1. 为鉴别化验准备陶瓷
<ol><li>在无菌组织培养罩中制备组织培养试剂和处理活细胞。</li>
	<li>制备 A0 培养基: Dulbecco 改性鹰中、火腿 F12 培养基 (DMEM/F12)、1x glutamax、1x 青霉素/链霉素。</li>
	<li>准备完整的 ASC 培养基: DMEM/F12, 1x glutamax, 1x 青霉素/链霉素, 10% 胎牛血清。</li>

	<li>将200µL 的细胞悬浮液转移到 microwell 板上, 以达到每井 5 x 103单元的最终细胞密度。每个治疗组需要四口水井 (三项技术重复和免疫细胞化学 (ICC) 的无主抗体控制)。</li>

</ol>2. 诱导分化陶瓷
<ol><li>用1µM dexamethasome、10µM 胰岛素和200µM 吲哚美辛补充完整的 ASC 培养基, 制备脂肪分化培养基。完全分化培养基稳定在4°c 1 月, 所以只做1的化验要求。存储在4°c 和当需要加热整除到37°c 在水浴。</li>

</ol>3. 鉴别染色-传统染料基污渍
<ol><li>用 PBS 稀释16% 甲醛, 制备4% 甲醛溶液。仅稀释实验所需的用量, 并在离心管中密封牢固。 注意: 甲醛是一种潜在的致癌物质, 在吸入时会引起鼻子、喉咙和肺部的刺激。执行与油烟机内甲醛有关的所有过程。</li>
	<li>在100毫升异丙醇中溶解300毫克, 制备油红色 O 型溶液。</li>
	<li>把 microwell 盘子从孵化器里拿出来。以下步骤可在无菌环境中执行。</li>
	<li>从井中取出所有介质, 用4% 甲醛固定细胞30分钟。</li>
	<li>在孵化时间, 准备油红色 O 工作解决方案。工作解决方案包括6件油红色 O 型股, 4 份蒸馏水 (dH2o)。混合好, 让坐在室温下20分钟, 过滤前使用0.2 µm 过滤器单元。工作解决方案只稳定2小时。</li>
	<li>吹打从水井中取出所有甲醛。</li>
	<li>用60% 异丙醇洗涤细胞, 并在继续前让水井完全干燥。</li>
	<li>添加50µL 的油红 O 工作溶液, 每井和孵化在室温下10分钟。</li>

</ol>4. 鉴别基因特异抗体的染色
<ol><li>通过溶解80克氯化钠、2克氯化钾和30克三基到 dH2O (pH 8) 的1升, 制备三缓冲盐水 (tb) 的库存溶液。使用 dH2o 在室温下存储, 以稀释1:10 来准备工作溶液。</li>
	<li>准备 immunobuffer (IB) (1% 胎牛血清 (血清) 在 TBS)。标签15毫升离心管与日期和存储在4摄氏度。IB 可用于1星期从日期。</li>
	<li>用200毫升磷酸盐缓冲盐水 (PBS) 溶解0.5 克酪蛋白和0.195 克叠氮化钠, 制备0.25% 酪蛋白阻滞剂。通过搅拌过夜, 完全溶解成分。整除15毫升离心管, 并贮存在-20 摄氏度的冷冻机中。解冻酪蛋白溶液可保持在4°c 1 月。</li>
	<li>通过稀释 DAPI 1:200 的 DAPI 溶液, 将其储存在4摄氏度, 免受光照。</li>

	<li>选择要图像的水井。</li>

</ol>6. 分析获取的图像

visualization and quantification of mesenchymal cell adipogenic differentiation potential with a lineage specific marker
