Den största nackdelen med binära gen uttryck växel är behovet att samtidigt leverera två separata plasmider (föraren och effektor) i målcellen. Flera olika gen växlar har undersökts för sin förmåga att just reglera transgenens uttryck i cellkultur och djurmodeller, med varierande framgång1,2,3. Potentiella gen switch system bör uppfylla flera stränga kriterier, inklusive: exakt riktad uttryck av transgenens, dos - och tidsberoende lyhördhet för inducerare, försumbar leakiness av arrangören, reversibilitet av transgen aktivering genom inducerare borttagning, och inducerare toxicitet nivåer tolerabla cellinjer och laboratorium djur1,2,3. Flera små molekyler inducerare har utnyttjats, inklusive tetracyklin4,5, rapamycin6,7,8, mifepriston9,10, och ecdysone11,12,13,14. I allmänhet dessa system kräver minst två plasmider som innehåller två eller tre enheter som diskret uttryck, en eller två som komponera ett föreskrivande element (inducerare plasmid) som binder en liten molekyl inducerare att få transkriptionell aktivitet; och en annan plasmid (effektor plasmid) som innehåller transgenens under kontroll av en regulatoriska DNA-region som tillåter åtkomst av elementet inducerare-bunden tillsyn. Den största nackdelen med det binära systemet är att det kräver samtidig införandet av två vektorer (inducerare och effektor) i målcellen. Alltså att kombinera alla komponenter krävs för transgenens uttryck och förordning i en enda vektor skulle vara perfekt att garantera samtidig uttryck för alla element som krävs i en enda cell och möjliggöra reglering av transgenens uttryck genom behandling med liten molekyl inducerare.
För att övervinna bristerna i binärt gen växlar, utvecklade vi nyligen en singular gen switch, med en Drosophila melanogaster REG-baserade gen inducerbara system15. Ecdysone förmedlar genuttryck när det binder till den REG/ultraspiracle (USP) heterodimer, som i sin tur inducerar bindande REG till DNA reglerande element3,11. Ryggradsdjur retinoid X receptorn (RXR), en ortholog av insekt USP, rekryterar REG för att bilda en aktiv transkriptionell aktivator16. Således av riktade uttryck för en transgen i ryggradsdjur celler eller vävnad, REG och RXR skall vara co uttryckt samtidigt innan stimuleras av ecdysone eller dess agonister. Eftersom funktionen heterodimeriskt av växeln REG-baserade genen kan påverkas av den endogena RXR nivå, Padidam et al. ersatte de REG DNA-bindning och aktivering domänerna med heterologt GAL4 DNA-bindande, herpes simplex virus protein vmw65 (VP16) aktiveringen domäner smält till REG ligand-bindande domänen, som sedan blev okänslig för endogena RXR och bildade en homodimer att få transkriptionell aktivitet17. VÃ¤xeln REG-baserade gen förbättrades ytterligare genom att konstruera en chimär protein består av minimal VP16 aktiveringen domän kombinerat med GvEcR, som bildade en homodimer och lokaliserade i cytosolen i avsaknad av den ecdysone agonist, Teb16, 18. Genom bindning till Teb, GvEcR ändrade dess subcellulär lokalisering från cytosolen till kärnan att känna igen en hybrid promotor består av zebrafisk E1b minimal arrangören kombinerat med en tio tandem upprepade uppströms aktiveringen sekvenser (högskolor) att inleda det transkription av genen mål16.
För att förenkla den tidigare rapporterade REG-baserade genen växlar16,18, vi kombinerat binära funktioner i systemet i en enda vektor utrustad med alla element som krävs för stimulerande transgenens uttryck, och sedan designerat den nyligen genererade vektorn, pEUI(+) (GenBank anslutningen nummer: KP123436, figur 1A)15. De effektor som svarar på den GvEcR drivrutinen (nedan) består av tio tandem UAS upprepar bredvid en E1b minimal arrangören följt av en flera kloning site (MCS) med mina, PmlI, NheI, BmtI, AfeI och AflII erkännande sekvenser (figur 1B). Dessutom att underlätta urvalet av transfekterade celler, en SV40 arrangören-driven puromycin N-acetyl-transferas (PAC) genen placerades mellan regionerna driver och effektor att upprätthålla stabil cellinjer (figur 1).

Exakt kontroll av transgenens uttryck är önskvärt i biologiska och kliniska studier. Eftersom funktionen binära av för närvarande anställd gen växlar kräver överföring av två terapeutiska uttryck enheter samtidigt till en enda cell, begränsas emellertid den praktiska tillämpningen av systemet för genterapi. För att förenkla systemet transgenens uttryck, genererade vi en gen switch betecknas som pEUI(+) omfattar en komplett uppsättning av transgenens uttryck moduler i en enda vektor. Bestående av GAL4 DNA-bindande domänen och modifierade REG (GvEcR), en minimal VP16 aktiveringen domän smält med en GAL4 DNA-bindande domän, samt en modifierad Drosophila ecdysone receptor (EcR), växeln nyutvecklade singular genen är mycket lyhörd för administrationen av en kemisk inducerare på ett sätt som tid - och dos-beroende. PEUI(+) vektorn är ett potentiellt kraftfulla verktyg för att förbättra kontrollen av transgenens uttryck i både biologisk forskning och kliniska studier. Här presenterar vi ett detaljerat protokoll för modulering av ett övergående och stabil transgenens uttryck med pEUI(+) vector genom behandling av tebufenozide (Teb). Dessutom delar vi viktiga riktlinjer för användningen av Teb som en kemisk inducerare.

GvEcR-baserade singular gen växeln avbildas i figur 1A. PEUI(+) vector var optimerad för reglerade transgenens uttryck genom behandling med Teb.
