De tarieven van de kolonie vorming verkregen met MCBM, is afhankelijk van het celtype. Voor niet-aanhanger cellen, wordt het aantal kolonies meestal veel hoger vergeleken met aanhangend cellen. We hebben vastgesteld dat de A549 cellen gevormd 30 tot 40 kolonies na 10 dagen. Eerder hebben we gemeld voor verschillende leukemie cellen dat aantal kolonies tussen de 200 tot 2509 is. Kanker wordt veroorzaakt door cellulaire mutaties en dientengevolge de biochemische signaalroutes triggering van ongecontroleerde celdeling en weerstand tegen celdood worden verstoord. Tumoren interfereren met fysiologische functies van spijsverteringsstelsel, zenuwachtig, bloedsomloop systems en vervolgens naburige weefsels1. Ondanks uitgebreid onderzoeksinspanningen blijft kanker de meest voorkomende levensbedreigende ziekte in de wereld2. Precisie geneeskunde is erkend als basis van toekomstige cancer therapeutics. Nieuwe moleculaire entiteiten worden regelmatig getest in combinatie met bestaande geneesmiddelen ter verbetering van de klinische resultaten.
Een van de belangrijke beperkingen in verband met de ontwikkeling van nieuwe efficiënte gerichte therapieën is echter de mislukking van veelgebruikte tests simuleren de exacte biologische uitkomst van drug blootstelling3. Kanker drugontdekking is nog steeds voornamelijk gebaseerd op testen van de werkzaamheid van de therapeutische agenten in kanker cellijnen gekweekte in 2D enkelgelaagde culturen, die moeilijk te valideren in klinische proeven4. Daarom is er een groeiende belangstelling voor de ontwikkeling van betere kanker modellen die beter na te de ingebouwde functies van tumoren in vivo5 bootsen. In de afgelopen jaren resulteerde een groeiende interesse in 3D cultuur modellen in de ontwikkeling van verbeterde methodologieën voor het produceren van 3D tumor modellen5.
Hier presenteren we een aanpak met drie verschillende 3D cel cultuur technieken waardoor ter verbetering van het inzicht in de potentie van de hydroxycoumarin OT486 in combinatie met BH3 mimetics voordat meer in de diepte in vivo tests. Onze werkwijze bestaat uit de kolonie en SFAs te combineren met een in vivo tumor vorming test gebaseerd op een model van de zebravis verder de werkzaamheid van de combinatie van OT48 en BH3 mimetics in Long kankercellen valideren en controleren van de progressie van kanker in een levende organisme.
Kolonie vorming testen worden routinematig gebruikt om te beoordelen van de werkzaamheid van anticancer agents voor zowel vaste als hematopoietische kankers. De test bepaalt de mogelijkheid van een cel te vermenigvuldigen voor onbepaalde tijd en kolonies7te vormen. Het effect van een antikanker agent op de kolonievormende vermogen van de cellen wordt bepaald door de daling van het aantal en/of grootte van de kolonies.
Spheroïden in vitro tumor modellen vertegenwoordigen en dienen als een goedkope screening platform voor anticancer agents. Spheroïden zijn aggregaten van cellen groeien in suspensie of ingesloten in een 3D-matrix. Deze aanpak wordt veel gebruikt voor drug screening en studies van tumor groei, proliferatie en immuun interactie8.
Om volledig te begrijpen de eigenschappen van een nieuwe drug, is het essentieel om uit te voeren in vivo experimenten met knaagdieren. Deze conventionele methode is echter duur en tijdrovend. In de afgelopen jaren werd zebravissen (Danio rerio) een grote schaal bestudeerde laboratorium organisme dat is goedkoper en sneller te verhogen. Tumoren in de zebravis vertegenwoordigen een 3D cel cultuur benadering maar binnen de in vivo -instelling van een gewervelde9ontwikkeld.
Over het geheel genomen gebruiken we hier drie verschillende 3D cultuur benaderingen, met inbegrip van GVO, SFAs en zebrafish in vivo de vorming van de tumor om aan te tonen van de antikanker capaciteit van hydroxycoumarin OT48 in een long kanker A549 cel model in combinatie met BH3 mimetics.

In vitro en in vivo pre-klinische screening van nieuwe therapeutische agenten zijn een essentieel onderdeel van de Geneesmiddelenontwikkeling kanker. Hoewel menselijke kanker cellijnen inspelen op de therapeutische bestanddelen in 2D (dimensionale) enkelgelaagde celculturen, werden 3D cultuur systemen ontwikkeld om te begrijpen van de werkzaamheid van geneesmiddelen in meer fysiologisch relevante modellen. In de afgelopen jaren, werd een paradigmaverschuiving waargenomen in de pre-klinisch onderzoek voor het valideren van de potentie van nieuwe moleculen in 3D cultuur systemen, preciezer het nabootsen van de communicatie van de tumor. Deze systemen karakteriseren de ziekte staat in een meer fysiologisch relevante wijze en helpen om beter mechanistische inzicht en begrip van de farmacologische werkzaamheid van een bepaalde molecule te winnen. Hier, we onderzocht de therapeutische werking van hydroxycoumarin OT48 alleen of in combinatie met BH3 mimetics in long kanker cellijn A549 met behulp van drie verschillende 3D cultuur systemen met inbegrip van de kolonie vorming assays (CFA), sferoïde vorming assay (SFA) en in vivo zebrafish xenografts.

In Figuur 2cel longkanker lijn A549 was uitgezaaid in MCBM aan vorm kolonies na behandeling met OT48 alleen of in combinatie met BH3 mimetische A1210477 bij de aangegeven concentraties. De resultaten toonden aan dat de combinatie aanzienlijk verminderd aantal, de grootte en de totale oppervlakte van de koloniën na 10 dagen incubatie.
