A principal desvantagem do uso de um interruptor de expressão do gene binário é a necessidade de entregar simultaneamente dois plasmídeos separados (motorista e efetoras) para a célula de destino. Isso pode levar a uma distribuição desigual de plasmídeos e resultam em uma resposta inconsistente do interruptor para o indutores1,2,3. Outra deficiência do sistema de dois vetores é que um mínimo de duas rodadas de seleç?...

Vários genes diferentes switches tem sido explorados por sua capacidade de regular precisamente a expressão do transgene em cultura de células e modelos animais, com diferentes graus de sucesso1,2,3. Sistemas de interruptor de gene potencial devem conhecer vários critérios rigorosos, incluindo: precisão direcional expressão do transgene, capacidade de resposta de dosagem e tempo-dependente para indutores, leakiness insignificante do promotor, reversibilidade do transgene ativação através da remoção de indutor e níveis de toxicidade do indutor toleráveis para linhas celulares e animais de laboratório a1,2,3. Indutores de várias pequenas moléculas têm sido explorados, incluindo tetraciclina4,5, rapamicina6,7,8, mifepristone9,10, e isoinokosterone11,12,13,14. Em geral, estes sistemas exigem pelo menos dois plasmídeos contendo duas ou três unidades discretas de expressão, um ou dois dos quais compõem um elemento regulatório (plasmídeo indutor) que vincula um indutor de pequenas moléculas para obter atividade transcricional; e outro plasmídeo (plasmídeo efetoras) contendo o transgene sob o controle de uma região de DNA reguladora que permite o acesso do elemento indutor-limite regulamentar. A principal desvantagem do sistema binário é que ele requer a introdução concomitante dos dois vetores (indutor e efetoras) na célula alvo. Em experimentos de expressão do transgene transitória, o transfection simultâneo de dois plasmídeos inevitavelmente produz uma população de células que isoladamente é transfectadas com o indutor ou o efetor, ou co transfectadas desigualmente. Além disso, o sistema binário requer pelo menos duas rodadas de seleção antibiótica para estabelecer linhas de célula estável, que é demorado e trabalhoso. Assim, combinar todos os componentes necessários para a expressão do transgene e regulamento em um único vetor seria o ideal para garantir a expressão concomitante de todos os elementos necessários em uma única célula e permitir a regulação da expressão do transgene através do tratamento com indutores de pequenas moléculas.
Para superar as deficiências do gene binário switches, recentemente desenvolvemos um interruptor de gene singular, usando uma Drosophila melanogaster gene Col-baseado sistema inducible15. Isoinokosterone Medeia a expressão genética quando se liga ao heterodímero Col/ultraspiracle (USP), que por sua vez induz a vinculação da colectânea de DNA dos elementos reguladores3,11. O receptor de vertebrados retinoide X (RXR), um ortholog de insecto USP, recrutas EcR para formar um ativador transcricional ativo16. Assim, a expressão orientada de um transgene em células de vertebrados ou tecido, o EcR e RXR devem ser co expressa simultaneamente antes de ser estimulado por isoinokosterone ou seus agonistas. Desde que o recurso heterodimérica dSwitch gene Col-baseado pode ser influenciado pela endógena RXR nível, Padidam et al. substituídos os domínios DNA EcR-vinculação e ativação com ligação de GAL4 DNA heteróloga, herpes simplex vírus proteína vmw65 domínios de ativação (VP16) fundiram-se ao domínio de ligação ligante EcR, que depois tornou-se insensível ao RXR endógena e formou um homodímero para ganhar a atividade transcricional17. O interruptor de gene Col-baseado foi melhorado através da construção de uma proteína quimérico composta por domínio de ativação VP16 mínimo combinado com GvEcR, que formou um homodímero e localizadas no citosol na ausência do agonista isoinokosterone, Teb16, 18. Por ligação a Teb, o GvEcR mudou sua localização subcellular do citosol para o núcleo de reconhecer um promotor híbrido composto de zebrafish E1b promotor mínimo combinado com um conjunto de dez repetidas sequências de ativação upstream (UASs) para iniciar o transcrição do gene alvo16.
Para simplificar o gene Col-baseado anteriormente relatado muda16,18, nós combinamos os recursos binários do sistema em um único vetor equipado com todos os elementos necessários para estimular expressão do transgene e então designado o vetor recém-gerado, pEUI(+) (número de adesão GenBank: KP123436, figura 1A)15. O efetor respondendo para o driver de GvEcR (doravante driver) consiste em dez em tandem UAS repete ao lado de um promotor de E1b mínimo seguido de um local de clonagem múltipla (MCS) com EcoRI, PmlI, NheI, BmtI, AfeI e AflII sequências de reconhecimento (figura 1B). Além disso facilitar a seleção de transfectadas cells, uma puromicina orientada por promotor de SV40 N-acetil-gene transferase (PAC) foi colocada entre as regiões de driver e efetoras para manter a linha celular estável (Figura 1).
Descreveremos um protocolo detalhado para a modulação transitória e estável da expressão do transgene usando pEUI(+). Além disso, nós fornecemos instruções detalhadas para o uso bem sucedido de Teb como um agonista isoinokosterone.

Controle preciso da expressão do transgene é desejável em estudos clínicos e biológicos. O vetor de pEUI(+) é uma ferramenta potencialmente poderosa para melhorar o controle da expressão do transgene em pesquisas biológicas e estudos pré-clínicos. Aqui, apresentamos um protocolo detalhado para a modulação de uma expressão do transgene transitória e estável usando vetor de pEUI(+) pelo tratamento de tebufenozide (Teb).
