단일 클론 항 체 (mAbs) 기초 연구와 의학에 대 한 수요는 매년 증가 하 고 있다. Hybridoma 기술 mAb 개발 이후 1975 년에 그것의 첫 번째 보고서에 대 한 지배적인 방법 되었습니다. 대체 기술, mAb 개발에 대 한 파지 디스플레이 방법으로 점점 더 매력적인 Humira, 첫 번째 페이지에서 파생 된 항 체와 베스트 셀러 mAbs 중은 2002 년에 류 마티스 관절염의 임상 치료에 대 한 승인 이후입니다. 비 동물 mAb 개발 기술을 기반으로, 살 균 소 전시 항 원 immunogenicity, 인간 화, 그리고 전통적인 hybridoma 기술 기반 항 체 개발에서 필요한 동물 유지 보수를 무시 합니다. 이 프로토콜에서 우리는 합성 살 균 소 표시 팹 라이브러리의 109-1010 단일 electroporation에 얻을 수의 다양성을 건설 하는 방법을 설명합니다. 이 프로토콜의 구성: 1) 고효율 전기 유능한 세포 준비; 2) 추출 uracil 포함 하는 단일 가닥 DNA (뒤-ssDNA); 3) Kunkel 메서드 기반 oligonucleotide 감독 mutagenesis; 4) electroporation 그리고 계산 라이브러리 크기; 5) 단백질 A/L 기반 효소 연결 된 immunosorbent 분석 결과 (ELISA) 폴딩에 대 한 기능적 다양성 평가; 그리고 6) 다양성의 DNA 순서 분석.
