Dit protocol beschrijft de geheel-mount immunonohistochemical beeldvorming van volwassen oor huid voor de analyse van de neuro-vasculaire structuren en immuun cel distributie. Huid bestaat uit drie lagen: de epidermis, de dermis en de hypodermis. Het is gebruikt als een modelsysteem om te studeren stamcel onderhoud, differentiatie en voedselproductie in ontwikkeling evenals de regeneratie, tumorvorming en ontstekingen bij volwassenen. Huid is rijkelijk gevacuoliseerd en geïnnerveerd zodanig zijn dat de ontwikkeling van het perifere zenuwstelsel en vasculaire systeem goed gecoördineerd.
Wij hebben eerder aangetoond een geheel-mount embryonale huid beeldvormende techniek met meerdere labelen om te studeren intact perifere zenuwen en bloedvaten, met inbegrip van hun cellulaire componenten1,2,3, 4: sensorische axonen, sympathieke axonen Schwann cellen in zenuwen, endotheliale cellen, pericytes en vasculaire zachte spiercellen (VSMCs) in de bloedvaten. Tijdens de angiogenese, een primaire capillaire netwerk ondergaat intensieve vasculaire remodeling en ontwikkelt zich tot een hiërarchisch vasculaire vertakkende netwerk. In de ontwikkelingslanden dermis/hypodermis, slagaders vertakken naast perifere sensoriële zenuwen en aders dan vormen grenzend aan de slagaders. Nadat het hiërarchische vasculaire netwerk grondig met VSMCs bedekt is, sympathische zenuwen uitbreiden langs en innervate grote diameter bloedvaten1,5,6. Ondanks het belang in de nauwe samenwerking tussen de ontwikkelingslanden nerveus en vasculaire systemen, een grote vraag geweest om te pakken wat gebeurt er met de neuro-vasculaire netwerken in diverse pathologische situaties bij volwassenen. Een driedimensionale high-resolution imaging is noodzakelijk om te waarderen de pathogenese, samen met anatomisch herkenbaar vertakkende genuitdrukking en patronen.
Neuronale en vasculaire morfogenese in volwassen muis huid is vaak geanalyseerd door weefsel sectie kleuring. Andere studies hebben geheel-mount beeldvorming van de huid gebruikt voor het visualiseren van de perifere zenuwen en bloedvaten, naast de haarzakjes sebaceous klieren en de arrector pili spieren7,8,9. Echter, de dikte van volwassen huid heeft het moeilijk gemaakt voor het analyseren van de huid over de gehele diepte.
In de huidige studie ontwikkelden we een nieuwe hoge resolutie geheel-mount beeldvorming van volwassen oor huid om deze uitdagingen te overwinnen. Oor huid is gemakkelijk toegankelijk zijn voor dissectie en latere geheel-mount beeldvorming van de huid over de gehele diepte. Dus, het is een eenvoudig en zeer reproduceerbare methode die kan worden toegepast om te vergelijken van driedimensionale architectuur van perifere zenuw- en vasculaire systemen in de huid, met uitgebreide kwantificering metingen. We toonden aan dat de uitlijning van perifere zintuiglijke en sympathische zenuwen met grote diameter bloedvaten bewaard in de volwassen huid gebleven is. Het doel van dit protocol is te visualiseren vertakkende genuitdrukking en de patronen van perifere zenuwen en bloedvaten, evenals de immuun cel distributie op een cellulair niveau in volwassen Muismodellen in verschillende omstandigheden zoals ontsteking en regeneratie weer gecontroleerd.

Hier presenteren we een protocol van de huid van een geheel-mount volwassen oor imaging techniek om te bestuderen van de uitgebreide driedimensionale neuro-vasculaire vertakkende genuitdrukking en patronen, evenals immuun cel distributie op een cellulair niveau. De analyse van perifere zenuwen en bloedvat anatomische structuren in volwassen weefsels biedt sommige inzicht in het begrip van functionele neuro-vasculaire bedrading en neuro-vasculaire degeneratie in pathologische omstandigheden zoals de wondgenezing. Als een zeer informatief modelsysteem, hebben we onze studies gericht op volwassen oor huid, dat gemakkelijk toegankelijk zijn voor dissectie is. Onze eenvoudige en reproduceerbare protocol biedt een nauwkeurige voorstelling van de cellulaire componenten in de gehele huid, zoals de perifere zenuwen (sensorische axonen, sympathieke axonen en cellen van Schwann), bloedvaten (endotheliale cellen en vasculaire zachte spiercellen ), en ontstekingscellen. Wij geloven dat dit protocol zal de weg effenen naar onderzoeken morfologische afwijkingen in het perifere zenuwen en bloedvaten en de ontsteking in de huid van de volwassen oor onder verschillende pathologische condities.
