Ce protocole illustre la première méthode de dépistage pour établir un système robuste de la culture pour l’entretien des hPSCs qualifiés. Tout d’abord, nous avons décrit comment préparer une plateforme mettant en vedette divers SEGDA artificiel et cellule ensemencement des densités à l’aide d’un dispositif microfluidique intégré avec un tableau de nanofibres, le tableau MACME. De1,(hPSCs) des cellules souches pluripotentes humaines2 se renouveler de manière illimitée et se différencient en diverses lignées de tissu, qui pourraient révolutionner le développement des médicaments, thérapies cellulaires, génie tissulaire et la médecine régénérative 3 , 4 , 5 , 6. plats de culture générale et de microplaques, cependant, ne sont pas conçus pour permettre la manipulation précise de cellules physiques et chimiques au niveau cellulaire avec la gamme de nano - de micromètres, qui est un facteur critique pour l’expansion cellulaire, l’auto-renouvellement et différenciation. Pour remédier à cet inconvénient, les études ont examiné le rôle des micro-environnements cellulaires dans la régulation des décisions de la cellule-destin et cellule fonctions4. Ces dernières années, un nombre croissant d’études ont été mené pour reconstruire des micro-environnements cellulaire in vitro7,8. Processus de nano - et microfabrication ont établi ces micro-environnements à travers la manipulation de produits chimiques9,10,11,12,13, 14,15,16,17 et physique18,19,20 signaux environnementaux. Jusqu'à présent, aucune n’a été d’étudier systématiquement les mécanismes sous-jacents des signaux environnementaux chimiques et physiques sur les décisions de la cellule-destin et fonctions au sein d’une plate-forme unique.
Ici, nous introduisons une stratégie fondée sur les principes de conception simple pour établir une plate-forme robuste de dépistage (Figure 1). Tout d’abord, les auteurs décrivent la procédure d’élaboration d’une plate-forme intégrée pour créer des micro-environnements cellulaires polyvalents, artificielles en utilisant un tableau de nanofibres et une structure microfluidique : tableau The multiplexés artificiel cellulaire microenvironnement (MACME) (Figure 1A et 2 a). Le tableau de nanofibres a 12 microenvironnements différents dans diverses combinaisons de matériaux de nanofibres et de densité. Électrofilage servait à fabriquer des nanofibres. Les matériaux de nanofibres, tels que le polystyrène (PS)21, polymethylglutarimide (PMGI)22et23de gélatine (GT), ont été conçus pour tester leurs propriétés chimiques, ce qui pourraient affecter adhésion cellulaire et maintien de la pluripotence ( Figure 2B). Les densités de nanofibres étaient variées en changeant de temps électrofilage et les nanofibres générés ont été définis selon leurs densités (DNF, avec D = XLow/Low/Mid/High). La structure de la microfluidique est composée de polydiméthylsiloxane (PDMS) hébergeant les 48 chambres de culture de cellules, qui peuvent être positionnés selon les dimensions standards de la Microplaque 96 puits. PDMS est un polymère biocompatible et gaz échangeable, généralement utilisé pour la fabrication de dispositifs microfluidiques24. Chaque canal microfluidique a été conçu pour être 700 µm large et 8,4 mm de long et avait deux anses sur ses bords (tableau 1). Les chambres ont des hauteurs différentes (250, 500 et 1000 µm) pour manipuler l’ensemencement des cellules densités initiales (0,3, 0,6 et 1,2 × 105 cellules/cm2), qui pourraient mettre en corrélation avec la survie, la prolifération et la différenciation des hPSCs25 (Figure 2C). Le nombre de cellules ensemencées dans une chambre est proportionnel à la densité de la colonne au-dessus du plancher de la chambre, et ainsi la cellule initiale, la densité de semis a contrôlé en introduisant la même suspension cellulaire dans des chambres de culture à différentes hauteurs. Tous les canaux ont été conçus pour être ≥ 250 µm-hauteur26 pour minimiser les effets de tension faible teneur en oxygène27 et contrainte de cisaillement28 sur les cellules. Hauteurs de canal de 250, 500 et 1000 µm sont abrégés ici comme XCD avec X = Low, Mid et High, respectivement. Les environnements avec des densités de nanofibres distinctes et densités d’ensemencement des cellules initiales ont été raccourcies comme « Material_NF density_Cell densité » (par exemple, GT_HighNF_HighCD : un environnement caractérisé par des nanofibres de GT haute densités et haute cellule-amorçage initial densité).
Par la suite, nous décrivons comment effectuer des analyses de cellules individuelles afin d’étudier systématiquement les comportement cellulaire en réponse à des facteurs environnementaux (Figure 1B). Comme une preuve de concept, nous avons identifié l’environnement cellulaire optimal pour hPSC autorenouvellement, qui est une fonction clée pour hPSC entretien (Figure 1B)29. Imageur cytométrie en flux, suivie d’analyses statistiques, permet une interprétation quantitative des réponses phénotypiques cellulaires individuelles aux environnements cellulaires. Parmi une variété de fonctions cellulaires, ce document fournit une procédure détaillée afin d’identifier les conditions optimales pour le maintien de hPSC l’auto-renouvellement.

Microenvironnements cellulaires se composent d’une variété d’indices, comme des facteurs de croissance, matrices extracellulaires et interactions intercellulaires. Ces signaux est bien orchestré et ils est essentiels à la régulation des fonctions cellulaires dans un système vivant. Bien qu’un certain nombre de chercheurs ont tenté d’enquêter sur la corrélation entre les facteurs environnementaux et les fonctions cellulaires désirées, une grande partie reste inconnue. C’est en grande partie en raison de l’absence d’une méthodologie appropriée pour imiter ces signaux environnementaux in vitroet de tester simultanément les différents signaux environnementaux sur les cellules. Nous rapportons ici une plateforme intégrée de canaux microfluidiques et un tableau de nanofibres, suivie haute teneur unicellulaires analyse, afin d’examiner les phénotypes de cellules souches modifiées par des facteurs environnementaux distincts. Pour illustrer l’application de cette plate-forme, cette étude se concentre sur les phénotypes de renouvellement automatique des cellules souches pluripotentes humaines (hPSCs). Nous présentons ici les procédures de préparation pour un tableau de nanofibres et de la structure de la microfluidique dans la fabrication d’un tableau du microenvironnement cellulaire artificiel multiplexés (MACME). En outre, les étapes globales de la seule cellule de profilage, cellule souillant avec plusieurs marqueurs fluorescents, imagerie de fluorescence multiples et des analyses statistiques, sont décrites.
