Proteaser regulere fysiologiske processer ved holde proteiner, og indledningen af protease aktivitet på bestemte tidspunkter og steder er reguleret1. Det er derfor vigtigt at identificere udtryk og aktivitet af væv, der reguleres lokalt og udvikle en ny metode til at registrere proteaser. Metoden, der foreslås i denne undersøgelse har til formål at identificere protease specificitet i parallel til at afsløre proteaser.
Der er nogle metoder til påvisning af protease specificitet. Metoden azocasein er kendt for sin brug i påvisning af proteaser2 men er begrænset i sin evne til at indhente yderligere oplysninger. Zymography er en anden omfattende protease påvisningsmetode, der kan bruges til at bestemme molekylvægt af proteaser3, men det kan ikke bruges til at undersøge substrat specificitet. Protease specificitet kan bestemmes af spektrometrisk assays, ved hjælp af substrater såsom p-nitroanilide4, og fluorometriske assays, ved hjælp af cumarin substrater5 eller fluorescens resonans energi overføre (FRET) assays6. Disse metoder aktiverer single-specificitet påvisning af substrater indeholdt i en enkelt brønd. I den foreliggende undersøgelse undersøges protease specificiteten af væv ekstrakter under forskellige betingelser, som disse ekstrakter indeholder flere proteaser. Protease aktivitet er reguleret af flere faktorer, herunder pH, coenzymer, prostetiske grupper og ion styrke. Dette papir beskriver en metode til at vurdere protease specificitet ved hjælp af MALDI-TOF-massespektrometri til at måle molekylvægt af unikke substrater. De molekylære former for kløvet substrater, sammen med deres teoretiske Molekylær vægte, er vist i figur 1 og tabel 1. Tidligere undersøgelser har brugt substrater indeholdende polyglycine spacere og biotin9; men disse substrater er mangelfuldt, fordi polyglycine sekvens kan kløves af enzymer, der anerkender glycin sekvensen. Derudover kan høj affinitet mellem begærlighed og biotin føre til en lav inddrivelsessats. At forbedre disse ulemper, i denne undersøgelse vi syntetiseret et unikt underlag, som var sammensat af polyethylenglycol (PEG), iminobiotin og en aminosyre, der kunne identificere kavalergang websted (figur 1). For at skelne mellem aminosyrer af lignende Molekylær vægte, blev D-Serin tilføjet mellem PIND spacer og aminosyre på webstedet kløvet.
N-terminalen af substratet var mærket med iminobiotin, som muliggør affinitet rensning fra rå prøver. Brug af iminobiotin i stedet for biotin er kritisk. Biotin har en stærk affinitet med begærlighed, hvilket resulterer i lav genfindingsprocent biotin-mærket underlag fra begærlighed harpiks, mens iminobiotin's affinitet for begærlighed kan ændres af pH. Iminobiotin-mærket substrater vil binde sig til begærlighed i betingelserne ovenfor pH 9, mens begærlighed frigiver iminobiotin-mærket substrater i betingelser under pH 4. Proteaser har flere biologiske funktioner, herunder protein Aktivering/inaktivering og fødevarer fordøjelse. At identificere protease specificitet er vigtigt for at afsløre protease funktion.
