HTRF insulin analysen beskrives her tilbyder en hurtig og effektivt system til at måle insulinsekretion fra en kulturperler celle-baseret system. Reguleringen af energimetabolisme er finjusteret af en større anabole hormon, insulin. Insulin er syntetiseret og udgivet af pancreas beta celler som reaktion på øget ekstracellulær glucose niveauer. Den frigivne insulin udløser optagelsen af glukose af insulin-følsomme væv1,2. Fysiologisk, er dette knyttet til udvidelse af glukose koncentration efter et måltid, efterfulgt af udskillelsen af insulin til at regulere glukoseoptagelse. Forstyrrelser i glukose homøostase fører til metabolisk funktionshæmninger kulminerede i insulinresistens og i sidste ende i debut af type 2 diabetes2,3,4.
Selv om insulinsekretion er blevet grundigt undersøgt, fortsat dets reguleringsmekanismer dårligt forstået. Et kritisk område af undersøgelsen har været identifikation af roman modulatorer af insulinsekretion betaceller5,6,7,8. Disse undersøgelser kræver en bedre forståelse af kobling forholdet mellem glucose stimulation og insulinsekretion. Evne til præcist at overvåge og kvantificere niveauerne af glucose-stimuleret insulinsekretion (GSIS) har derfor været afgørende. Til dato, dog var kun et begrænset antal metoder til rådighed til at tillade kvantificering af GSIS ved hjælp af insulin-secernerende cellelinjer og/eller bugspytkirtlen Holme. En er radioimmunoassay (RIA), som udnytter radioisotop-tagged insulin og antistoffer. De vigtigste begrænsninger af denne tilgang omfatter sikkerhedsspørgsmål som følge af håndtering og bortskaffelse af radioaktivt materiale. AlphaScreen teknologi er baseret på konvertering af luftens ilt ind i en spændt ilt singlet tilstand, som kan reagere med kemiluminescens arter, resulterer i generation af chemiluminescence. Fordi analysen er homogen, er mange af de vask skridt tilknyttet RIA og ELISA elimineret. Men ustabilitet af signalet på grund af karakteren af reaktionen er en begrænsende faktor, der kan påvirke udlæsning af analysen. (TR-KNIBTANG, udviklet af Heyduk og kolleger9, er en anden homogen tilgang til insulin måling baseret på bindingen af to separate antistoffer til forskellige epitoper på insulin molekyle. Antistofferne er hver kemisk forbundet med dobbelt strandede DNA med korte supplerende enkelt strandede DNA markiser. Binding af antistoffer mod insulin samler dem og fører til en dobbelt strandede DNA duplex. Hver antistof er også forbundet med en respektive donor eller acceptor fluorophore, og sammenslutningen af DNA duplex samler disse fluorophores til at generere Förster resonans energioverførsel (FRET). En potentiel begrænsning af TR-KNIBTANG, hviler dog med ÆRGRE sig selv. Manglende evne til hurtigt sprede baggrund fluorescens under FRET reaktion kan føre til relativt høje niveauer af baggrunden fluorescens og et lavt signal til støjforhold i analysen. Derfor findes der stadig et behov for en pålidelig, robust og omkostningseffektiv analyse til kvantificering af GSIS i en høj overførselshastighed måde.
De seneste fremskridt i biofysik har kulminerede i udviklingen af en homogen tidsopløst fluorescens energi resonans overførsel (HTRF) baseret assay. Specifikt, mens energien overføres inden for kan analysen betegnes som FRET-baseret, mere præcist, HTRF bygger på luminescence energi resonans overførsel (LRET)10 , som er ikke-radiative overførsel af energi mellem donor og acceptor arter11,12,13. Denne sondring er vigtig, da timingen af et fluorescens eller dæmper baseret FRET interaktion er meget anderledes end for LRET, men de samme typer af gating kan også bruges til FRET og LRET. Desuden brug af sjældne jordarters lanthanide cryptate forbindelser såsom europium eller terbium cryptate i HTRF producerer lang fluorescens halveringstider12,14. Dette giver den unikke fordel af indførelsen af en forsinkelse (µsec) mellem donor excitation og måling af emission fra acceptoren (dvs., tidsopløst assay). Denne forsinkelse giver mulighed for tilstrækkelig tid til baggrunden fluorescens at sprede før måling af acceptoren emission fluorescens. Udlæsningen er derfor gratis ikke-specifik fluorescens og således et højt signal / støj-forhold opnås (figur 1). Desuden, den ensartede karakter af HTRF eliminerer behovet for vaske trin til at vaske den ubundne arter, hvilket gør analysen meget hurtigere end ELISA eller RIA-baserede metoder.

Her, vi leverer en detaljeret protokol for at benytte en HTRF-baseret tilgang til bestemmelse af GSIS fra INS-1E celler, en veletableret insulin-secernerende beta celle-afledte rotte insulinom celle linje15. Derudover kan dette assay bruges til at identificere den farmakologiske profil af molekylære regulatorer af insulinsekretion. Vi anvender denne HTRF-baserede insulin analyse for at undersøge dopamin D2-indkvartering receptor regulering af GSIS. homogeneous time-resolved f&#246;rster resonance energy transfer-based assay for detection of insulin secretion

Påvisning af insulinsekretion er kritisk for informative mekanismer til reguleret sekretion såvel som i undersøgelser af metabolisme. Selvom mange insulin assays har eksisteret i årtier, har den nylige fremkomsten af homogene tidsopløst Förster resonans energi Transfer (HTRF) teknologi væsentligt forenklet disse målinger. Dette er en hurtig, omkostningseffektiv, reproducerbare og robust optisk assay afhængig af antistoffer konjugeret med lyse fluorophores med langvarige emission, som letter tidsopløst Förster resonans energi Transfer. Desuden er HTRF insulin opdagelse modtagelig for udviklingen af high throughput screening-assays. Her bruger vi HTRF til at opdage insulinsekretion i INS-1E celler, en rotte insulinom-afledte cellelinie. Dette giver os mulighed at vurdere basal niveauer af insulin og deres ændringer som reaktion på glucose stimulation. Derudover bruger vi denne insulin detection system til at bekræfte betydningen af dopamin som en negativ regulator af glucose-stimuleret insulinsekretion (GSIS). I en lignende måde, andre dopamin D2-ligesom receptor agonister, quinpirole og Bromocriptin, reducere GSIS i en koncentration-afhængige måde. Vores resultater fremhæve nytten af HTRF insulin assay format i bestemmelse af mange stoffer i GSIS og deres farmakologiske profiler betydning.

Vi validerede vores insulin HTRF analyse ved at generere en insulin standardkurve ved hjælp af oprenset humant insulin standarder af foruddefinerede koncentrationer (figur 2).
