Satserne for kolonien dannelse fremstillet med MCBM afhænger celletype. Normalt, for ikke-tilhænger celler, antallet af kolonier er meget højere i forhold til vedhængende celler. Kræft er forårsaget af cellulære mutationer og følgelig biokemiske signaling veje afbrydes udløser ukontrolleret celledeling og modstand til celledød. Tumorer forstyrre fysiologiske funktioner i fordøjelsessystemet, nervøs, kredsløbssystemet og efterfølgende omkringliggende væv1. Trods omfattende forskningsindsats, kræft er stadig den mest udbredte livstruende sygdom i verden2. Precision medicin er blevet anerkendt som grundlaget for fremtidige kræft therapeutics. Nye molekylære enheder testes rutinemæssigt i kombination med eksisterende stoffer for at forbedre kliniske udfald.
En af de betydelige begrænsninger i forbindelse med udviklingen af nye effektive målrettede behandlinger er imidlertid den manglende almindeligt anvendte assays til at simulere det nøjagtige biologiske resultatet af drug eksponering3. Kræft drug discovery er stadig for det meste afhængig teste effekten af terapeutiske agenter i kræft cellelinjer kulturperler i 2D éncellelag kulturer, som er vanskelige at validere i kliniske forsøg4. Derfor er der en stigende interesse i at udvikle bedre kræftmodeller, der bedre efterligner de indfødte funktioner af tumorer i vivo5. I de seneste år resulterede en stigende interesse i 3D kultur modeller i udviklingen af forbedrede metoder til at fremstille 3D tumor modeller5.
Her præsenterer vi en strategi med tre forskellige 3D celle kultur teknikker gør det muligt for at forbedre forståelsen af styrken af hydroxycoumarin OT486 i kombination med BH3 mimetics før mere i dybden i vivo assays. Vores metode består af kombinerer koloni og SFAs med en i vivo svulst dannelse test baseret på en zebrafisk model til yderligere validering af effekten af kombination af OT48 og BH3 mimetics i lunge kræftceller og overvåge kræft progression i en levende organisme.
Koloni dannelse assays er rutinemæssigt anvendes til at vurdere effekten af anticancer agenter for både solid såvel som hæmatopoietisk kræftformer. Analysen bestemmer evne til en celle for at formere sig på ubestemt tid og danne kolonier7. Effekten af anticancer agent på kolonidannende evne til celler bestemmes af fald i antallet og/eller størrelsen af kolonierne.
Spheroids repræsenterer in vitro- tumor modeller og tjene som en billig screening platform for anticancer agenter. Spheroids er aggregater af celler vokser i suspension eller indlejret i et 3D matrix. Denne tilgang er meget udbredt til stof screening og undersøgelser af tumor vækst, spredning og immun interaktion8.
Fuldt ud at forstå egenskaberne for et nyt lægemiddel, er det vigtigt at gennemføre i vivo forsøg på gnavere. Denne konventionelle metode er imidlertid dyrt og tidskrævende. I de seneste år blev zebrafisk (Danio rerio) et almindeligt studerede laboratorium organisme, der er billigere og hurtigere at rejse. Tumorer udviklet i zebrafisk repræsenterer en 3D celle kultur tilgang, men inden i vivo indstilling af en hvirveldyr9.
Helt, vi bruger her tre forskellige 3D kultur metoder herunder CFAs, SFAs og zebrafisk i vivo tumordannelse for at påvise kræft kapaciteten af hydroxycoumarin OT48 i en lunge cancer A549 celle model i kombination med BH3 mimetics.

In vitro og i vivo præklinisk screening af nye terapeutiske agenter er et vigtigt redskab i kræft drug discovery. Selv om menneskelige cancer cellelinjer reagere terapeutiske forbindelser i 2D (dimensional) éncellelag cellekulturer, blev 3D kultur systemer udviklet for at forstå effekten af lægemidler i mere fysiologisk relevante modeller.
