Dit protocol wordt de eerste screeningsmethode om een systeem van de robuuste cultuur voor het onderhoud van gekwalificeerde hPSCs gedemonstreerd. Eerst, beschreven we hoe voor te bereiden op een platform met gevarieerde kunstmatige ECMs en cel dichtheden zaaien met behulp van een microfluidic apparaat geïntegreerd met een nanofiber-serie, de MACME array. Menselijke pluripotente stamcellen (hPSCs)1,2 zelf vernieuwen onbeperkt en onderscheid in verschillende geslachten van weefsel, die kunnen een revolutie Geneesmiddelenontwikkeling, cel-gebaseerde therapieën, weefselkweek en regeneratieve geneeskunde 3 , 4 , 5 , 6. algemene cultuur gerechten en microtiterplaat platen, echter zijn niet ontworpen om precieze cel van de fysische en chemische manipulatie op cellulair niveau met het bereik van nano - tot micro-meters, die een kritische factor voor cellulaire expansie is, zelf-vernieuwing en differentiatie. Om aan te pakken dit nadeel, hebben studies onderzocht de rol van cellulaire microenvironments bij het reguleren van de cel-lot besluiten en cel functies4. In de afgelopen jaren zijn een toenemend aantal studies uitgevoerd om te reconstrueren van cellulaire microenvironments in vitro7,8. Nano - en micro-fabricage processen hebben deze microenvironments door de manipulatie van chemische9,10,11,12,13, vastgesteld 14,,15,16,17 en fysieke18,19,20 milieu signalen. Tot nu toe waren er geen meldingen te systematisch onderzoeken van de onderliggende mechanismen van chemische en fysische milieu signalen op cel-lot besluiten en functies binnen een enkel platform.
Hier introduceren we een strategie gebaseerd op eenvoudig ontwerpprincipes om een robuuste screening platform (Figuur 1). Eerst beschrijven we de procedure van de ontwikkeling van een geïntegreerd platform voor het maken van de veelzijdige, kunstmatige cellulaire microenvironments met behulp van een nanofiber array en een microfluidic structuur: de Multiplexed kunstmatige cellulaire communicatie (MACME) matrix (Figuur 1A en 2A). De nanofiber array heeft 12 verschillende microenvironments in wisselende combinaties van nanofiber materialen en dichtheden. Electrospinning werd gebruikt om nanofibers te fabriceren. De nanofiber materialen, zoals polystyreen (PS)21, polymethylglutarimide (PMGI)22en gelatine (GT)23, werden ontworpen voor het testen van hun chemische eigenschappen, die later nog van invloed kunnen zijn op cel adhesie en het onderhoud van pluripotent ( Figuur 2B). Nanofiber dichtheden waren gevarieerd door het veranderen van electrospinning tijd en de geproduceerde nanofibers werden gedefinieerd volgens hun dichtheid (niet gefinisht, met D = XLow/Low/Mid/High). De microfluidic structuur bestaat uit Polydimethylsiloxaan (PDMS) 48 celkweek kamers, die kunnen worden geplaatst langs de standaard afmetingen van de 96-Wells-microplate herbergen. PDMS is een biocompatibel en gas-inwisselbaar polymeer gewoonlijk gebruikt voor het fabriceren van microfluidic apparaten24. Elk kanaal van de microfluidic werd ontworpen om 700-µm breed en 8.4-mm lang en had twee inhammen aan de randen (tabel 1). De kamers hadden verschillende hoogtes (250, 500 en 1000 µm) te manipuleren van de eerste cel-zaaien dichtheden (0.3 en 0.6 1,2 × 105 cellen/cm2), die met overleven, proliferatie en differentiatie van hPSCs25 correleren kunnen (Figuur 2C). Het aantal cellen zaadjes in een kamer is evenredig met de dichtheid van de kolom boven de vloer van de kamer, en dus de eerste cel zaaien dichtheid werd gecontroleerd door de invoering van de dezelfde celsuspensie in cultuur kamers met verschillende hoogtes. Alle zenders werden ontworpen om ≥ 250-µm hoge26 om te minimaliseren van de effecten van lage-zuurstof-spanning27 en schuifspanning28 op de cellen. Kanaal hoogten van 250, 500 en 1000 µm hier hierna afgekort als XCD met X = Low, Mid, en hoog, respectievelijk. De omgevingen met verschillende nanofiber dichtheden en eerste cel-zaaien dichtheden waren afgekort als "Material_NF density_Cell dichtheid" (bijvoorbeeld, GT_HighNF_HighCD: een milieu gekenmerkt door high-density GT nanofibers en hoge initiële cel-zaaien dichtheid).
Vervolgens beschrijven we hoe eencellige analyses voor het systematisch onderzoek naar cel gedrag in reactie op milieu-factoren (Figuur 1B) uitvoeren. Als een bewijs-van-concept geïdentificeerd we de optimale cellulaire omgeving voor hPSC zelf-vernieuwing, dat is een belangrijke functie voor hPSC onderhoud (Figuur 1B)29. Beeld-gebaseerde cytometry, gevolgd door statistische analyses, zorgt voor kwantitatieve interpretatie van individuele cellulaire fenotypische reacties op cellulaire omgevingen. Onder een verscheidenheid van cellulaire functies biedt dit Witboek een gedetailleerde procedure ter identificatie van de optimale voorwaarden voor het behoud van hPSC zelf-vernieuwing.

Cellulaire microenvironments bestaan uit een aantal signalen, zoals groeifactoren, extracellulaire matrices en intercellulaire interacties. Deze signalen zijn goed georkestreerd en cruciaal bij het reguleren van de functies van de cel in een levend systeem. Hoewel een aantal onderzoekers hebben geprobeerd te onderzoeken van de correlatie tussen milieufactoren en gewenste cellulaire functies, er nog veel onbekend. Dit is grotendeels te wijten aan het ontbreken van een juiste methodologie te bootsen zo'n milieu signalen in vitro, gelijktijdig testen met verschillende milieu aanwijzingen op cellen. Wij rapporteren hier, een geïntegreerd platform van microfluidic kanalen en een array van de nanofiber, gevolgd door hoge-eencellige inhoudsanalyse, te onderzoeken van de stamcel fenotypen gewijzigd door verschillende omgevingsfactoren. Om aan te tonen van de toepassing van dit platform, deze studie concentreert zich op de fenotypen van zelf vernieuwen van menselijke pluripotente stamcellen (hPSCs). Hier presenteren we de procedures van de voorbereiding voor een matrix van de nanofiber en de microfluidic-structuur in de fabricage van een array Multiplexed kunstmatige cellulaire communicatie (MACME). Bovendien de algemene stappen voor de eencellige profiling, cel kleuring met meerdere fluorescente markeringen, meerdere fluorescentie imaging en statistische analyses, worden beschreven.

MACME arrays: ontwerp en fabricatie: In combinatie met nanofiber technologie gebruikten we de cultuur van de cel van de microfluidic- en screeningsprocedures van eerder gebruikte om te identificeren van de optimale omstandigheden voor hPSC zelf-vernieuwing of differentiatie35,36 (Figuur 1) technieken.
