Microfluidic tools bieden veelvoudige voordeel halen uit het bestuderen van wormen. Beeldvorming in een PDMS biedt apparaat hogere grafische kwaliteit ten opzichte van een standaard NGM agarplaat. Meerdere afbeeldingen kunnen afkomstig zijn uit een enkele worm, in tegenstelling tot de traditionele methoden waarin dieren zijn geplukt uit de plaat en gemonteerd op een microscoopglaasje voor imaging. Wijzigingen in de omgevingscondities kunnen leiden tot activatie van genetische programma's begeleid door inductie en onderdrukking van de expressie van specifieke genen1,2. Deze kinetische veranderingen kunnen variabele onder weefsels in dezelfde dieren en tussen verschillende dieren. Studies van dergelijke genetische programma's pleit daarom voor methoden die verlenen nauwkeurige dynamische controle van milieuomstandigheden en longitudinale beeldvorming van individuele dieren.
In de afgelopen jaren zijn microfabricated fluidic apparaten gebruikt om het bestuderen van vele aspecten van respons en gedrag bij kleine dieren, zoals wormen, vliegen, water beren en meer3,4,5,6, 7. Toepassingen omvatten bijvoorbeeld, diepe fenotypering, optogenetic opnemen van neuronale activiteit in reactie op chemische stimuli en volgen van gedrag van de motor zoals motoriek en pompen8,9,10 , 11.
Microfluidic-gebaseerde benaderingen houden veel eigenschappen die op lange termijn longitudinale beeldvorming van antwoord op het milieu signalen, met inbegrip van nauwkeurige dynamische controle van het lokale communicatie, flexibel ontwerp waarmee onderhoud van kunnen profiteren individuele dieren in de afzonderlijke kwartalen en gunstige kenmerken voor imaging. Behoud van dieren in een microfluidic kamer voor een lange tijd met een minimale negatieve impact op hun mens goed is echter een uitdaging, waarvoor bijzondere aandacht in het ontwerp van het microfluidic-apparaat alsook bij de uitvoering van het experiment.
We laten hier zien dat het gebruik van WormSpa, een microfluidic apparaat voor longitudinale beeldvorming van Caenorhabditis elegans. 5 individuele wormen zijn beperkt in de kamers. Een constante lage stroom van vloeibare en bacteriële suspensie garandeert dat wormen zijn goed gevoed en voldoende actief goede gezondheid te behouden en te verlichten stress, en de structuur van de kamers laat wormen om eieren te leggen. De eenvoud van het ontwerp en de werking van WormSpa moeten onderzoekers zonder eerdere ervaring in microfluidics te nemen van dit apparaat in hun eigen onderzoeksplannen toestaan.

In het laatste decennium, microfluidic technieken zijn toegepast om te bestuderen van kleine dieren, met inbegrip van de nematode Caenorhabditis elegans, en zijn nuttig als een handige levende imaging platform bieden mogelijkheden voor een nauwkeurige besturing van gebleken experimentele omstandigheden in real time. Ter illustratie van het vermogen, wij aantonen van de principiële experimenten waarin wormen in het apparaat met pathogene bacteriën besmet waren, en de dynamiek van de expressie van genen van de immuunrespons en het leggen van eieren waren doorlopend gecontroleerd in individuele uitgevoerd dieren. Het eenvoudige ontwerp en de werking van dit apparaat maken het geschikt voor gebruikers geen ervaring heeft met microfluidic gebaseerde experimenten. Wij stellen voor dat deze aanpak nuttig voor veel onderzoekers ook geïnteresseerd in de lengterichting opmerkingen van biologische processen onder welomschreven voorwaarden zitten zal.

Leeftijd-gesynchroniseerde jonge volwassen wormen (46 uur post L1 larvale arrestatie bij 25 ° C)12 werden geladen in het apparaat, zoals beschreven in het protocol.
