Ackumulerande bevis tyder på att exponering för miljö kemikalier spelar en viktig roll i utvecklingen av T2DM. Xenobiotika-inducerad ROS har erkänts som en potentiell etiologisk faktor som bidrar till utvecklingen av T2DM. Människor utsätts för ett brett utbud av xenobiotiska kemikalier och det finns ett stort behov av ny forskningstekniker att effektivt identifiera de pankreas gifter och utreda mekanismen av toxicitet specifika för bukspottkörtelns celler.

Ökningar i förekomsten av T2DM har blivit en global hälsa kris under de senaste åren innebär ett allvarligt hot mot folkhälsan1. Många faktorer har konstaterats vara kausalt kopplat till utvecklingen av T2DM, bland vilka återkommande fynd tyder på att en gemensam konvergerande punkt för dessa faktorer är induktion av oxidativ stress som leder till generation av överdriven ROS2 , 3.
Ett brett spektrum av miljömässiga kemikalier inklusive PCB, dioxiner och BaP har konstaterats för att inducera oxidativ stress, vilket kan försämra funktionen av pankreas β-celler och leda till insulinresistens och T2DM4. Även om den fysiologiska nivån av ROS spelar en viktig roll i cellulära funktioner, exponering för ROS som överskrider kapaciteten hos antioxidant systemet resulterar i skador på cellerna eller vävnaderna och leder till sjukdomar5. Pankreas β-celler uttrycker en låg nivå av antioxidant enzym och således är måltavla känslig för oxidativ stress-medierad skada6,7. Kronisk exponering för höga nivåer av ROS har visat sig orsaka stressinducerade bukspottskörteln cell dysfunktion5 samt insulinresistens i levern och fettväv8.
Det övergripande målet med detta projekt är att utveckla en cellbaserade test för skärmen kemikalier för deras diabetogent potential baserat på deras induktion av ROS i bukspottkörtelns celler. Bukspottkörteln saknar metabola avgiftning och är en känslig mål för xenobiotiska-inducerad skada6,7. Denna analys bör därför, genom att direkt mäta ROS genereras i bukspottkörtelns celler, innehålla en direkt uppskattning av den kemiska-inducerad skadan i bukspottkörteln. För att utveckla denna metod, vi isolerade mus Langerhanska odlade den isolerade islet cell kultur villkor med kemikalier och utnyttjade den kemisk-inducerad ROS generationen som utslaget. Proceduren är enkel och effektiv att identifiera ROS-inducerande kemikalier i isolerade Holmen; Det kan vidareutvecklas för undersökning av toxicitet mekanismer som är specifika för bukspottkörteln in vitro.

Exponering för vissa miljömässiga kemikalier i människa och djur har visat sig orsaka cellskada i pankreas β-celler som kommer att leda till utvecklingen av typ 2-diabetes mellitus (T2DM). Även om mekanismerna för de kemisk-inducerad β cellskador var oklart och kan vara komplexa, är ett återkommande fynd att dessa kemikalier inducera oxidativ stress leder till generation av överdriven reaktiva syreradikaler (ROS) som inducerar skador till β-cellen.
