Hoewel microinjection van WCR met dsRNAs met het oog op RNAi gerapporteerde9, dit is het eerste protocol bij het vaststellen van beste praktijken voor het microinjecting van de precellular WCR embryo's, een kritische proces voor het uitvoeren van germline transformatie en/of CRISPR/Cas9 genoom bewerken in deze soort. Succesvolle microinjection van WCR embryo's is afhankelijk van vele factoren, zoals transformatie efficiëntie. Hieronder besproken zijn enkele van de belangrijke kwesties invloed op ...

Westerse maïswortelboorder (WCR), Diabrotica virgifera virgifera, is een belangrijke plaag van maïs1. Interessant, WCR lijkt te overwinnen van de controle maatregelen sneller dan meest agrarische plagen omdat ze niet alleen fysiologisch maar ook gedragsgestoorde2,3,4,5, passen 6. het afgelopen decennium, RNA-interferentie (RNAi), een krachtige functioneel genomisch tool, heeft onderzocht als een potentiële beheermethode voor WCR7,8, en is ook gebruikt als een middel om te studeren gene functie9. Terwijl RNAi wordt vaak uitgevoerd door microinjection van double-stranded RNA (dsRNA) bij andere diersoorten, is injectie gebaseerde RNAi echter zeldzaam in WCR. In feite zijn er slechts enkele rapporten van RNAi via microinjection van dsRNA in WCR9. De reden is dat WCR hoge niveaus van gene vechtpartij via ingestie van dsRNA7,10 bereiken kan, zelfs het toelaat van de studie van embryonale effecten door dsRNA vervoederen aan de moeder11. Terwijl deze methode WCR een uitstekend onderwerp voor functionele genoom analyse via RNAi maakt, is het vooruitgang op de ontwikkeling van methodologieën voor embryonale microinjection in deze soort vertraagd.
Ondanks de kracht van RNAi zijn er een paar nadelen. Niet alle genen reageren bijvoorbeeld even op RNAi. Deze variabiliteit kunnen interpreteren van de resultaten van een functionele genomica assay moeilijker. Ook RNAi voorbijgaande aard is en genereert geen erfelijke mutaties. Aan de andere kant, kan germline transformatie, een ander functioneel genomisch hulpmiddel, erfelijke mutaties via het invoegen van een gemarkeerde transposons element in het genoom12,13genereren. Dit maakt germline transformatie een uitstekend hulpmiddel voor het maken van gemuteerde stammen voor gebruik in genetische langetermijnstudies. Transformatie kan ook worden gebruikt om te redden van een mutatie door het leveren van een functionele kopie van het gen tot een mutant genoom14. Bovendien is germline transformatie de hoeksteen van een grote verscheidenheid van moleculaire genetische technieken. Naast uitsparen en/of redden van de genfunctie, kan germline transformatie worden gebruikt voor enhancer overlapping15, gene overlapping16, ectopische expressie Gal4 gebaseerde17en genoom-brede mutagenese18.
Meer recentelijk, gereedschappen waarmee geavanceerde genoom bewerken zijn ontwikkeld. Deze tools omvatten transcriptie Activator-Like Effector nucleasen (TALEN) en de CRISPR/Cas9-nuclease systeem19,20. Het voordeel van deze nieuwe instrumenten is dat ze onderzoekers de mogelijkheid bieden voor het opwekken van een double-stranded DNA pauze op bijna elke plek binnen bijna elke genoom. Zodra geïnduceerde, deze onderbrekingen kunnen worden gerepareerd via niet-homologe weerszijden toe te treden, die kan introduceren verwijderingen of inserties in het beoogde gen, of via homologie geleide reparatie, die in het bijzijn van een gemodificeerde constructie, kan katalyseren de vervanging van de gehele genen of de genetische gebieden21,22. Echter, deze methoden vereisen microinjection van DNA, RNA en/of eiwit in zeer jonge embryo's.
Nog belangrijker is, in tegenstelling tot de dsRNAs gebruikt voor RNAi, nucleïnezuren en proteïnen gebruikt voor germline kruisen transformatie en genoom bewerken niet gemakkelijk celmembranen. Daarom, microinjection van de ANI van de plasmide, mRNAs, en/of eiwitten moet plaatsvinden tijdens de syncytieel blastoderm fase (dat wil zeggen voordat het insect embryo cellularizes). Dit maakt de timing van injecties een kritieke factor. Bijvoorbeeld bij Drosophila melanogastermoeten embryo's binnen twee uur na ei12 leggenworden geïnjecteerd. Daarom moet de ontwikkeling van een succesvolle embryonale microinjection protocol voor WCR rekening houden met de beste voorwaarden voor vrouwelijke leggen van eieren, evenals de beste methode voor het verzamelen van voldoende hoeveelheden van precellular embryo's.
Een poging om een breed scala aan moleculaire genetische hulpmiddelen te oefenen op WCR biologie vereist de ontwikkeling van methoden voor het verzamelen en microinjecting van de precellular WCR embryo's. Hier voorzien wij gedetailleerde instructies, samen met tips en trucs, om anderen transformatie gebaseerde technieken gebruiken in WCR onderzoek te helpen. De westerse maïswortelboorder (WCR) is een belangrijke plaag van maïs en staat bekend om zijn vermogen om snel aanpassen aan de pest beheersstrategieën. RNA-interferentie (RNAi) is een krachtig hulpmiddel voor de studie van biologie van de WCR gebleken, heeft maar zijn beperkingen. Specifiek, RNAi zelf voorbijgaande aard is (d.w.z. niet leidt tot een langdurige Mendelian inheritance van het bijbehorende fenotype), en het vereist de opeenvolging van DNA van het doel-gen weten. De laatste kan worden beperkt als het fenotype van belang wordt gecontroleerd door slecht bewaard gebleven, of zelfs nieuwe genen, omdat identificeren nuttige doelen zou worden uitdagend, zo niet onmogelijk. Dus, het aantal gereedschappen in de WCR genomic gereedschapset moet worden uitgebreid door de ontwikkeling van methoden die gebruikt kunnen worden maken van stabiele gemuteerde stammen en schakelen sequence-onafhankelijke onderzoeken van de WCR-genoom. Een belangrijke overweging bij het ontwikkelen van dit protocol werd het stadium van de embryonale ontwikkeling ten tijde van microinjection. In het bijzonder vereist germline omzetting microinjection van ANI voorafgaand aan embryonale cellularization. Dit is omdat het DNAS-systeem niet gemakkelijk kruisen celmembranen.
