Dit protocol bespreekt het gebruik van split fluorescerende eiwitten voor het meten van caspase geïnduceerde nabijheid. Split Venus werd gekozen voor deze techniek, omdat het is zeer helder, zeer photostable en de uiteinde is snel 13. Dus, de analyse van Venus uiteinde op caspase geïnduceerde nabijheid kan bieden bijna real-time schattingen van caspase-eiwit interactie dynamiek. Venus is opgesplitst in twee iets overlappende fragmenten, het N-terminus van Venus (Venus-N of VN) aminozuren bestaan...

Het caspase protease familie staan bekend om hun belangrijke rol in apoptosis en aangeboren immuniteit 1. Vanwege hun belang, wanneer, waar, en bepalen hoe efficiënt specifieke caspases worden geactiveerd kan bieden cruciaal inzicht in de mechanismen van caspase activering trajecten. De visualisatie van de vroegste stappen in de opeenvolging van de activering van caspase kunnen de imaging gebaseerd protocol beschreven hier. Deze techniek maakt gebruik van de dynamische eiwit: eiwitinteractie dat station caspase-activering.
De caspases kunnen worden onderverdeeld in twee groepen: de caspases van de initiatiefnemer (caspase-1, -2, -4 -5, -8, -9, -10 en -12) en de caspases van de beul (caspase-3-6 en -7). De beul caspases zijn aanwezig in de cel als voorgevormde Dimeren en worden geactiveerd door de splitsing tussen de grote en kleine subeenheid 2. Wanneer geactiveerd, klieven zij talrijke structureel en regelgevend proteïnen wat resulteert in apoptosis 3. De initiatiefnemer caspases zijn de eerste caspases in het algemeen leiden tot de activering van de caspases van de beul te worden geactiveerd in een traject. In tegenstelling tot de beul caspases, worden initiatiefnemer caspases geactiveerd door dimerisatie 4,5. Deze dimerisatie wordt vergemakkelijkt door de aanwerving van inactieve monomeren aan specifieke grote molecuulgewicht complexen bekend als activering platformen. Vergadering van activering platformen wordt beheerst door een reeks van specifieke eiwit: eiwitinteractie. Deze zijn gemedieerd door geconserveerde eiwit interactie motieven aanwezig in de proform van de initiatiefnemer caspase en omvatten het dood domein (DD), dood effector domein (DED) en caspase werving domein (kaart) 6 (figuur 1A). Activering platformen omvatten over het algemeen een receptor eiwitten en een adapter-eiwit. De receptor is meestal geactiveerd bij de binding van een ligand, inducerende een conformationele verandering die het mogelijk voor de oligomerisatie van talloze moleculen maakt. De receptor werft vervolgens ofwel de caspase direct of adapter moleculen die op hun beurt de caspase aan het complex brengen. Zo komen talrijke caspase-moleculen in de nabijheid van dimerisatie toelaat. Dit staat bekend als de nabijheid van de geïnduceerde model 7. Zodra dimerized, ondergaat de caspase autoprocessing, die dient om de actieve enzym 4,8te stabiliseren. Vergadering van de Apaf1 apoptosome wordt bijvoorbeeld geactiveerd door cytochroom c na haar vrijlating uit de mitochondriën in een proces genaamd mitochondriale buitenmembraan permeabilization (MOMP). Apaf1 werft beurtelings caspase-9 tot en met een interactie die wordt gemedieerd door een kaart in beide eiwitten 9aanwezig. Soortgelijke eiwitinteractie resulteren in de vergadering van de CD95 dood inducerende signalering complexe (DISC) dat tot activering van caspase-8 leidt; de PIDDosome, die caspase-2 activeren kan; en verschillende inflammasome complexen dat caspase-1 activering 10,11,12te leiden. Dus, initiatiefnemer caspases worden gerekruteerd voor specifieke activering platformen door een gemeenschappelijk mechanisme wat resulteert in de nabijheid van de geïnduceerde en dimerisatie, waarzonder activering niet zal plaatsvinden.
Caspase Bimolecular fluorescentie complementatie (BiFC) is een imaging gebaseerde bepaling, die is ontwikkeld voor het meten van deze eerste stap in de activering van de caspases van de initiatiefnemer, waardoor directe visualisatie van caspase geïnduceerde nabijheid na activering platform vergadering. Deze methode maakt gebruik van de eigenschappen van de split fluorescent proteïne Venus. Venus is een helderder en meer photostable versie van gele fluorescerende eiwit (YFP) dat kan worden verdeeld in twee niet-belichting fluorescerende en iets overlappende fragmenten: de N-terminus van Venus (Venus N of VN) en de C-terminus van Venus (Venus C of VC). Deze fragmenten behouden de mogelijkheid om refold en fluorescerende wanneer in de directe nabijheid van 13geworden. Elk fragment Venus is gesmolten tot de prodomain van de caspase, de minimale gedeelte van de caspase dat zich aan het platform van de activering bindt. Dit zorgt ervoor dat de caspases niet enzymatische activiteit behouden en gelijktijdige analyse van downstream gebeurtenissen gekoppeld aan endogene gebeurtenissen dus mogelijk is. De Venus fragmenten refold wanneer de caspase-prodomains worden gerekruteerd voor de activering platform en ondergaan van de nabijheid van de geïnduceerde. (Figuur 1B). De resulterende fluorescentie van Venus kan worden gebruikt voor het nauwkeurig en specifiek bewaken de subcellular localisatie, de kinetiek en de efficiëntie van de vergadering van initiatiefnemer caspase activering platforms in afzonderlijke cellen. Imaging gegevens kan worden verkregen door confocale microscopie of standaard fluorescentie microscopie en kan worden aangepast aan een aantal verschillende microscopie benaderingen, met inbegrip van: time-lapse imaging als u wilt bijhouden van de activering van caspase in real-time; hoge resolutie beeldvorming voor de nauwkeurige analyse van subcellular localisatie; en eindpunt kwantificering van de efficiëntie van de activering.
Deze techniek werd eerst ontwikkeld om de activering van caspase-214onderzoeken. Terwijl het platform van de activering van caspase-2 wordt beschouwd als de PIDDosome, bestaande uit de receptor PIDD (p53-geïnduceerde proteïne met een dood domein) en de adapter RAIDD (RIP-geassocieerde ICH-1/CAD-3 homologe eiwit met een dood domein), PIDDosome-onafhankelijke caspase-2 activering heeft gemeld. Dit suggereert dat extra activeren platforms voor caspase-2 15,16bestaan. Ondanks de niet wetende de volledige onderdelen van de activering van caspase-2-platform, heeft de caspase BiFC techniek toegestaan voor succesvolle ondervraging van de signaalroutes caspase-2 op het moleculaire niveau 14,17. Wij hebben ook met succes aangepast dit protocol voor de inflammatoire caspases (caspase-1, -4 en -5 -12) 18 en, in beginsel dezelfde aanpak zou voldoende moeten zijn ook het analyseren van elk van de resterende caspases van de initiatiefnemer. Dit protocol kan ook worden aangepast om te onderzoeken van andere wegen waren dimerisatie is een belangrijk signaal voor activeren. Bijvoorbeeld, worden STAT eiwitten geactiveerd door dimerisatie na fosforylering door Janus kinase (JAK) 19. Dus, het BiFC-systeem kan worden gebruikt om te visualiseren voor STAT activering evenals vele andere trajecten dynamische eiwitinteractie geregeld. Het volgende protocol biedt stapsgewijze instructies voor de invoering van de verslaggevers in cellen en methodologieën Beeldacquisitie en analyse.

lighting up the pathways to caspase activation using bimolecular fluorescence complementation

Het caspase-familie van proteasen spelen een essentiële rol in apoptosis en aangeboren immuniteit. Daaronder een deelgroep is bekend als initiatiefnemer caspases zijn de eerste in deze trajecten moet worden geactiveerd. Deze groep omvat caspase-2, -8, -9, alsmede de inflammatoire caspases, caspase-1, -4 en -5. De initiatiefnemer caspases worden alle geactiveerd door dimerisatie na aanwerving aan specifieke multiprotein complexen genaamd activering platformen. Caspase Bimolecular fluorescentie complementatie (BiFC) is een imaging gebaseerde benadering waar gesplitst fluorescente proteïnen gesmolten aan initiator caspases worden gebruikt voor het visualiseren van de aanwerving van initiatiefnemer caspases aan hun activering platformen en de daaruit voortvloeiende geïnduceerde nabijheid. Deze fluorescentie biedt een uitlezing van één van de vroegste stappen die nodig zijn voor initiator caspase activering. Het gebruik van een aantal verschillende microscopie gebaseerde benaderingen, kan deze techniek kwantitatieve gegevens bieden op de efficiëntie van caspase activering op het bevolkingsniveau van een, alsook de kinetiek van de activering van caspase en de grootte en het aantal caspase activeren complexen op basis van de per cel.

Een voorbeeld van caspase-2 BiFC geïnduceerd door DNA-beschadiging is afgebeeld in Figuur 3. Camptothecine, een topoisomerase ik inhibitor, werd gebruikt voor het opwekken van DNA-schade en caspase-2-activering.
