Avgjørende skritt av kirurgiske prosedyren er å finne riktig inngangspunkt for skruen implantasjon i femur-Kondyler på intercondylar hakket samt optimal retningen på nålen parallelt med bein aksen for brotsjing av den mulig intramedulær hulrom. For å unngå en feil oppføring posisjon, bør kirurgen forberede hakket til en optimal visning er oppnådd. Bestemme retningen under brotsjing, bør femur av mus holdes med fingrene i en stabil posisjon. Bein helbredende studier i mus er svært etterspurt et bredt spekter av antistoffer og genetisk endret dyr. Disse fakta kan studere molekylære mekanismer av bein healing1. I de siste årene utviklet ulike bein healing modeller for mus har vært2. Disse modellene kan deles inn i åpne modeller, som benet er osteotomized med en åpen lateral kirurgisk tilnærming og lukket modeller, der benet er brukket basert på brudd modell introdusert av Bonnares og Einhorn3. Bruker denne teknikken, standardisert tverrgående brudd kan produseres av en 3-punkts bøying enhet og mulig intramedulær implantater kan settes gjennom en liten mediale parapatellar snitt i en minimalt invasiv teknikk unngå en stor bløtvev traumer.
Mulig intramedulær skruen kan brukes for lukket brudd stabilisering i mus. Skruen tilbyr roterende og aksial stabilitet. Dette oppnås ved brudd komprimering gjennom en proksimale tråd og distale hodet4. Ytterligere fordeler av skruen er enkel kirurgiske teknikken, invasivity, lav lav karakteren implantat vekt og, spesielt, en høyere stabilitet gir standardisert og kontrollert biomekaniske forhold sammenlignet med andre mulig intramedulær implantater5. Faktisk mest lukket brudd modeller, fragmenter er stabilisert bare av enkel pinner, som er forbundet med en fullstendig mangel på roterende og aksial stabilitet og en høy risiko for pin og også brudd forvridning. Dette kan betydelig påvirke helbredelsesprosessen, som kan føre til forsinket helbredelse eller ikke-union formasjonen.
Det er velkjent at stabiliteten i brudd fiksering har en enorm innvirkning på den helbredende prosess6,7. En høy stive fiksering resulterer i intramembranous healing, mens en mindre rigid fiksering, som kan micromovements brudd gapet, resulterer i endochondral healing. Stabilisering av brudd med mulig intramedulær skruen viser hovedsakelig en endochondral healing med en stor mengde callus vev, særlig etter 2 uker brudd healing. Muligheten til å høste mye callus vev gjør analyse av flere parametere av forskjellige teknikker.
Her rapportere vi om design og anvendelse av mulig intramedulær skruen i mus, samt fordeler og ulemper i eksperimentelle studier på normal endochondral bein helbredelse.

Bein helbredende modeller er nødvendig for å analysere komplekse mekanismer for brudd healing for å forbedre klinisk brudd behandling. I løpet av det siste tiåret, ble økt bruk av musen modeller i Ortopedisk forskning bemerket, sannsynligvis fordi musen modeller tilbyr en rekke genetisk endret stammer og spesiell Antistoffene for analyse av molekylære mekanismer av brudd healing. For å kontrollere biomekaniske betingelsene, er godt karakterisert osteosynthesis teknikker obligatorisk, også i mus. Her rapportere vi om utforming og bruk av en lukket bein healing modell å stabilisere femur brudd i mus. Mulig intramedulær skruen, medisinsk kvalitet rustfritt stål gir gjennom sprekken komprimering en aksial og roterende stabilitet sammenlignet med mest brukte enkel mulig intramedulær pinner, som viser en fullstendig mangel på aksial og roterende stabilitet. Stabiliteten av mulig intramedulær skruen tillater analyse av endochondral healing. En stor mengde callus vev, mottatt etter stabilisering med skruen, tilbyr ideelle forhold å høste vevet for biokjemiske og molekylære analyser. Flere fordeler av skruen er det faktum at skruen kan settes inn i femur med en minimal invasiv teknikk uten å indusere skade bløtvev. Avslutningsvis er skruen en unik implantatet som ideelt kan brukes i lukkede brudd healing modeller tilbyr standardiserte biomekaniske forholdene.

Driftstiden fra huden snitt å sår nedleggelse var 20 min. Operasjonen kan utføres uten stereo-mikroskop. Postoperatively, var dyrene overvåket daglig. Postoperativ analgesi ble avsluttet etter 2 dager fordi ingen av dyrene viste bevis av smerte etter denne tidsperioden. Dyrene viste også vanlig vektbærende innen 2 dager etter operasjonen. Sårinfeksjoner var ikke observert i hele observasjon perioden.
