Denne protokollen omhandler bruk av delt fluorescerende proteiner måle caspase indusert nærhet. Split Venus ble valgt for denne teknikken fordi det er svært lyse, svært photostable og den refolding er rask 13. Caspase protease familien er kjent for sin avgjørende roller i apoptose og medfødt immunitet 1. På grunn av deres betydning, avgjøre når, hvor og hvor effektivt bestemte caspases aktiveres kan gi viktig innsikt i mekanismer for caspase aktivisering trasé. Den tenkelig basert protokollen beskrevet her kan effekten av de tidligste trinnene i caspase aktivisering kaskade. Denne teknikken utnytter dynamisk protein: protein interaksjoner som stasjonen caspase aktivisering.
I caspases kan deles inn i to grupper: initiator-caspases (caspase-1, -2, -4, 5,-8,-9, -10 og -12) og bøddelen caspases (caspase-3-6 og -7). Bøddelen caspases finnes i cellen som preformed dimers og aktiveres som spalting mellom store og små delenhet 2. Når aktivert, hveran mange strukturelle og regulatoriske proteiner som resulterer i apoptose 3. Initiator-caspases er det første caspases aktiveres i en sti og generelt utløse aktivering av bøddelen caspases. I motsetning til bøddelen caspases aktiveres initiatoren caspases som dimerization 4,5. Denne dimerization er tilrettelagt av rekruttering av inaktive monomerer til bestemte store molekylvekt komplekser kalles aktivisering plattformer. Montering av aktivisering er underlagt en rekke bestemt protein: protein interaksjoner. Disse er formidlet av bevarte protein samhandling motiver i den proform av initiatoren caspase og inkluderer død domene (DD), død effektor domene (DED) og caspase rekruttering domene (kort) 6 (figur 1A). Aktivisering plattformer inkluderer vanligvis en reseptor protein og en adapter protein. Reseptoren aktiveres vanligvis ved binding av en ligand, indusere en conformational endring som tillater oligomerization av mange molekyler. Reseptoren deretter rekrutter enten til caspase direkte eller adapter molekyler som igjen bringer caspase til komplekset. Dermed kommer mange caspase molekyler i nærheten tillater dimerization. Dette kalles indusert nærhet modell 7. Når dimerized, gjennomgår caspase autoprocessing, som serverer å stabilisere aktiv enzym 4,8. Apaf1 rekrutterer igjen caspase-9 til en samhandling som er formidlet av et kort i begge proteiner 9. PIDDosome, som kan aktivere caspase-2; og ulike inflammasome komplekser som starter caspase-1 aktivering 10,11,12. Dermed er initiatoren caspases rekruttert til bestemte aktivisering plattformer av en felles mekanisme indusert nærhet og dimerization, uten noe som ikke skjer aktivisering.
Caspase resultatet av dette fluorescens Complementation (BiFC) er en bildebehandling-basert analyse som ble utviklet for å måle denne steg for aktivering av initiatoren caspases, slik at direkte visualisering av caspase indusert nærhet etter aktivisering plattform montering. Denne metoden tar fordel av egenskapene til split fluorescerende protein Venus. Venus er en lysere og mer photostable versjon av gule fluorescerende protein (YFP) som kan deles inn i to ikke-fluorescerende og litt overlappende fragmenter: N-terminus Venus (Venus N eller VN) og C-terminus Venus (Venus C eller VC). Disse fragmentene beholder refold og bli fluorescerende i nærheten 13. Hver Venus fragment er del av prodomain av caspase, som er den minimale delen av caspase som binder aktivisering-plattformen. Dette sikrer at caspases beholder ikke enzymatisk aktivitet og derfor samtidige analyse av nedstrøms hendelser knyttet endogene hendelser er mulig. Venus fragmenter refold når caspase prodomains rekrutteres aktivisering-plattformen og gjennomgå indusert nærhet. (Figur 1B). Den resulterende Venus fluorescensen kan brukes til nøyaktig og spesielt overvåke den subcellular lokaliseringen, kinetics og effektiviteten av montering av initiatoren caspase aktivisering plattformer i enkeltceller. Bildebehandling data kan skaffes ved AC confocal mikroskopi eller standard fluorescens mikroskopi og kan tilpasses mange forskjellige mikroskopi tilnærminger inkludert: time-lapse imaging for å spore caspase aktivisering i sanntid; Høyoppløselig bilde for presise avgjørelser av subcellular lokalisering; og sluttpunkt kvantifisering av effektiviteten av aktivisering.
Denne teknikken ble først utviklet for å undersøke aktivering av caspase-214. Mens aktivering plattformen for caspase-2 antas å være PIDDosome, bestående av reseptoren PIDD (p53-indusert protein med et død domene) og adapter RAIDD (RIP-assosiert ICH-1/CAD-3 homologe protein med en død-domene), PIDDosome-uavhengig caspase-2 aktivisering er rapportert. Dette tyder på at ekstra aktivering plattformer for caspase-2 finnes 15,16. Til tross for ikke å vite full komponentene i caspase-2 aktivisering plattformen, har caspase BiFC teknikken tillatt for vellykket avhør av caspase-2-signalveier på molekylært nivå 14,-17. Vi har også lykkes tilpasset denne protokollen for den inflammatoriske caspases (caspase-1-4, 5 og -12) 18 og i prinsippet den samme tilnærmingen bør være tilstrekkelig tilsvarende analysere hver av de gjenværende initiatoren caspases. Denne protokollen kan tilsvarende tilpasses å undersøke andre veier ble dimerization er store aktivering signal. For eksempel er STAT proteiner aktivert ved dimerization etter fosforylering av Janus kinase (JAK) 19. Dermed kan BiFC systemet brukes til å visualisere for STAT aktivisering, samt mange andre baner regulert av dynamisk protein interaksjoner.
