Ein gut etabliertes Protokoll für decellularized ECM-Herstellung sollten die native ECM Zusammensetzung beim Entfernen von zellulären Bestandteilen in Geweben behalten fast komplett21. Trotz aktuell gemeldete Decellularization Protokolle erfordern Perfusion durch das Gefäßsystem, zellulare Werkstoffe durch konvektiven Transport entfernen, mechanische Agitation hier, wurde bekannt als eine traditionelle einfache und preiswerte Methode22 , 23<...

Die Zunge hat verschiedene wichtige Funktionen wie Schlucken, Artikulation und Verkostung. Die Beeinträchtigung der Funktion der Zunge hat so großen Einfluss auf Patienten Lebensqualität1. Die häufigsten malignen Erkrankungen in der Mundhöhle ist Zunge Squamous Zelle Krebsgeschwür (TSCC), die in der Regel tritt bei Menschen, die Alkohol trinken oder Rauchen Tabak2.
In den letzten Jahren wurden in der Grundlagenforschung auf TSCC wenig Fortschritte erzielt. Der Mangel an effizienter in-vitro- Forschung Modelle bleibt eines der größten Probleme sein. So entpuppt sich die extrazelluläre Matrix (ECM) eine mögliche Lösung sein. Da ECM ein komplexes Netzwerk Frame hochorganisierte Matrix Komponenten zusammengesetzt ist, wäre eine ECM-artige Struktur und Zusammensetzung Gerüstmaterial zuständigen für die Krebsforschung. Decellularized ECM bieten perfekt die Nische für die Zellen aus der gleichen Herkunft in Vitro, die entpuppt sich als der wichtigste Vorteil von ECM.
ECM kann mit zellulären Komponenten aus dem Gewebe durch die Decellularization mit Wasch- und Enzyme entfernt wird einbehalten. Verschiedenen ECM-Komponenten, einschließlich Kollagen, Fibronektin und Laminin in decellularized Matrix bieten eine Native-Gewebe-Like Mikroumgebung für kultivierten Zellen, fördert das Überleben, Proliferation und Differenzierung von Zellen3. Darüber hinaus kann die Immunogenität zur Transplantation auf einem minimalen Niveau mit der Abwesenheit von zellulären Bestandteilen in ECM reduziert werden.
Bisher sind Fertigungsverfahren für decellularized ECM in verschiedenen Geweben und Organen, wie Herz4,5,6,7, Leber8,9,10 versucht worden ,11, Lunge12,13,14,15,16,17und Niere18,19 , 20. keine relevanten Forschung hat jedoch auf ähnliche Arbeiten in der Zunge nach bestem Wissen und gewissen gefunden werden.
In dieser Studie wurde decellularized Zunge extrazelluläre Matrix (TEM) durch eine Reihe von physikalischen, chemischen und enzymatischen Behandlung effizient und kostengünstig hergestellt. Dann wurde die TEM zur TSCC in Vitrorekapitulieren zeigt eine entsprechende Simulation für TSCC Verhalten und Entwicklung. TEM hat gute Biokompatibilität sowie die Fähigkeit, die Zellen der Gewebe-spezifische Nische zu führen gibt an, dass TEM ein großes Potenzial in TSCC Forschung3haben kann. Die hier gezeigte Protokoll bietet eine Auswahl für Forscher auf Pathogenese oder Therapien von TSCC.

Um eine effektive und realistische Modell für Zunge Plattenepithelkarzinom Karzinom (TSCC) in Vitrozu konstruieren, die Methoden zu produzieren decellularized Zunge extrazelluläre Matrix (TEM) entstanden funktionelle Gerüste für TSCC Bau bietet. TEM bietet eine in-vitro- Nische für Zellwachstum und Differenzierung Zellmigration. Die Mikrostrukturen der native extrazelluläre Matrix (ECM) und biochemische Kompositionen aufbewahrt in der decellularized Matrix bieten gewebespezifischen Nischen für die Verankerung von Zellen. Die Herstellung von TEM kann durch Deoxyribonuclease (DNase) Verdauung begleitet mit einer schweren organischen oder anorganischen Vorbehandlung realisiert werden. Dieses Protokoll ist einfach zu bedienen und sorgt für hohe Effizienz für die Decellularization. Die TEM zeigte günstige Cytocompatibility für TSCC Zellen unter statischen oder gerührt Kulturbedingungen, ermöglicht den Bau des Modells TSCC. Ein self-made-Bioreaktor wurde auch für die anhaltende gerührt Bedingung für die Zellkultur verwendet. Rekonstruierte TSCC mit TEM zeigte die Merkmale und Eigenschaften ähnlich klinische TSCC Histopathologie, was darauf hindeutet das Potenzial in der TSCC Forschung.

Dieses Protokoll für die Vorbereitung der TEM entpuppt sich als effizient und angemessen. Die TEM zeigte perfekte Decellularization im Vergleich mit Muttersprache Gewebe.
