Ces dernières décennies ont vu le développement explosif du champ de photoacoustique biomedical imagerie1,2,3,4,5,6,7 ,,8. Basé sur la conversion de l’énergie de la lumière en son, l’imagerie photoacoustique émergents peuvent visualiser des échantillons biologiques à échelles des organelles, cellules, tissus, organes de petits animaux tout le corps et peut révéler sa anatomiques, fonctionnelles, moléculaires, génétiques, et information métabolique1,2,9,10,11,12. L’imagerie photoacoustique a trouvé des applications dans une gamme de domaines biomédicaux, comme cell biology13,14, biologie vasculaire15,16, neurologie17,18 , oncologie19,20,21,22,23de la dermatologie, pharmacologie24et hématologie25,26. Son application en ophtalmologie, c'est-à-dire photoacoustique oculaire imaging, a attiré des intérêts substantiels à la fois scientifiques et cliniciens et est actuellement à l’étude active.
Contrairement aux couramment utilisé oculaire d’imagerie technologies27, tels que l’angiographie à la fluorescéine (FA) et angiographie de vert d’indocyanine (ICGA) (basé sur le contraste de fluorescence), tomographie à cohérence optique (OCT) (basé sur le contraste de la dispersion des optiques) et ses dérivé angiographie OCT (basé sur le contraste de la motion des globules rouges), photoacoustique oculaire d’imagerie d’absorption optique utilise le mécanisme de contraste. Cela diffère des technologies d’imagerie oculaires classiques et offre un outil unique pour l’étude des propriétés d’absorption optique de le œil, qui sont généralement associées à l’État physiopathologique des tissus oculaires28. À ce jour, significative, excellent travail a été fait dans photoacoustique oculaire d’imagerie29,30,31,32,33,34,35, 36,37, mais ces études se concentrent sur le segment postérieur des yeux des petits animaux, comme les rats et les souris. Le pionnier dans l’étude bien démontre la faisabilité de l’imagerie photoacoustique en ophtalmologie, mais il y a encore un long chemin à parcourir vers la traduction clinique de la technologie depuis tailles de globe oculaire des rats et des souris sont beaucoup plus petits (moins d’un tiers) à celle de l’homme. En raison de la propagation des ondes ultrasoniques sur une beaucoup plus longues distances, signal intensité et qualité d’image peut grandement souffrir lorsque la technique est utilisée pour l’imagerie du segment postérieur de plus grands yeux.
Pour atteindre cet objectif, nous avons récemment rapporté les non invasive, imagerie exempte d’étiquette Chorio-rétinienne chez les lapins vivants à l’aide intégrée microscopie photoacoustique (PAM) et le domaine spectral OCT (SD-OCT)38. Le système a d’excellentes performances et peut visualiser la rétine et la choroïde des yeux des animaux plus gros absorption endogène et contrastes de diffusion des tissus oculaires. Chez les lapins, les résultats préliminaires montrent que la PAM pourrait distinguer non invasive de vaisseaux sanguins rétiniens et choroïdienne individuels à l’aide d’une dose d’exposition laser (80 ~ nJ) significativement inférieures à la limite de sécurité American National Standards Institute (ANSI) (160 nJ) à 570 nm39; et l’outil OPO pourrait résoudre clairement la sclérotique, la choroïde et différentes couches rétiniennes. C’est la toute première démonstration de l’imagerie du segment postérieur de plus grands animaux à l’aide du PAM et pourrait être une étape importante vers la traduction clinique de cette technologie étant donné que la taille du globe oculaire de lapins (18,1 mm)40 est près de 80 % de la longueur axiale de humains (23,9 mm).
Dans ce travail, nous fournir une description détaillée du système dual-modalité d’imagerie et des protocoles expérimentaux utilisés pour l’imagerie non invasive et sans étiquette Chorio-rétinienne chez les lapins vivants et démontrer les performances du système par le biais de rétine représentatif et résultats de l’imagerie choroïdienne.

novel photoacoustic microscopy and optical coherence tomography dual-modality chorioretinal imaging in living rabbit eyes

Photoacoustique imagerie oculaire est une émergence ophtalmique technologie qui peut visualiser une façon non envahissante de tissus oculaires en convertissant l’énergie lumineuse en ondes sonores et est actuellement à l’étude intensive de l’image. Cependant, les plus signalés à ce jour se concentre sur l’imagerie du segment postérieur des yeux des petits animaux, comme les rats et les souris, qui pose des défis pour la traduction humaine clinique en raison des tailles petit globe oculaire. Ce manuscrit décrit une microscopie photoacoustique roman (PAM) et le système de double-modalité de tomographie par cohérence optique pour l’imagerie du segment postérieur des yeux des grands animaux, tels que des lapins. La configuration du système, alignement du système, préparation animaux et dual-modalité protocoles expérimentaux pour in vivo, non invasive et sans étiquette Chorio-rétinienne d’imagerie chez les lapins sont détaillées. L’efficacité de la méthode est démontrée par les résultats expérimentaux représentatifs, dont la vascularisation rétinienne et choroïdienne obtenue par le PAM et les PTOM. Ce manuscrit fournit un guide pratique pour reproduire les résultats de l’imagerie chez le lapin et l’avancement photoacoustique imagerie oculaire chez les animaux plus gros.

Le système dual-modalité d’imagerie et le protocole expérimental ont été testés avec succès en laboratoire des auteurs à l’aide de quatre lapins néo-zélandais blancs. Ce qui suit présente quelques résultats représentatifs.
