Hier berichten wir über eine einfache Methode zur Herstellung von MWCNTs mit hinterlegten Ag Nanopartikel. Diese silberhaltigen Nanomaterial zeigt erhebliche antibakterielle Aktivität und minimalen Potenzial für unkontrollierte Aufnahme von Silber-Nanopartikeln in den Körper. Wir zeigen, dass 30 µg/mL der synthetisierten Ag-MWCNTs ist eine wirksame antibakterielle Aktivität gegen Methylobacterium spp. mit vernachlässigbar Zytotoxizität für Leber Säugerzellen. Das ultimative Ziel dieser Studie ist zu umweltfreundlichen antibakteriellen Nanomaterialien, die das Wachstum von Bakterien hemmen können diese Form Biofilmen. Diese antibakteriellen Nanomaterialien haben das Potenzial, die Toxizität und Antibiotikum Resistenzprobleme von häufig verwendeten Chemikalien oder Antibiotika chemischer Verbindungen zu überwinden. Ein Biofilm ist eine hydratisierte extrazellulären Polymeren Substanz (EPS), die besteht aus Polysacchariden, Proteinen, Nukleinsäuren und Lipiden1,2. Biofilme verhindern das Eindringen von Fremdstoffen und helfen Bakterien3,4kräftig wachsen. Biofilme verursachen Geruch und chronische Infektionskrankheiten5,6. Methylobacterium Spp., zum Beispiel, wächst durch das Festhalten an Orten, wo immer Wasser vorhanden ist oder ist es schwierig, bakterielle Beseitigung auf einer kontinuierlichen Basis, wie z. B. Klimaanlage Wärmetauscher, Duschräume und medizinische Geräte zu gewährleisten. Diese Arten von Biofilmen auslösen Geruch und chronische Infektionskrankheiten5,6.
Normalerweise sind Chemikalien oder Antibiotika chemischen Verbindungen verwendet, um das Wachstum von Bakterien hemmen, die Biofilme bilden. Die Entstehung von Antibiotika-resistenten Bakterien und Sicherheitsbedenken in Vivo von Chemikalien fahren die Notwendigkeit zur Entwicklung neuer Materialien, um die Bildung von Biofilmen zu verhindern und das Wachstum von Bakterien hemmen.
In dieser Studie werden antimikrobielle Nanomaterialien synthetisiert, die frei von Antibiotika-Resistenz und Toxizität sind. Silber ist ein bekannter antimikrobieller Wirkstoff, und jüngste Entwicklungen im Bereich der Nanowissenschaften und Nanotechnologie führten zu aktive Forschung in die antimikrobielle Wirkung von Metall-Nanopartikeln7,8. Jüngste Studien haben berichtet, dass die geringe Größe und hohe Oberflächen-Volumen-Verhältnisses der Nanopartikel erhöhte antibakterielle Aktivität9,10,11führen.
Die enthaltenen Nanomaterialien kombinieren Silber-Nanopartikel mit erhöhten antimikrobielle Eigenschaften und Kohlenstoff-Nanoröhrchen mit einem hohen Seitenverhältnis, wodurch sich die Fläche pro Volumeneinheit. Die fabrizierten Silber Nanopartikel-Carbon-Nanotube-Composite weist beträchtliche antimikrobielle Eigenschaften und minimale Toxizität für menschlichen und tierischen Zellen. Die synthetischen Prozesse in früheren Studien verwenden gefährlicher Reduktionsmittel wie NaBH4, Formamid, Dimethylformamid und Hydrazin. Der Prozess ist kompliziert, gefährlich und zeitaufwendig. Das synthetische Verfahren berichtet verwendet hier Ethanol als deutlich weniger gefährlichen Reduktionsmittel.
Die Hemmung Zone und minimale bakterizide Konzentration (MBC) für die Ag-MWCNTs wurden gemessen; Lebenden/Toten und Trypan blau-Assays wurden zur Toxizität und antibakteriellen Eigenschaften zu messen. Minimalen inhibitorischen Konzentration (MIC) und mitochondriale Toxizität (MTT) Assays wurden nicht durch die ungewöhnliche Farbe der Kohlenstoff-Nanoröhren durchgeführt, die mit der Assays gestört haben würde. Zu guter Letzt wurde die minimale Konzentration zu verhindern das Wachstum von Methylobacterium spp. ohne Säugetier-Zellen bestimmt.

In dieser Studie wurden mit einer wässrigen Schwefelsäure-Lösung zu einer Sauerstoff-basierte Sachgruppe bilden Multi-walled Carbonnanotubes (MWCNTs) behandelt. Silber MWCNTs wurden von der reduktiven Abscheidung von Silber aus einer wässrigen Lösung von AgNO3 auf die oxidierte MWCNTs vorbereitet. Die einzigartige Farbe der CNTs war es nicht möglich, auf der minimalen inhibitorischen Konzentration oder mitochondriale Toxizität Assays, die Toxizität und antibakterielle Eigenschaften, zu bewerten, da sie die Assays behinderen würde anzuwenden. Die Hemmung Zone und minimale bakterizide Konzentration für die Ag-MWCNTs wurden gemessen und Live/Dead und Trypan blau-Assays wurden verwendet, um die Toxizität und antibakterielle Eigenschaften zu messen, ohne die Farbe der CNTs zu stören.

Transmission Electron Microscopy (TEM) Bilder bestätigen die Bildung der Ag-MWCNTs (Abb. 1A und 1 b). Ihre gelungene Synthese wurde durch die Veränderung der Oberflächenladung bestätigt.
