Sitrus greening, også kjent som huanglongbing, har forårsaket betydelig tap av sitrus trær. En representant saken er Florida, der sykdommen er forecasted for å føre en 70% reduksjon i produksjonen av Florida oransje bokser fra 244 millioner bokser i 1997-1998-sesongen å 70 millioner bokser i 2016-2017 sesong1. 90% av sitrus trær i Florida er over infisert2, og det er anslått at Florida sitrus næringen taper ca en milliard dollar hvert år på grunn av sitrus sykdommer, der sitrus greening spiller en viktig rolle3. Formålet med denne metoden er å nøyaktig og identifisere nøyaktig motile bakterie Candidatus liberibacter spp., den utløsende agenten for sitrus greening, bor i phloem av infiserte sitrustrær5. Genomic DNA er Hentet fra hele blad vev, som inneholder live bakterier. Dette utdraget genomisk DNA brukes som mal for konvensjonelle PCR, hvor oligonucleotides utfyllende til bakteriens 16S rDNA sekvens brukes til å forsterke denne sekvensen. Oligonucleotides komplementær til en sitrus FBOX genet brukes som en intern forsterkning kontroll. Vi valgte å bruke denne metoden, fordi det har vist seg for å være vellykket i tidligere forskning6.
Denne metoden har klar fordel av å være enkel, relativt billig og kan utføres i alle normalt utstyrt biokjemi lab. I tillegg fortsatt PCR mest nøyaktig og presis metoden for å oppdage denne patogen, på grunn av vanskelighetene med dyrking denne patogen7og den patogene evne til å overleve og reprodusere i år i en asymptomatisk vert8. Mange forskjellige spesialitet PCR analyser har blitt brukt til å oppdage denne patogen; konvensjonelle PCR imidlertid enkleste analysen for nøyaktig og bestemt deteksjon, spesielt innen asymptomatisk verter8.

Sitrus greening, også kjent som huanglongbing, er en destruktiv sitrus sykdom herjer sitrus gårder globalt. Denne sykdommen fører asymmetrisk gult blad mottling blodåre gulfarging, den høye saltholdigheten, rot forfall og til slutt død av sitrus anlegget. Denne metoden er enkel, effektiv, kostnadseffektiv og tilpasses kvantitativ analyse. Denne metoden kan tilpasses for bruk på alle sitrus vev; men er det potensielt begrenset av mengden patogen i vevet. Denne metoden vil likevel tillate sitrus bønder å identifisere infiserte sitrus planter tidligere og dempe spredningen av denne ødeleggende sykdommen før det kan videre spres.

Et positivt resultat gir en tydelig band tilsvarer 500 bp Candidatus Liberibacter asiaticus Las606/Lss6, og/eller en distinkt band tilsvarer 700 bp for Laa2/Laj5, 16S ribosomal β bilag operon9.
