Cette méthode peut aider à répondre aux questions clés dans le domaine de la thrombose et de l’hémostase identifiant des sujets plus sensibles aux processus athérothrombotiques en particulier les patients à haut risque traités avec de l’aspirine. Le principal avantage de cette technique est qu’elle est peu coûteuse, simple à utiliser, pratique, a une efficacité élevée et permet l’identification rapide des polymorphismes C924T. Bien que cette méthode puisse fournir un aperçu dans le domaine de l’athérothrombose, elle peut également être appliquée à d’autres systèmes tels que les polymorphismes impliquant la médecine personnalisée ou des médicaments spécifiques afin de comprendre la réponse individuelle pharmacologique.
Le Docteur Sebastiano Ursi, technicien de notre laboratoire, démontrera l’intervention. Pour préparer le tampon Tris EDTA au pH 8.0, ajouter 200 microlitres de 0,5 molaire EDTA et un millilitre d’un chlorure tris molaire dans un bécher, puis ajuster le volume à 100 millilitres avec de l’eau stérile et les conserver à température ambiante. Ensuite, préparez un litre de solution de stock 10X de tampon d’électrophoresis en ajoutant tous les reagents, puis stockez le tampon à température ambiante.
Ensuite, préparez le colorant de chargement de gel en ajoutant tous les reagents puis apportez le volume à 100 millilitres avec de l’eau stérile. Une fois préparé, conserver le colorant à quatre degrés Celsius pendant quelques mois. Ajouter 450 millilitres d’eau à 50 millilitres de tampon TBE 10X pour préparer une solution de tampon TBE 1X.
Ajouter quatre grammes d’agarose dans 200 millilitres de tampon 1X TBE ensemble dans un bécher de 600 millilitres pour préparer 200 millilitres de gel 2% agarose. Le gel d’agarose solidifié stocké à température ambiante peut être redissolé sur un bain d’eau bouillante à 60 degrés pendant 15 à 20 minutes ou dans un four à micro-ondes pendant 35 minutes en ajoutant un petit volume d’eau pour compenser l’augmentation de la concentration d’agarose. Continuer à remuer le bécher sur un mélangeur magnétique pendant environ cinq minutes jusqu’à ce que l’agarose soit suspendue, puis chauffer l’agarose sur une plaque chaude pendant environ 10 minutes ou dans un four à micro-ondes pendant trois minutes pour qu’il se dissolve complètement.
Après purification de l’ADN, mélanger délicatement les échantillons d’ADN frais à l’aide de tubes de pipetage à plusieurs reprises. Il est obligatoire d’obtenir 10 à 50 nanogrammes d’un modèle d’ADN de bonne qualité extrait d’échantillons humains. Pour cette raison, nous préférons utiliser une purification automatisée de l’ADN plutôt qu’une purification semi-automatisée ou manuelle.
Pour commencer la purification, d’abord quantifier et calculer la pureté de l’ADN en mesurant les absorbants à 260, 280 et 320 nanomètres, puis diluer les échantillons à l’aide d’eau stérile. Ensuite, calibrez le spectrophotomètre. Pour constituer la réaction PCR, ajouter tous les reagents pour préparer 25 microlitres de mix master PCR dans un tube de microamplification de 0,2 millilitre.
Placez ensuite les tubes PCR dans un thermocycleur automatisé et amplifiez les échantillons d’ADN purifiés. Une fois le programme terminé, arrêter la réaction en laissant les tubes à quatre degrés Celsius. Ajouter 10X enzyme tampon cinq unités par enzyme de restriction microlitre dans l’eau libre de l’ADN à un volume de 22,5 microlitres.
Ensuite, ajoutez 2,5 microlitres de produit PCR à un nouveau tube PCR puis ajoutez les 22,5 microlitres du mélange de digestion de restriction au produit PCR. Incuber le mélange maître de digestion de restriction de produit de PCR à 37 degrés Celsius pendant quatre heures, puis ajouter 0.5 microgramme par millilitre de bromure d’Ethidium pour tacher le gel pendant 10 minutes. Verser le gel sur le plateau de gel avec le peigne bien en place pour que le gel se solidifie.
Maintenant, versez la boîte de gel avec tampon 1X TBE jusqu’à ce que le gel est couvert. Utilisez des pointes fines pour charger six microlitres de l’ADN digéré amplifié dans les puits formés dans le gel, puis ajoutez un marqueur d’ADN aussi dans un parallèle distinct avec l’échantillon. Ensuite, allumez l’unité d’électrophorèse.
Afin d’identifier le polymorphisme génétique C924T dans le gène TBXA2R, l’analyse RFLP est effectuée. Dans le gel, il affiche deux bandes à 395 et 144 paires de base pour la version sauvage de l’allèle majeur, tandis qu’une seule bande à 539 paires de base pour la version mutante de l’allèle mineur est vu indiquant aucune coupe par l’enzyme de restriction. Fait intéressant, trois groupes de 395, 144 et 539 paires de base sont présents pour l’allèle hétérozygote.
Ensuite, le polymorphisme C924T est confirmé à l’aide de l’analyse des séquences. La séquence mise en surbrillance représente le polymorphisme. Une fois maîtrisée, cette technique peut être effectuée en environ huit heures si elle est effectuée correctement.
Tout en essayant cette procédure, il est important de se rappeler que cette méthode ne peut être utilisée que pour un petit nombre de snips et le pour quelques échantillons dans une session de travail. Après cette procédure, d’autres méthodes comme l’analyse snip peuvent être effectuées afin de répondre à des questions supplémentaires comme l’importance d’autres voies impliquant des processus athérothrombotiques. Après son développement, cette technique a ouvert la voie à des chercheurs dans le domaine de la biologie moléculaire pour explorer les variations génétiques exceptionnelles dans ce début de développement et de progression.
