Senaste decennierna har bevittnat den explosiva utvecklingen av området för biomedicinsk photoacoustic imaging1,2,3,4,5,6,7 ,8. Baserat på energi omvandlingen av ljus till ljud, den framväxande photoacoustic imaging kan visualisera biologiska prover på skalor från organeller, celler, vävnader och organ till små djur hela kroppen och kan avslöja dess anatomiska, funktionella, molekylär, genetisk, och metaboliska information1,2,9,10,11,12. Photoacoustic imaging har funnit unika tillämpningar i en rad biomedicinska forskningsområden, exempelvis cellbiologi13,14, vaskulärbiologi15,16, neurologi17,18 , onkologi19,20,21,22, dermatologi23, farmakologi24och hematologi25,26. Systemet har utmärkta prestanda och kan visualisera näthinnan och åderhinnan ögonen på större djur baserat på endogena absorption och spridning kontrasten i okulära vävnader. Preliminära resultat i kaniner visar att PAM noninvasivt kunde urskilja enskilda blodkärl på näthinnan och koroidal som använder en laser exponeringsdos (~ 80 nJ) betydligt under ANSI American National Standards Institute () säkerhet gräns (160 nJ) på 570 nm39; och ULT kunde tydligt lösa olika retinala lagren, åderhinnan och sklera. Det är den första demonstrationen av bakre segment avbildning av större djur med PAM och kan vara ett stort steg mot klinisk översättning av tekniken med tanke på att ögongloben storleken på kaniner (18,1 mm)40 är nästan 80% av axiell längd människor (23,9 mm).
I detta arbete, vi ger en detaljerad beskrivning av dual-modalitet bildsystem och experimentella protokoll som används för icke-invasiv, etikett-fri korioretinal imaging i levande kaniner och visar systemets prestanda genom representativa retinal och koroidal bildbehandling resultat.

novel photoacoustic microscopy and optical coherence tomography dual-modality chorioretinal imaging in living rabbit eyes

Photoacoustic okulär bildbehandling är en framväxande oftalmologiska bildteknik som noninvasivt kan visualisera okulär vävnad genom att omvandla ljusenergi till ljudvågor och är för närvarande under intensiv undersökning. De flesta rapporterade dock hittills är inriktad på avbildning av bakre segment ögonen på små djur, som råttor och möss, som innebär utmaningar för kliniska mänsklig översättning på grund av små ögongloben storlekar. Detta manuskript beskriver en roman photoacoustic mikroskopi (PAM) och optisk koherens tomografi (ULT) dual-modalitet system för bakre segment avbildning av ögonen av större djur såsom kaniner. Systemkonfiguration, system justering, djur förberedelse och dual-modalitet experimentella protokoll för i vivo, icke-invasiv, etikett-fri korioretinal imaging hos kaniner är detaljerade. Effektiviteten i metoden demonstreras genom representativa experimentella resultat, inklusive näthinnan och koroidal vaskulatur erhålls av PAM och OCT. Detta manuskript ger en praktisk guide till reproducera de bildbehandling resultat hos kaniner och främja photoacoustic okulär imaging i större djur.

Den dubbla-modalitet bildsystem och experimentellt protokoll har framgångsrikt testats i författarnas laboratorium med fyra nya Zeeland vita kaniner. Följande visar upp några representativa resultat.
