Le principal inconvénient de l’utilisation d’un commutateur d’expression de gène binaire est la nécessité d’offrir simultanément deux plasmides distincts (pilote et effectrices) dans la cellule cible. Cela peut conduire à une répartition inégale des plasmides et aboutir à une réponse incohérente du commutateur pour les inducteurs1,2,3. Une autre lacune du système deux-vecteur est qu’un minimum de deux tours...

Plusieurs gènes différents commutateurs ont été étudiés pour leur capacité à précisément régulent l’expression de transgènes dans les cultures cellulaires et des modèles animaux, avec des degrés de succès1,2,3. Des systèmes de commutation de gènes potentiels se réunisse plusieurs critères stricts, notamment : précise directionnelle expression du transgène, réactivité de la fonction du dosage et temps à inducteurs, de perméabilité négligeable du promoteur, de réversibilité du transgène activation par le biais de suppression de l’inducteur et des niveaux de toxicité inducteur tolérables de lignées cellulaires et les animaux de laboratoire1,2,3. Plusieurs inducteurs de petites molécules ont été exploitées, y compris la tétracycline4,5, la rapamycine6,7,8, mifépristone9,10, et l’ecdysone11,12,13,14. En général, ces systèmes nécessitent au moins deux plasmides contenant deux ou trois unités discrètes d’expression, un ou deux qui composent un élément régulateur (plasmide inducteur) qui lie un inducteur de petite molécule d’acquérir l’activité transcriptionnelle ; et un autre plasmide (plasmide effecteur) contenant le transgène sous le contrôle d’une région régulatrice de l’ADN qui permet l’accès de l’élément réglementaire lié aux inducteur. Le principal inconvénient du système binaire est qu’il exige l’introduction concomitante de deux vecteurs (inducteur et effectrices) dans la cellule cible. Dans les expériences d’expression transitoire transgène, la transfection simultanée des deux plasmides produit inévitablement une population de cellules qui est transfectées avec l’inducteur ou l’effecteur isolément ou conjointement transfectées inégalement. En outre, le système binaire nécessite au moins deux tours de sélection antibiotique d’établir des lignées cellulaires stables, qui est longue et laborieuse. Ainsi, combinant tous les composants requis pour l’expression du transgène et de réglementation dans un unique vecteur serait idéale pour garantir l’expression simultanée de tous les éléments requis dans une seule cellule et permettre la régulation de l’expression du transgène Grâce à un traitement avec des inducteurs de petites molécules.
Pour remédier aux insuffisances des commutateurs de gène binaire, nous avons développé récemment un commutateur de gène du singulier, utilisant un Drosophila melanogaster axée sur les EcR gène inductible système15. L’ecdysone intervient dans l’expression des gènes, lorsqu’il se lie à l’hétérodimère EcR/ultraspiracle (USP), qui induit à son tour la liaison du recueil d’ADN des éléments régulateurs3,11. Le récepteur de vertébrés retinoid X (RXR), un orthologues d’insecte USP, recrute des EcR pour former un actif activateur de la transcription16. Ainsi, pour l’expression ciblée d’un transgène dans les cellules de vertébrés ou d’un tissu, le recueil et le RXR doivent être conjointement exprimé simultanément avant étant stimulée par l’ecdysone ou ses agonistes. Étant donné que la fonctionnalité d’hétérodimères du commutateur génique à base EcR peut être influencée par paquet niveau, RXR endogène et al. remplacé les domaines de liaison à l’ADN de l’EcR et l’activation avec le liant à l’ADN de GAL4 hétérologue, herpès simplex virus protéine vmw65 domaines d’activation (VP16) fondu vers le domaine de liaison du ligand EcR, qui ensuite devenue insensible aux RXR endogène et forme un homodimère pour obtenir de l’activité transcriptionnelle17. Le commutateur de gène axée sur les EcR a été encore amélioré par la construction d’une protéine chimérique composée d’un minimum domaine d’activation VP16 combiné avec GvEcR, qui forme un homodimère et localisée dans le cytosol en l’absence de l’agoniste de l’ecdysone, Teb16, 18. En se liant à Teb, le GvEcR a changé sa localisation subcellulaire du cytosol vers le noyau de reconnaître un promoteur hybride composé du poisson-zèbre E1b promoteur minimal combiné avec un tandem dix répété séquences d’activation en amont (HES) pour initier la transcription du gène cible16.
Pour simplifier le gène axée sur les EcR rapporté antérieurement commutateurs16,18, nous avons combiné les dispositifs binaires du système dans un vecteur unique équipé de tous les éléments requis pour stimuler l’expression transgénique et ensuite désignée le vecteur généré récemment, pEUI(+) (nombre de GenBank accession : KP123436, la Figure 1 a)15. Le processeur d’effets en réponse à la GvEcR (ci-après pilote) se compose de dix tandem SAMU répète à côté d’un promoteur minimal E1b suivi d’un site multiple de clonage (MCS) avec des séquences de reconnaissance EcoRI PmlI, NheI, société, AfeI et AflII (Figure 1 b). En outre, pour faciliter la sélection des transfecté des cellules, une puromycine de pilotée par le promoteur SV40 N-acétyl-transférase (PAC) a été placée entre le pilote et effecteur de maintenir les lignées cellulaires stables (Figure 1).
Les auteurs décrivent un protocole détaillé pour la modulation transitoire et stable de transgene expression à l’aide de pEUI(+). En outre, nous fournissons des instructions détaillées pour l’utilisation réussie de Teb comme un agoniste de l’ecdysone.

an ecdysone receptor-based singular gene switch for deliberate expression of transgene with robustness, reversibility, and negligible leakiness

Un contrôle précis de l’expression du transgène est souhaitable dans les études cliniques et biologiques. Cependant, parce que la fonction binaire d’interrupteurs de gène actuellement employé exige le transfert de deux unités d’expression thérapeutique simultanément en une seule cellule, l’application pratique du système pour la thérapie génique est limitée. Pour simplifier le système d’expression de transgènes, nous avons généré un commutateur de gène dénommé pEUI(+) qui englobe un ensemble complet de modules d’expression du transgène dans un vecteur unique. Comprenant le domaine de liaison à l’ADN de GAL4 et mis à jour le recueil (GvEcR), un domaine d’activation minimal VP16 fusionné avec un domaine de liaison à l’ADN de GAL4, mais aussi un mis à jour le récepteur de l’ecdysone Drosophila (EcR), le commutateur de gène du singulier nouvellement mis au point est très sensible à l’administration d’un inducteur chimique d’une manière dépendante du temps et dosage. Le vecteur pEUI(+) est un outil potentiellement puissant pour améliorer le contrôle de l’expression du transgène en recherche en biologie et en études précliniques. Nous présentons ici un protocole détaillé pour la modulation d’une expression du transgène transitoire et stable en utilisant le vecteur pEUI(+) par le traitement de tébufénozide (Teb). En outre, nous partageons des orientations importantes pour l’utilisation de Teb comme un inducteur chimique.

Le commutateur axée sur les GvEcR du singulier gène est représenté dans la Figure 1 a. Le vecteur de pEUI(+) a été optimisé pour l’expression de transgènes réglementés par le traitement avec Teb.
