Cel infiltratie is kritisch voor de regeneratie en remodelleert van de vasculaire graft in vivo16. Beperkte cel infiltratie is vaak gerelateerd aan de relatief kleine poriën van de prothese, die een belemmering vormen voor de migratie van de cellen in de wand van de prothese. Om aan te pakken dit probleem, we een gewijzigde methode ontwikkeld om electrospun PCL vaattransplantaten met grote-poriënstructuur bereiden. Vaattransplantaten gemaakt van synthetische polymeren worden algemeen gebruikt in de kliniek voor de behandeling van cardiovasculaire aandoeningen (CVDs). Helaas, in het geval van kleine diameter vaattransplantaten (D < 6 mm) er zijn geen succesvolle producten beschikbaar vanwege de lage bij veroorzaakt door verminderde bloed stroomsnelheid, die vaak tot trombose, intima hyperplasie en andere leidt complicaties1.
Weefselengineering biedt een alternatieve strategie te realiseren op lange termijn bij en homeostase op basis van een steiger geleide vasculaire regeneratie en wederopbouw. In detail, de vasculaire prothese, als een driedimensionale sjabloon, kon bieden mechanische ondersteuning en structurele begeleiding tijdens de regeneratie van vaatweefsel en invloed cellulaire functies, met inbegrip van cel adhesie, migratie, proliferatie, en secretie van extracellulaire matrix2. Tot nu toe zijn verschillende synthetische polymeren geëvalueerd voor toepassingen in vaatweefsel engineering. Onder deze polymeren, is poly(ε-caprolactone) (PCL) intensief onderzocht vanwege goede cel compatibiliteit en langzame afbraak, variërend van enkele maanden tot twee jaar3. In een rat aorta model4,5,6, PCL vaattransplantaten verwerkt door electrospinning tentoongesteld uitstekende structurele integriteit en bij, zo goed als continu verhoogde cel invasie en neovascularization in de Graft muur voor maximaal 6 maanden. Echter nadelige weefsel remodeling, met inbegrip van regressie van cellen en de haarvaten en verkalking, werden ook waargenomen bij langere timepoints, omhoog tot 18 maanden.
Cellularization van de vasculaire prothese is een sleutelfactor bepalen Weefselregeneratie en omvorming van7. Electrospinning, heeft als een veelzijdige techniek, grote schaal gewerkt voor de bereiding van vaattransplantaten met nano-vezelige structuur8. Helaas, de relatief kleine poriënstructuur leidt vaak tot onvoldoende cel infiltratie in de electrospun vasculaire prothese, waardoor de latere weefselregeneratie wordt beperkt. U kunt dit probleem oplossen, zijn diverse technieken geprobeerd poriegrootte en algehele porositeit, met inbegrip van het zout/polymeer uitloging9,10, wijziging van verzamelaar apparaten, nabehandeling door laserstraling11 te verhogen , enz. In feite, is de structuur van electrospun transplantaties (met inbegrip van de diameter van de vezel, poriegrootte en porositeit) nauw verwant aan de verwerking voorwaarden12,13. Tijdens electrospinning, kan de diameter van de vezels gemakkelijk worden gecontroleerd door het veranderen van de parameters, zoals de concentratie van de polymeeroplossing, debiet, spanning, etc. 14 , 15, en dus de poriën en porositeit dienovereenkomstig zijn verbeterd.
Onlangs meldden wij een gemodificeerde PCL electrospun transplantaat met macroporeuze structuur (vezels met diameter van 5-7 µm en poriën van 30-40 µm). In vivo implantatie door vervanging van de abdominale aorta rat toonde hoge tarief van bij, evenals de goede endothelialization en gladde spieren regeneratie op 3 maanden na operatie16. Wat nog belangrijker is, kon geen negatieve weefsel remodeling met inbegrip van verkalking en cel regressie worden waargenomen zelfs na een jaar van implantatie.

implantation of electrospun vascular grafts with optimized structure in a rat model

Hier presenteren we een protocol om te fabriceren macroporeuze PCL-vasculaire graft en beschrijven van een evaluatie-protocol met behulp van een model van de rat van vervanging van de abdominale aorta. De electrospun vaattransplantaten bezitten vaak relatief kleine poriën, die beperken cel infiltratie in de transplantaties en de regeneratie en verbouwing van de neo-aders belemmeren. In deze studie werden PCL vaattransplantaten met dikkere vezels (5-6 µm) en grotere poriën (~ 30 µm) vervaardigd met behulp van een gemodificeerde gegevensverwerking. De prestaties op lange termijn van de prothese werd beoordeeld door de inplanting in een rat abdominale aorta-model. Echografie-analyse toonde aan dat de transplantaten bleef octrooi zonder aneurysma of stenose die zelfs na 12 maanden innesteling intreedt. Macroporeuze structuur verbeterd de cel ingroei en dus bevorderd weefsel geregenereerd op 3 maanden. Wat nog belangrijker is, was er geen teken van nadelige Remodellerend, zoals verkalking binnen de stadsmuur van het transplantaat na 12 maanden. Daarom houdt electrospun PCL vaattransplantaten met gemodificeerde macroporeuze verwerking potentieel als een slagader substituut voor lange termijn implantatie.

De PCL-transplantaties waren transplanteren op 3 maanden en 12 maanden post-operatively en geanalyseerd door histologische standaardtechnieken voor haematoxyline en eosine (H & E), Masson trichrome, Verhoeff-van Gieson (VVG), Von Kossa en immunofluorescentie kleuring voor α-SMA, MYH, vWF en elastine. De histologische beelden werden genomen met behulp van een rechtop Microscoop, en de immunofluorescentie beelden werden genomen met behulp van een microscoop met ﬂuorescence.
