimales Wachstum der Pretest, während die Ergebnisse anhand von Referenz-Antimykotika als Steuerelemente Standardisierung. Diese Methode, mit der Verwendung von Multi-gut Kultur Platten ermöglicht es schnell und zuverlässig eine große Anzahl von Verbindungen auf den Bildschirm.
Aufgrund seiner Flexibilität kann dieses Protokoll mit unterschiedlichen chemischen Klassen von Verbindungen und gegen andere Mikroorganismen, mit wenigen Anpassungen verwendet werden.

Pilzinfektionen sind in den letzten Jahrzehnten eine wichtige medizinische Voraussetzung geworden, aber die Anzahl der verfügbaren Antimykotika ist begrenzt. In diesem Szenario ist die Suche nach neuen Antimykotika notwendig. Das Protokoll berichtet hier beschreibt eine Methode, um Bildschirm Peptide für ihre antimykotischen Eigenschaften. Es basiert auf der Microdilution-Empfindlichkeitstest Brühe aus den Clinical and Laboratory Standards Institute (CLSI) M27-A3-Richtlinien mit Modifikationen an der Erforschung der antimikrobielle Peptide als potenzielle neue Antimykotika. Dieses Protokoll beschreibt einen funktionelle Assay zur Beurteilung der Aktivität von Antimykotika Verbindungen und kann leicht geändert werden, um einer bestimmten Klasse von Molekülen unter Untersuchung zu entsprechen. Dieses Verfahren wird veranschaulicht, wie ein standardisierte und einstellbare klinisches Protokoll die Bank-Arbeit Verfolgung neuer Moleküle, die Therapie von Pilzerkrankungen zu verbessern helfen kann.

Das MIC ist definiert als die niedrigste antimikrobielle Compoundkonzentration, die vollständig sichtbare Pilzwachstum am Ende der Inkubationszeit hemmt.
