Denne protokollen beskriver hele-mount immunonohistochemical avbilding av voksen øre hud for analyse av Nevro vaskulære strukturer og immun celle distribusjon. Huden består av tre lag: overhuden, dermis og epidermis. Det har blitt brukt som et modellsystem for å studere stamcelleforskningen vedlikehold, differensiering og morphogenesis i utvikling og regeneration, tumorigenesis og betennelse i voksen. Huden er rikt Stangeriaceae og innerveres slik at utviklingen av det perifere nervesystemet og vaskulære system er godt koordinert.
Vi har tidligere vist en tenkelig teknikk som helhet-mount embryonale huden med flere merking for å studere intakt perifere nerver og blodkar inkludert deres cellulære komponenter1,2,3, 4: sensoriske axons sympatisk axons, Schwann celler i nerver, endotelceller, pericytes og vaskulær glatt muskelceller (VSMCs) i blodkar. Under angiogenese, en primær capillary nettverket gjennomgår intensiv vaskulær remodeling og utvikler seg til en hierarkisk vaskulær forgrening nettverk. I utviklingsland dermis/epidermis, arterier gren perifere sensoriske nerver og årer og danne ved siden av arteries. Når hierarkisk vaskulære nettverket er grundig dekket med VSMCs, sympatiske nerver strekker seg og innervate stor-diameter blodkar1,5,6. Til tross for betydningen i nær tilknytning mellom de utvikle nervesystemet og vaskulære systemene er et viktig spørsmål å ta hva skjer til Nevro vaskulære nettverk i ulike patologisk situasjoner i voksne. En tredimensjonal høyoppløselige bilder er nødvendig å verdsette patogenesen, anatomisk gjenkjennelig branching morphogenesis og mønstre.
Neuronal og vascular morphogenesis i voksen mus huden er vanligvis analysert av vev delen flekker. Andre studier har brukt hele-mount avbilding av huden for å visualisere perifere nerver og blodkar, i tillegg til hårsekkene, talgkjertlene og arrector pili muskler7,8,9. Men har tykkelsen av voksen hud gjort det vanskelig å analysere huden over hele dybden.
Studien utviklet vi en roman høyoppløselig hele-mount avbilding av voksen øre hud å løse disse utfordringene. Øre hud er lett tilgjengelig for disseksjon og påfølgende hele-mount bildebehandling av huden over hele dybden. Således, det er en enkel og svært reproduserbar metode som kan brukes til å sammenligne tredimensjonale arkitektur av perifere nervesystemet og vaskulære systemer i huden, med omfattende kvantifisering målinger. Vi viste at justeringen av eksterne sensorisk og sympatiske nerver med stor-diameter blodkar opprettholdes i voksen huden. Målet med denne protokollen er å visualisere branching morphogenesis og mønstre av eksterne nerver og blodkar, samt immun celle fordelingen på cellenivå i voksen mus modeller i ulike forhold som betennelse og regenerering.

Her presenterer vi en protokoll av en hel-mount voksen øre hud tenkelig teknikk for å studere omfattende tredimensjonale Nevro-vaskulær branching morphogenesis og mønstre, samt immun celle distribusjon på cellenivå.
