Die Preise der Kolonie Formation mit MCBM erhalten richtet sich nach dem Zelltyp. Für nicht-anhaftende Zellen, wird die Anzahl der Kolonien in der Regel viel höher mit adhärenten Zellen verglichen. Krebs wird durch zelluläre Mutationen verursacht und folglich biochemischen Signalwege gestört werden, unkontrollierte Zellteilung und Widerstand zum Zelltod auszulösen. Tumoren stören mit physiologischen Funktionen von Verdauungs, Nervensystem, Herz-Kreislauf-Systems und anschließend benachbarte Gewebe1. Trotz umfangreicher Forschungsbemühungen bleibt Krebs die häufigste lebensbedrohliche Krankheit in der Welt2. Präzisions-Medizin wurde als Grundlage für zukünftige Krebstherapie anerkannt. Neue Wirkstoffe werden routinemäßig getestet in Kombination mit vorhandenen Drogen, klinische Outcome zu verbessern.
Eine erhebliche Einschränkung verbunden mit der Entwicklung der neuen effiziente zielgerichtete Therapien ist jedoch das Scheitern von häufig verwendeten Assays, die genaue biologische Ergebnis der Droge Exposition3zu simulieren. Krebs Wirkstoffforschung stützt sich nach wie vor überwiegend auf testen die Wirksamkeit therapeutischer Wirkstoffe in Krebszelllinien kultiviert in 2D Monolayer-Kulturen, die schwer in klinischen Studien4zu validieren sind. Daher gibt es ein wachsendes Interesse, bessere Krebs Modelle zu entwickeln, die die systemeigene Funktionen von Tumoren in Vivo5besser zu imitieren. In den letzten Jahren führte ein zunehmendes Interesse an 3D Culture Modelle die Entwicklung von verbesserten Methoden, 3D Tumor Modelle5zu produzieren.
Hier stellen wir einen Ansatz mit drei verschiedenen 3D Zelle Kulturtechniken zur Verbesserung des Verständnisses der Potenz des Hydroxycoumarin OT486 in Kombination mit BH3-Mimetika vor mehr in Tiefe in Vivo Tests. Unsere Methode besteht darin, Kolonie und SFAs mit einem in Vivo Tumor Bildung Test basiert auf einem Zebrafisch-Modell weiter bestätigen die Wirksamkeit der Kombination von OT48 und BH3-Mimetika in Lungenkrebszellen und überwachen Krebsentwicklung in einer lebendigen verbinden Organismus.
Kolonie-Bildung-Assays sind routinemäßig zur Wirksamkeit der Anti-Krebs-Agents für solide sowie hämatopoetischen Krebsarten zu beurteilen. Der Test bestimmt die Fähigkeit einer Zelle zu vermehren auf unbestimmte Zeit und bilden Kolonien7. Die Wirkung von Anti-Krebs-Agent auf die koloniebildenden Fähigkeit der Zellen wird durch Abnahme der Anzahl und/oder Größe der Kolonien bestimmt.
Sphäroide vertreten in Vitro Tumormodellen und dienen als eine kostengünstige Screening-Plattform für Anti-Krebs-Agenten. Sphäroide sind Aggregate von Zellen in Suspension wachsen oder in einer 3D Matrix eingebettet. Dieser Ansatz ist für Wirkstoff-Screening und Studien von Tumor Wachstum, Vermehrung und Immune Interaktion8verbreitet.
Um die Eigenschaften eines neuen Medikaments vollständig zu verstehen, gilt es in Vivo Experimente auf Nagetiere durchzuführen. Diese herkömmliche Methode ist jedoch teuer und zeitaufwendig. Zebrafisch (Danio Rerio) wurde in den letzten Jahren ein weit studierte Labor-Organismus, der ist billiger und schneller zu erhöhen. Tumoren entwickelt im Zebrafisch repräsentieren eine 3D Zelle Kultur Ansatz aber innerhalb der in Vivo -Einstellung eines Wirbeltier-9.
Insgesamt verwenden wir hier drei verschiedene 3D Culture Ansätze einschließlich aufgewendete, SFAs und Zebrafisch in Vivo Tumorbildung um die Anti-Krebs der Hydroxycoumarin OT48 in einem Lungenkrebs Krebs A549 Zellmodell in Kombination mit BH3-Mimetika unter Beweis zu stellen.

In Vitro und in Vivo prä-klinischen Screening von neuartiger Therapeutika sind ein wesentliches Instrument in der Drogeentdeckung Krebs. Obwohl menschliche Krebszelllinien therapeutischen Verbindungen in 2D (dreidimensionale) Monoschicht Zellkulturen Rechnung zu tragen, wurden 3D Culture Systeme entwickelt, um die Wirksamkeit von Medikamenten in mehr physiologisch relevanten Modelle zu verstehen. In den letzten Jahren verzeichneten ein Paradigmenwechsel in der präklinischen Forschung, die Potenz der neuen Moleküle in 3D Culture Systeme, genauer imitiert den Tumor Mikroumgebung zu validieren. Hier untersuchten wir die therapeutische Wirksamkeit des Hydroxycoumarin OT48 allein oder in Kombination mit BH3-Mimetika in Lunge-Krebs-Zell-Linie A549 mithilfe von drei verschiedenen 3D Culture-Systeme einschließlich der Kolonie Bildung Assays (CFA), Sphäroid Bildung Assay (SFA) und in Vivo Zebrafisch Xenotransplantate.

In Abbildung 2Lungenkrebs Zelllinie, die A549 in MCBM Form Kolonien nach der Behandlung mit OT48 allein oder in Kombination mit BH3 mimetischen A1210477 in den angegebenen Konzentrationen ausgesät wurde.
