In de huidige kwalitatieve methode, verkregen we MS3 product ionen een SPC en een N-acyl deel daarvan. Het is essentieel voor het correct toewijzen van zowel een SPC en een N-acyl deel daarvan. Te dien einde moet opgemerkt worden dat andere phosphorylcholine-bevattende moleculen kunnen ook worden gedetecteerd als MS3 product ionen. Diacyl-dunlaag (PC) en plasmalogen-PC zijn overvloedig aanwezig in cellen van zoogdieren, en hun hydrophobicity is vergelijkbaar met die van SM. Daarom,...

Sphingomyelinase (SM) is een gemeenschappelijk sfingolipiden bij zoogdiercellen. SM is gesynthetiseerde intracellulair1 en heden als een voorloper voor andere sfingolipiden zoals een sphingosine-1-fosfaat en een ceramide, die hebben een cruciale rol in immuun cel mensenhandel en huid barrière homeostase, respectievelijk2, 3. De nauwkeurige analyse van de SM-metabolisme is dus belangrijk voor het ophelderen van de functies van het fysiologische en pathologische van sfingolipiden.
SM is samengesteld uit een ceramide en een phosphorylcholine die is gekoppeld aan de 1-hydroxy groep van het ceramide, dat verder uit een sphingosine en een N bestaat-acyl deel daarvan. Een verscheidenheid in de koolstof- en dubbele binding getallen in zowel de sphingosine en N-acyl deel daarvan resulteert in het aantal ceramide (en SM) soorten. Recente vooruitgang in LC-ESI-MS/MS heeft kwantitatieve en kwalitatieve analyse van SM4,5. In de kwalitatieve analyse, werd het aantal koolstof en dubbele bindingen van een sphingoid LCB van SM geïdentificeerd door product ion spectra van LCB toe te wijzen. Echter structuurgegevens van de N-acyl deel daarvan was niet direct verkregen omdat de bijbehorende product-ionen zijn niet gemeld, en dan ook N-acyl wordt werden afgeleid door differentiële analyse tussen voorloper ionen en product ionen overeenkomt met LCB in zowel positieve als negatieve ion modi4,5. In dit rapport presenteren we een methode voor het detecteren van de product-ionen van LCB zowel N-acyl groep gelijktijdig in MS3 modus met behulp van triple vierpolige en vierpolige lineaire ion trap massaspectrometrie, dat de precieze structuur vergemakkelijkt speculatie van elke SM soorten6.
De ion onderdrukking (of toebehoren) effecten, veroorzaakt door de matrix in biologische monsters belemmeren de nauwkeurige kwantificering in LC-ESI-MS-analyse, en daarom is het wenselijk voor de bouw van de kalibratiekrommen voor alle analyten van belang in de identieke matrix van het biologische monster. Deze strategie is echter niet haalbaar omdat het bijna onmogelijk om te bereiden alle SM soorten in biologische monsters, met name in de uitgebreide analyse is. Dus, het is praktisch om te bouwen van een kalibratiekromme en de kwantitatieve bereik met behulp van een representatieve soorten van de SM in de biologische monsters spiked bepalen. Wij twee ionenpaar-geëtiketteerden SM soorten gebruikt voor de constructie van een ijkcurve; een werd gebruikt voor een interne standaard en de andere voor een samengestelde norm. We een kleine hoeveelheid ionenpaar-geëtiketteerden SM soorten gedetecteerd als een standaard compound spiked in biologische monsters en verkregen met succes een kalibratiekromme en de kwantitatieve bereik6.

quantitative and qualitative method for sphingomyelin by lc-ms using two stable isotopically labeled sphingomyelin species

Presenteren we een methode voor het analyseren van sphingomyelinase (SM) kwalitatief en kwantitatief door vloeibare chromatografie-electrospray ionisatie-tandem massaspectrometrie (LC-ESI-MS/MS). SM is een gemeenschappelijk sfingolipiden samengesteld uit een phosphorylcholine en een ceramide als de hydrofiele en hydrofobe component, respectievelijk. Een aantal soorten SM in zoogdiercellen wegens een verscheidenheid in de sphingoid lange keten base (LCB) aanwezig zijn en een N-acyl deel in het ceramide. In dit verslag, tonen we een methode voor het schatten van het aantal koolstof en dubbele bindingen in een LCB en een N-acyl groep op basis van hun overeenkomstige product-ionen in MS/MS/MS (MS3) experimenten. Daarnaast presenteren wij een kwantitatieve analysemethode voor SM met behulp van twee stabiele ionenpaar gelabelde SM soorten, dat vergemakkelijkt het meetbereik in SM kwantificatie bepalen. De huidige methode zal zitten nuttig in karakterisering van een verscheidenheid van SM soorten in biologische monsters en industriële producten zoals cosmetica.
