Oligodendrocytes (عملية شريان الحياة) هي خلايا ميليناتينج الجهاز العصبي المركزي (CNS)1. بالعزلة وثقافة oligodendrocytes الأولية في بيئة محكم تنظيم أداة قيمة للدراسة في المختبر لتطوير أوليجوديندروجليا، فضلا عن بيولوجيا دملينتينغ الأمراض مثل التصلب المتعدد2 . وهذا يتطلب oligodendrocyte فعالة وقوية العزلة وثقافة أسلوب3. في هذه الدراسة، ونحن استغل للتعبير عن علامة سطح الخلية oligodendrocyte المميزة لتنفيذ تقنية عزل معدلة التي سريعة ومحددة.

rapid and specific immunomagnetic isolation of mouse primary oligodendrocytes

عزلة فعالة وقوية وثقافة oligodendrocytes الأولية (عملية شريان الحياة) أداة قيمة للدراسة في المختبر لتطوير أوليجوديندروجليا، فضلا عن بيولوجيا دملينتينغ الأمراض مثل التصلب المتعدد و بليزايوس-ميرتسباخر-مثل مرض (PMLD). هنا، فإننا نقدم بسيطة وتحقيق الكفاءة في اختيار الأسلوب لعزل إيمونوماجنيتيك المرحلة O4 ثلاث+ الخلايا بريوليجوديندروسيتيس من الجراء الفئران حديثي الولادة. منذ را غير ناضجة تشكل أكثر من 80% من هذه المسألة الأبيض القوارض في الدماغ عند الولادة يوم 7 (P7) لا يضمن هذا الأسلوب عزل عالية الغلة الخلوية، ولكن أيضا عزلة محددة من شريان الحياة للسودان ملتزمة بالفعل النسب أوليجوديندروجليال، تناقص إمكانية عزل الخلايا تلويث مثل أستروسيتيس والخلايا الأخرى من الدماغ الماوس. هذا الأسلوب هو تعديل التقنيات التي ذكرت سابقا، ويوفر oligodendrocyte إعداد نقاوة أعلى من 80% في حوالي 4 ح.
