Verschillende stappen in het protocol kunnen worden onderworpen aan wijzigingen onder bepaalde omstandigheden. De duur van fixatie hangt bijvoorbeeld af van de leeftijd, van de embryo's, variërend van 2 h tot 1 of meer dagen met behulp van vers-en-klare paraformaldehyde. Aangezien de fixatie wordt uitgevoerd vóór de hele berg immunokleuring, op antilichamen die gevoelig voor eiwit crosslinking zijn, kan methanol worden gebruikt als een alternatieve kleefpoeders agent. Spinale MNs zijn het deel van het centrale zenuwstelsel maar innervate perifere spieren om te controleren van de beweging. De ontwikkelende ruggenmerg, zijn volgens de signalen afkomstig van de chorda en de aangrenzende somieten MN progenitoren (pMNs) gevestigd. Alle post mitotische MNs gedifferentieerd zijn vervolgens gegenereerd op basis van pMNs, die uiteindelijk aanleiding geven tot een reeks van MN subtypen langs de as van de rostrocaudal van het ruggenmerg1,2. Spinale MNs zijn topografisch en anatomisch goed georganiseerd. Hun morfologische overeenkomst correleert met de positie van hun respectieve doel in de periferie-3. Bij het bereiken van hun doelen spier, axonen ontvangen neurotrophic cellulaire en exogene factoren die veroorzaken ze uit te breiden en tak verder in spieren. Innervatie arm en vertakkende gebreken kunnen bijdragen aan het uitblijven van neuromusculaire kruispunten (NMJ) vormen. Bijvoorbeeld gliale-afgeleide neurotrophic factoren (GDNF)-geïnduceerde Pea3 is onmisbaar voor axon arborization in cutane maximus (CM) en latissimus dorsi (LD) spieren4,5. Bovendien, tonen DINE knockout muis embryo's defecte arborization van phrenic zenuwen in het middenrif, respiratoir falen en mortaliteit veroorzaakt onmiddellijk na de geboorte6,7. Deze laatste stap van MN rijping (d.w.z., axonale projectie en vertakking) is daarom van cruciaal belang om communicatie tussen de neuronen en doelcellen.
Als u wilt bekijken arborization patronen, gedrag onderzoekers normaal confocal imaging of de lichte microscopie van de twee-foton fluorescentie van verdeelde of geheel-mount monsters8,9,10. Beide microscopie technieken genereren aanvaardbare resolutie en diepte penetratie. De lichte microscopie van twee-foton fluorescentie omvat excitatie van fluorophores door gelijktijdige absorptie van twee lage-energie fotonen11. Aangezien twee-foton excitatie nabij-infrarood straling gebruikt, bijdraagt de verminderde excitatie-frequentie aan verminderde verstrooiing en betere weefsel penetratie tot 1 mm in weefsel, waardoor beeldbewerking met meer diepte. Confocale microscopie verwijdert u door filters de out-of-focus signalen en verzamelt enkel licht binnen het brandvlak12. Met deze aanpak, kunnen beelden van monsters van verschillende focale vlakken worden gecombineerd voor de productie van een driedimensionaal (3D) beeld via de functie van een Z-stack. Niettemin, signaalsterkte wordt verminderd, zoals de meeste van het licht wordt geblokkeerd en hoge numerieke openingen de diepte-van-veld verdoezelen. Nog belangrijker, bijdragen beide technieken tot de ernstige photodamage en fototoxiciteit aangezien het hele model verlichting, krijgt zelfs wanneer slechts één vliegtuig is beeld op een bepaald moment.
Om te omzeilen deze tekortkomingen, LSFM een favoriet alternatief, met de voordelen van het snel en licht-efficiënt, en minder fototoxische13,14geworden. Verder staat LSFM Multi-View imaging. Deze aanpak is bijzonder geschikt voor het visualiseren van motorische axonen en hun verspreiden terminals zoals ze verspreid via 3D-ruimte. LSFM beter presteert dan de andere twee opties omdat monsters zijn gemonteerd op een podium waarmee rotatie rond een verticale as en bewegingen langs de x-, y- en z-assen. Niet alleen kunt deze set-up voor een minimaal geblokkeerde weergave van het monster, maar ook de keuze van een wenselijk verlichting pad, een tekortkoming van de twee-foton en confocal microscopie, die beide montage van monsters op een vlakke dia vereisen. LSFM is daarom het meest geschikte instrument voor 3D beeldvorming van axon arborization en kwantificering van de terminals van de motorische zenuw in muis embryo's.

Spinale motorische neuronen (MNs) breiden hun axonen om te communiceren met hun innervating doelen, waardoor beheersing van beweging en complexe taken in gewervelden. Het is dus van cruciaal belang voor het ontdekken van de moleculaire mechanismen van hoe motor axonen Navigeer naar arborize en hun perifere spier innervate richt zich tijdens het ontwikkelen en degeneratie. Hoewel transgene Hb9::GFP muis lijnen lang diende hebben te visualiseren motor axon trajecten tijdens de embryonale ontwikkeling, gedetailleerde beschrijvingen van het volledige spectrum van axon terminal arborization onvolledig als gevolg van de complexiteit van de patroon blijven en beperkingen van de huidige optische microscopie. Hier beschrijven we een verbeterde protocol dat combineert licht blad fluorescentie microscopie (LSFM) en robuuste beeldanalyse kwalitatief en kwantitatief visualiseren ontwikkelen van motorische axonen. Dit systeem kan gemakkelijk worden vastgesteld om het Kruis genetische mutanten of MN ziekte modellen met Hb9::GFP lijnen, onthullende roman moleculaire mechanismen die tot defecten in motor axon navigatie en arborization leiden.

LSFM biedt gedetailleerde 3D visualisatie van MN baan en axon arborization in muis embryo's. Onder heldere veld verschijnen weefsels volledig transparant na ondergedompeld worden in de commerciële clearing-reagens. Geen krimp of zwelling van het monster werd opgemerkt na maximaal een week van opslag in het reagens voor imaging ontruimen.
