Økning i utbredelsen av T2DM har blitt en global helse-krise i årene utgjør en alvorlig trussel mot folkehelsen1. Mange faktorer er funnet å være causally knyttet til utviklingen av T2DM, blant annet, regelmessige funn tyder på at en vanlig konvergent punkt for disse faktorene er induksjon av oksidativt stress som fører til generasjon av overdreven ROS2 , 3.
Et bredt spekter av miljømessige kjemikaliene som PCB, dioksiner og BaP har blitt funnet for å indusere oksidativt stress, som kan forringe funksjonen i bukspyttkjertelen β celler og føre til insulinresistens og T2DM4. Selv om hvilket fysiologiske ROS spiller en viktig rolle i cellulære funksjoner, eksponering for ROS som overstiger kapasiteten av antioksidant resulterer i skade celler/vev og fører til sykdommer5. Det overordnede målet med dette prosjektet er å utvikle et cellebasert analysen skjermen kjemikalier for deres diabetogenic potensialer basert på deres induksjon av ROS i bukspyttkjertelen celler. Bukspyttkjertelen mangler metabolske avgiftning og er en følsom mål for xenobiotic oval-indusert skade6,7. Derfor ved å måle direkte ROS generert i bukspyttkjertelen celler, bør denne analysen gi en direkte tilnærming til kjemisk-indusert skaden i bukspyttkjertelen. Eksponering for visse miljømessige kjemikalier i menneske og dyr har funnet cellular skade bukspyttkjertelen β cellene som vil føre til utvikling av type 2 diabetes mellitus (T2DM). Selv om mekanismene for kjemisk-indusert β celleskader var uklart og sannsynlig å bli komplekse, er en regelmessig finne at disse kjemikaliene induserer oksidativt stress fører til generering av overdreven reaktive oksygen arter (ROS) som induserer skade β cellen.
