Denne protokollen viser den første screening metoden å etablere et robust kultur system for vedlikehold av kvalifiserte hPSCs. Først beskrev vi hvordan forberede en plattform med mangfoldig kunstig ECMs og celle seeding tettheter ved hjelp av en microfluidic enhet integrert med et nanofiber utvalg, MACME array. Menneskelige pluripotent stamceller (hPSCs)1,2 selvstendig fornye unlimitedly og skille ut forskjellige vev linjene, som kan revolusjonere narkotika utvikling, celle-basert behandling, vevet engineering og regenerativ medisin 3 , 4 , 5 , 6. generell kultur retter og være ferdig innen plater, men er ikke utformet slik at presis fysiske og kjemiske celle manipulasjon på cellenivå med et utvalg av nano - til mikro-meter, som er en kritisk faktor for mobilnettet ekspansjon, selvtillit fornyelse, og differensiering. For å løse denne ulempen, har studier undersøkt rollene til mobilnettet microenvironments i å regulere celle skjebne beslutninger og celle funksjoner4. De siste årene, har et økende antall studier vært gjennomført for å rekonstruere mobilnettet microenvironments i vitro7,8. Nano - og mikro-metallbearbeiding prosesser har etablert disse microenvironments gjennom manipulering av kjemiske9,10,11,12,13, 14,15,16,17 og fysiske18,19,20 miljømessige signaler. Inntil nå er det ikke systematisk undersøke de underliggende mekanismene av kjemiske og fysiske miljømessige signaler på cellen skjebne beslutninger og funksjoner i en enkelt plattform.
Her introduserer vi en strategi basert på enkel designprinsipper å etablere en robust screening plattform (figur 1). Først vi beskrive utviklingen prosedyren av en integrert plattform for å lage allsidig, kunstige mobilnettet microenvironments ved hjelp av en nanofiber-matrise og en microfluidic struktur: The multiplekset kunstig Cellular MicroEnvironment (MACME) matrise (Figur 1A og 2A). Nanofiber matrisen har 12 ulike microenvironments med ulike kombinasjoner av nanofiber materialer og tettheter. Electrospinning ble brukt til å dikte opp nanofibers. Nanofiber materialer, som polystyren (PS)21, polymethylglutarimide (PMGI)22og gelatin (GT)23, ble utviklet for å teste deres kjemiske egenskaper som kan påvirke celleadhesjon og vedlikehold av pluripotency ( Figur 2B). Nanofiber tettheter var varieres med electrospinning gang og de genererte nanofibers ble definert etter deres tettheter (BRUTT, med D = XLow/lav/Mid/høy). Microfluidic strukturen består av polydimethylsiloxane (PDMS) skjuler 48 celle-kultur chambers, som kan plasseres i standard dimensjoner i 96-brønnen microplate. PDMS er en biokompatible og gass-utskiftbare polymer som vanligvis brukes til å dikte microfluidic enheter24. Hver mikrovæskekanalen laget som 700-µm bred og 8.4 mm lang og hadde to innganger i kantene (tabell 1). Chambers hadde ulike høyder (250 500 og 1000 µm) å manipulere de første cellen-såing tettheter (0,3 og 0,6 1.2 × 105 celler/cm2), som kan relatere til overlevelse, spredning og differensiering av hPSCs25 (Figur 2C). Antall celler seeded i et kammer er proporsjonal med kolonnen tetthet over kammeret gulvet, og dermed første celle seeding tetthet ble kontrollert ved å innføre samme celle suspensjon i kultur kamre med ulike høyder. Alle kanaler var ment å være ≥ 250 µm høy26 å redusere effekten av lav oksygen spenning27 og skjæring stress28 på cellene. Kanal høyder av 250 500 og 1000 µm er forkortet her som XCD x = lav, Mid, og høy, henholdsvis. Miljøer med forskjellige nanofiber tettheter og første celle-såing tettheter var forkortet som "Material_NF density_Cell tetthet" (f.eks GT_HighNF_HighCD: et miljø preget av høy tetthet GT nanofibers og høy første celle-såing tetthet).
Deretter beskriver vi hvordan å utføre encellede analyser systematisk undersøke celle atferd svar for miljøfaktorer (figur 1B). Som en proof-of-concept identifisert vi det optimale mobilnettet miljøet for hPSC selv-fornyelse, som er en viktig funksjon hPSC vedlikehold (figur 1B)29. Image-basert cytometri, etterfulgt av statistiske analyser, gir kvantitative tolkning av individuelle mobilnettet fenotypiske svar på cellular miljøer. Blant en rekke cellulære funksjoner gir dette dokumentet en detaljert prosedyre for å identifisere de optimale forholdene for å opprettholde hPSC selvtillit fornyelse.

Mobilnettet microenvironments består av en rekke signaler, som vekstfaktorer, ekstracellulære matriser og intercellulære interaksjoner. Disse pekepinner er godt organisert og er avgjørende i å regulere celle funksjoner i en levende system. Selv om mange forskere har forsøkt å undersøke sammenhengen mellom miljøfaktorer og ønsket cellulære funksjoner, fortsatt mye ukjent. Dette er hovedsakelig på grunn av mangel på en riktig metode for å etterligne slike miljømessige signaler i vitro, og samtidig teste ulike miljømessige signaler på celler. Her rapporterer vi et integrert plattform microfluidic kanaler og en nanofiber matrise, etterfulgt av høyt innhold én celle analyse, undersøke stamcelleforskningen fenotyper endres av forskjellige miljøfaktorer. For å demonstrere anvendelsen av denne plattformen, fokuserer denne studien på av fenotyper av selvtillit fornyelse menneskelige pluripotent stamceller (hPSCs).
