Últimas décadas têm testemunhado o desenvolvimento explosivo do campo da biomédica fotoacústico imagem1,2,3,4,5,6,7 ,8. Baseia-se a conversão de energia da luz, em som, a imagem de fotoacústico emergentes pode Visualizar amostras biológicas em escalas de organelas, células, tecidos e órgãos para pequenos animais de corpo inteiro e pode revelar sua anatômica, funcional, molecular, genética, e informações metabólicas1,2,9,10,11,12. Imagem latente fotoacústico encontrou aplicações únicas em uma variedade de campos biomédicos, como célula biologia13,14, biologia vascular15,16,,Neurologia1718 , oncologia19,20,21,22, Dermatologia23, farmacologia,24e25,de hematologia26. Sua aplicação em oftalmologia, ou seja, fotoacústico ocular imaging, tem atraído interesse substancial de cientistas e clínicos e é atualmente sob investigação.
Em contraste com a usada rotineiramente ocular de imagem tecnologias27, tais como a angiografia fluoresceína (FA) e angiografia indocianina verde (ICGA) (baseado no contraste de fluorescência), tomografia de coerência óptica (OCT) (baseado no contraste de dispersão óptica) e seus derivada angiografia de OCT (baseado no contraste do movimento das células vermelhas do sangue), ocular fotoacústico absorção óptica de usos como o mecanismo de contraste de imagem. O sistema tem excelente desempenho e pode visualizar a retina e a coroide dos olhos de animais maiores, com base na absorção endógena e contraste de espalhamento de tecido ocular. Os resultados preliminares em coelhos mostram que o PAM canaliza poderia distinguir individuais vasos sanguíneos da retina e da coroide usando uma dose de exposição do laser (~ 80 nJ) significativamente abaixo do limite de segurança de American National Standards Institute (ANSI) (160 nJ) no 570 nm,39; e a OCT claramente poderia resolver diferentes camadas da retina, coroide e a esclera. É a primeira demonstração da imagem latente de segmento posterior de animais maiores usando PAM e pode ser um passo importante para a tradução clínica da tecnologia considerando-se que o tamanho do globo ocular de coelhos (18,1 mm)40 é quase 80% do comprimento axial do seres humanos (23,9 mm).

novel photoacoustic microscopy and optical coherence tomography dual-modality chorioretinal imaging in living rabbit eyes

Imagem de ocular fotoacústico é um emergente oftálmica tecnologia que canaliza pode visualizar o tecido ocular, convertendo energia luminosa em ondas sonoras e está atualmente sob investigação intensiva de imagem. No entanto, a maioria relatou que trabalho até à data é focado sobre a imagem do segmento posterior dos olhos de pequenos animais, como ratos e camundongos, o que coloca desafios clínico tradução humana devido a tamanhos pequeno globo ocular. Este manuscrito descreve um romance fotoacústico microscopia (PAM) e o sistema de dual-modalidade de tomografia computadorizada (OCT) de coerência óptica para a imagem latente de segmento posterior dos olhos de animais maiores, tais como coelhos. A configuração do sistema, alinhamento de sistema, preparação de animais e dual-modalidade protocolos experimentais para vivo em, não invasivo, livre de rótulo choriorétiniennes imagem em coelhos são detalhados. A eficácia do método é demonstrada através de resultados experimentais representativos, incluindo a vasculatura da retina e da coroide, obtida pelo OCT e PAM. Este manuscrito fornece um guia prático para reproduzir os resultados de imagem em coelhos e avançando fotoacústico imagem ocular em animais maiores.
