De studie van de meeste aspecten van zoogdieren orgel ontwikkeling is bevorderd door in vitro -systemen. Twee belangrijkste methoden worden nu gebruikt voor de cultuur van vestibulaire zintuigen: vrij zwevende1 en aanhangend2 preparaten. Beide methoden toestaan dat het onderzoek van Haarcel kwetsbaarheden3 en regeneratie1,4 in vitro. Bovendien hebben de ontwikkelingstoxiciteit rollen van de inkeping5,6, Wnt7,8en epidermale groeifactor receptor (EGFR)9,10 signalering cascades in het binnenoor vastgesteld, gedeeltelijk door het gebruik van in vitro culturen van sensorische epitheel. Echter celgroei en differentiatie worden beheerd, niet alleen door middel van de signalering door morphogens, maar ook door middel van fysieke en mechanische signalen zoals intercellulaire contacten, de stijfheid van de extracellulaire matrix, en mechanische uitrekken of vernauwing. De rol van dergelijke mechanische stimuli is uitdagend om te onderzoeken in de ontwikkelingslanden binnenoor in vivo. Bovendien, bestaande vrij zwevende en aanhangend cultuur methoden zijn niet geschikt voor dergelijk onderzoek in vitro. Hier beschrijven we een methode voor driedimensionale organotypic cultuur in collageen gels ik verschillende stijfheid. Deze methode grotendeels bewaart de in vivo -architectuur van de vestibulaire en cochleair zintuigen en laat onderzoek naar de gevolgen van mechanische kracht voor de groei en differentiatie11.
Omdat de mechanische prikkels zijn bekend om te activeren downstream moleculaire gebeurtenissen, zoals de Hippo signalering traject12,13,14,15, is het belangrijk om het combineren van mechanische stimulatie te kunnen met biochemische en genetische manipulaties. De hier beschreven methode van cultuur virus-gemedieerde gene levering toestaat en kan daarom gebruikt worden om te studeren zowel mechanische als moleculaire signalering tijdens binnenoor ontwikkeling11.

three-dimensional organotypic cultures of vestibular and auditory sensory organs

De zintuigen van het binnenoor zijn uitdagend om te studeren bij zoogdieren als gevolg van hun onbereikbaarheid experimentele manipulatie en optische observatie. Bovendien, hoewel bestaande cultuur technieken toestaan biochemische verstoringen, deze methoden bieden niet een middel tot het bestuderen van de effecten van mechanische kracht en weefsel stijfheid tijdens de ontwikkeling van de zintuigen van het binnenoor. Hier beschrijven we een methode voor driedimensionale organotypic cultuur van de intact lymfkliertest utricle en slakkenhuis overwint deze beperkingen. De techniek voor aanpassing van de stijfheid van een drie-dimensionale matrix hier beschreven staat manipulatie van de elastische kracht tegen weefsel groei. Deze methode kan daarom worden gebruikt om te studeren van de rol van mechanische krachten tijdens de ontwikkeling van het binnenoor. Bovendien, toestaan de culturen virus-gemedieerde gene levering, die kan worden gebruikt voor experimenten van de winst - en verlies-van-functie. Deze methode van cultuur behoudt aangeboren haarcellen en ondersteunende cellen en fungeert als een potentieel superieur alternatief voor de traditionele tweedimensionale cultuur van vestibulaire en auditieve zintuigen.
