Toeprinting est une technique puissante pour mesurer directement la capacité d’un ARN d’intérêt pour soutenir la formation de translation initiation complexes dans une situation très contrôlée. Ce protocole décrit une technique simplifiée pour toeprinting ARN chez les mammifères. Comme traduction consomme de l’énergie plus cellulaire, il est logique que la traduction est étroitement réglé1. À l’inverse, le dérèglement de la traduction- et les altérations qui en résulte dans la production de protéine-est souvent observée aux réponse au stress et la maladie, comme le cancer1,2. Un avantage majeur de contrôle de la traduction est la vitesse avec laquelle les cellules peuvent modifier leur production de protéines afin de répondre aux divers stimuli3. Règlement de traduction représente donc un mécanisme important qui peut influer sur la survie des cellules et la mort1,2,3. Les étapes de la traduction, l’initiation est la plupart très réglementé et complexe3. Brièvement, l’ARNm eucaryotes plus contiennent une 5' m7G PAC est presque toujours indispensable pour leur traduction. Initiation PAC-dépendant nécessite initiation eucaryote facteurs eIF4E, eIF4G et eIF4A (le complexe de cap-reconnaissance) pour interagir avec l’extrémité 5' de l’ARNm. 43 s pré-initiation ribosome complex, qui contient eIF2 lié aux initiateur tRNA et eIF3, est recruté à l’extrémité 5' de l’ARNm via une interaction entre eIF4G et eIF3. La complexe de pré-initiation est pensée pour numériser des ARNm, aidé par eIF4A (une hélicase de RNA) jusqu'à ce que le codon de début (AUG) se trouve. La complexe d’initiation de 48 s est formée par la suite et l’ARNt est livré dans le site P du ribosome. Enfin, les sous-unités de ribosomes 60 s et 40 s sont Unies pour former la complexe, d’initiation 80 s suivi de translation élongation1,3,4. En revanche, les sites d’entrée des ribosomes interne (IRESes) contournement l’exigence pour un plafond de 5' en recrutant la sous-unité ribosomique 40 s directement à l’initiation codon3. Conditions de stress physiologique atténuent traduction ARNm globale en raison de modifications des facteurs d’initiation eucaryote générales clés (SIFE). Cependant, mécanismes d’initiation de traduction non canoniques permettant la traduction sélective de certains ARNm qui souvent codent pour des protéines de la réponse au stress, et le dérèglement de l’initiation de la traduction non canonique est impliqué dans des États de maladie comme le cancer 1 , 2. éléments régulateurs RNA discrets, tels que IRESes cellulaires, permettant la traduction de ces ARNm2,3.
Un aspect particulièrement intéressant de contrôle de la traduction est de comprendre les mécanismes de canonique versus non canonique traduction d’un ARNm donnée. Toeprinting est une technique qui permet l’étude mécanistique détaillée d’initiation de la traduction de l’ARN spécifique in vitro. L’objectif global du toeprinting consiste à évaluer la capacité d’un ARN d’intérêt de nucléée la formation d’une complexe avec le ribosome dans une variété de conditions, d’initiation de traduction afin de déterminer quelles séquences, éléments structuraux ou accessoire facteurs sont requis pour l’initiation de la traduction efficace. Par exemple, recrutement du ribosome pourrait être entravé en l’absence d’un plafond de 5' mais stimulé par la présence d’une IRES.
Le principe de la technique est à in vitro transcrire un ARN d’intérêt, il incuber en présence des extraits cellulaires contenant des composants de translation (ou les composantes purifiées) permettant de transcrivent les complexes de l’initiation à la forme et d’inverser l’ARN avec une amorce spécifique. Des complexes stables de RNA-ribosome provoquera la transcription inverse à caler au bord 3' du ribosome, le soi-disant « toeprint »5,6,7.
Dans ce protocole, les sous-unités ribosomiques, SIFE, ARNt et IRES trans-des facteurs (ITAFs) sont idéalement fournis par lysate de reticulocyte de lapin (RRL). Un autre avantage de ce protocole est l’utilisation d’une amorce marquée fluorescent et imageur de gel à base de fluorescence, plutôt qu’un apprêt radiomarqué. Cela permet d’éliminer des étapes supplémentaires, y compris le radiomarquage l’amorce, ainsi que le gel de séchage et exposer à un écran renforçateur. Les bandes fluorescentes sont enregistrés en temps réel, comme les essais de gel, ce qui permet une plus grande résolution. Non plafonné liée à le X inhibiteur de l’apoptose protéine (XIAP) IRES ARN est utilisé comme un exemple ici, bien que plafonnés ARNm peut également être analysés par cette technique8.
Contrairement à l’analyse de l’Ouest, qui mesure la production finale de la traduction dans les lysats cellulaires, toeprinting est une approche in vitro pour mesurer la formation du complexe initiation traduction de l’ARN. Cette approche réductrice permet l’étude très détaillée des substrats ou facteurs qui régulent l’initiation de la traduction (e.g., plafonnés ou non plafonnés ARNm, IRES structure, présence ou absence de queue polyA, disposition des facteurs protéiques spécifiques, " etc..). Par conséquent, toeprinting peut être utilisé pour étudier les différents modes de traduction8 ou les effets de structures de l’ARNm, comme IRESes, sur la synthèse de protéines9,10.

Initiation de la traduction est l’étape limitante de la synthèse protéique et représente un point essentiel au cours de laquelle les cellules régulent leur production de protéines. Régulation de la synthèse de la protéine est la clé de la réponse cellulaire au stress, et le dérèglement est au cœur de nombreux états pathologiques, comme le cancer. Par exemple, bien que le stress cellulaire entraîne l’inhibition de la traduction globale par atténuation dépendante de PAC initiation, certaines protéines de la réponse au stress sont sélectivement traduits d’une manière indépendante du cap. Discrets éléments régulateurs de RNA, tels que les sites d’entrée cellulaire ribosome interne (IRESes), permettant la traduction de ces ARNm spécifiques. Identification de ces ARNm et la caractérisation de leurs mécanismes de régulation, ont été un domaine essentiel en biologie moléculaire. Toeprinting est une méthode pour l’étude de structure d’ARN et de la fonction en ce qui touche à l’initiation de la traduction.
