Autophagie wurde ursprünglich gedacht, um ein nicht-selektiven Verfahren zur Masse Abbau von beschädigten Proteinen, aber neuere Studien haben gezeigt, dass Autophagie ist sehr selektiv zu den Katabolismus von Protein-Aggregate und dysfunktionalen Organellen, die nicht Abbau über andere Protein-Clearance Mechanismen11zugänglich. Der Darm von C. Elegans ist das große metabolische Gewebe in Worms und es ist eine kritische Regulator des systemischen Homöostase und Lebensdauer. Der Fadenwurm Caenorhabditis Elegans (C. Elegans) ist ein Modell, das häufig verwendet wird, um Langlebigkeit und Entwicklungsstörungen Wege zu studieren. Solche Studien werden erleichtert durch die Transparenz des Tieres, die Fähigkeit, weiterleiten und umkehren, genetische Tests, die relative Leichtigkeit der Generierung von eindringmittel markierte Proteine und die Verwendung von Fluoreszenzfarbstoffen, die entweder in den frühen mikroinjiziert werden können Embryo oder integriert in seine Nahrung (E. Hier präsentieren wir Ihnen die Verwendung von einem pH-sensitiven Fluoreszenzfarbstoff, der Darm Lysosomen Flecken bietet eine visuelle Auslesen der dynamischen, physiologische Veränderungen in lysosomalen Säure im Leben Würmer. Dieses Protokoll misst nicht lysosomale pH-Wert, sondern vielmehr soll eine zuverlässige Methode zur Beurteilung relevanten physiologischen Variationen in lysosomalen Säure. cDCFDA ist eine Zelle-permeationsfähigen Substanz, die in den fluoreszierenden Fluorophor 5-(and-6)-carboxy-2',7'-dichlorofluorescein (cDCF) nach Hydrolyse von intrazellulären Esterasen umgewandelt wird. Protonierung in Lysosomen fallen cDCF in diese Organellen, wo es sich ansammelt. Aufgrund seiner niedrigen pKa von 4,8 wurde dieser Farbstoff als pH-Sensor in der Hefe verwendet. Hier beschreiben wir die Verwendung von cDCFDA als Nahrungsergänzungsmittel zu beurteilen, den Säuregehalt des intestinalen Lysosomen in C. Elegans. Diese Technik ermöglicht die Erkennung von alkalisierenden Lysosomen mit lebenden Tieren, und hat ein breites Anwendungsspektrum experimentelle Studien über das Altern, Autophagie und lysosomale Biogenese einschließlich.
