Hier, zijn eenvoudig in vitro methoden beschreven waarmee de demonstratie van de generatie van cytotoxische amyloids tijdens de infectie met een longontsteking veroorzaken organisme. Patiënten die longontsteking overleven hebben verheven sterftecijfers in de maanden na ziekenhuis geen kwijting1,2,3,,4,,5,6. In de meeste gevallen overlijden door een soort eind-orgaanfalen zoals renale, pulmonale, cardiale, of lever evenementen, evenals lijn5,6. De reden voor de verhoogde sterfte in deze populatie is nooit vastgesteld.
Longontsteking is geclassificeerd als zijnde hetzij communautaire verworven of ziekenhuizen opgelopen (nosocomiale) en agenten die longontsteking kunnen veroorzaken bevatten bacteriën, virussen, schimmels en chemicaliën. Een van de belangrijkste oorzaken van nosocomiale pneumonie is de bacterie Pseudomonas aeruginosa. P. aeruginosa is een gram-negatieve organisme waarmee een type III secretie systeem overbrengt van verschillende effector moleculen, genoemd exoenzymes, rechtstreeks naar het cytoplasma van target cellen7,8. Tijdens de infectie van pulmonaire endotheliale cellen richten de exoenzymes verschillende intracellulaire eiwitten, met inbegrip van een endotheel vorm van de microtubulus-geassocieerde eiwit tau9,10,11,12 , leidt tot endothelial barrière verdeling resulterend in ernstige longoedeem, daalde pulmonaire functie en vaak dood.
Zoals eerder vermeld, hebben patiënten die de eerste longontsteking overleven sterftecijfers in de eerste 12 maanden na ziekenhuis kwijting verheven. Een potentiële mechanisme voor het verklaren van dit verschijnsel is dat een soort van langlevende toxine wordt gegenereerd tijdens de initiële infectie die tot slechte op lange termijn resultaat leidt. Twee opmerkingen ondersteunen deze mogelijkheid. Eerste, gekweekte pulmonaire endotheliale cellen die in eerste instantie worden behandeld met P. aeruginosa niet vermenigvuldigen voor tot een week nadat de bacteriën worden gedood door de antibiotica13. Tweede, langlevende prionen en agenten met prion kenmerken zijn aangetoond in verschillende menselijke en dierlijke ziekten, met name het zenuwstelsel14,15is gekoppeld.
Methoden voor de behandeling van de potentiële productie van langlevende cytotoxische agenten tijdens de pulmonaire infectie zijn nooit beschreven. Hier een aantal eenvoudige in vitro testen worden beschreven die kunnen worden gebruikt voor het onderzoeken van de cytotoxin productie en activiteit na infectie met behulp van een gemeenschappelijk longontsteking veroorzaken agent, P. aeruginosa. Deze testen moeten gemakkelijk aan te passen aan het onderzoeken van mogelijke cytotoxin inductie na infectie met behulp van andere stoffen die longontsteking veroorzaken, en het supernatant die worden gegenereerd ook moet nuttig zijn voor het onderzoeken van de effecten van de cytotoxins geheel organen of dieren. Tot slot zal de tests die worden beschreven hier waarschijnlijk worden aangepast aan het testen van dierlijke en menselijke biologische vloeistoffen voor de productie van cytotoxins tijdens longontsteking.

Patiënten die longontsteking overleven hebben verheven sterftecijfers in de maanden na ziekenhuis kwijting. Het heeft zijn veronderstelde dat besmetting van pulmonaire weefsel tijdens longontsteking resulteert in de productie van langlevende cytotoxins die tot daaropvolgende einde orgaanfalen leiden kunnen. Wij hebben ontwikkeld in vitro testen om te testen van de hypothese dat cytotoxins worden geproduceerd tijdens de pulmonaire infectie. Geïsoleerde rat pulmonaire endotheliale cellen en de bacterie Pseudomonas aeruginosa worden gebruikt als modelsystemen, en de productie van cytoxins na infectie van de endotheliale cellen van de bacteriën wordt aangetoond door middel van cultuur van de cel gevolgd door directe kwantificatie met behulp van lactaat dehydrogenase tests en een nieuwe microscopische methode ImageJ technologie. De amyloïde aard van deze cytotoxins werd aangetoond door thioflavin T bindende analyses en door immunoblotting en immunodepletion met behulp van de A11 anti-amyloid antilichaam. Verdere analyses met behulp van immunoblotting aangetoond dat oligomere tau en Aβ werden geproduceerd en uitgebracht door endotheliale cellen na infectie door P. aeruginosa. Deze methoden moeten gemakkelijk aanpasbaar aan analyses van menselijke klinische monsters.

Een eenvoudige in vitro -assay is ontwikkeld om assay voor de aanwezigheid van cytotoxins in supernatant van cellen die zijn besmet met de bacterie P. aeruginosa.
