Citrus greening, noto anche come huanglongbing, ha causato una perdita significativa di alberi di agrumi. Un caso rappresentativo è Florida, dove la malattia è prevista per causare una riduzione del 70% nella produzione di scatole di Florida arancione da scatole di 244 milioni nella stagione 1997-1998 a caselle 70 milioni a partire dal 2016-2017 stagione1. Oltre il 90% degli alberi di agrumi in Florida sono infetti2, e si stima che l'industria dell'agrume di Florida perde circa 1 miliardo di dollari ogni anno a causa di malattie degli agrumi, in cui citrus greening gioca un ruolo importante3. Citrus greening è trasmesso principalmente per la dilagante psyllid agrume asiatico Diaphorina citri, ma può essere trasmessa anche mediante innesto di tessuto infetto. Il DNA genomico è estratto dal tessuto foglia intera, che contiene i batteri vivi. Questo DNA genomico estratto viene utilizzato come un modello per PCR convenzionale, dove oligonucleotidi complementari alla sequenza del rDNA 16S del batterio sono utilizzati per amplificare questa sequenza. Oligonucleotidi complementari ad un gene FBOX agrumi sono utilizzati come un controllo di amplificazione interna. Abbiamo scelto di utilizzare questo metodo, perché essa ha dimostrato di avere successo nella precedente ricerca6.
Questo metodo ha il chiaro vantaggio di essere semplice, relativamente poco costoso e in grado di essere eseguita in qualsiasi laboratorio di biochimica normalmente attrezzata. Inoltre, PCR rimane il metodo più accurato e preciso per la rilevazione di questo agente patogeno, a causa della difficoltà di coltura di questo agente patogeno7e capacità del patogeno di sopravvivere e riprodursi per anni all'interno di un host asintomatico8. Molte specialità diverse analisi di PCR sono state usate per rilevare correttamente questo agente patogeno; Tuttavia, PCR convenzionale rimane il più semplice test per la rilevazione accurata e specifica, soprattutto all'interno di host asintomatico8.

Citrus greening, noto anche come huanglongbing, è una malattia distruttiva agrumi devastando agrumeti a livello globale. Questa malattia provoca asimmetrico foglia giallo chiazzatura, vena ingiallimento, defogliazione, decadimento di radice e in ultima analisi, la morte della pianta di agrumi. Quando infetta, le piante di agrumi hanno stentata crescita e producono fiori fuori stagione.
