Mehrere Schritte im Protokoll können unter bestimmten Umständen Änderungen unterzogen werden. Zum Beispiel hängt die Dauer der Fixierung auf das Alter des Embryos, variierend von 2 h bis 1 oder mehrere Tage mit frisch zubereiteten Paraformaldehyd. Da Fixierung vor dem ganzen Berg Immunostaining für Antikörper durchgeführt wird, die empfindlich auf Protein Vernetzung kann Methanol als alternative Fixativ Agent verwendet werden. Spinale MNs sind der Teil des zentralen Nervensystems aber innervieren peripheren Muskeln, um die Bewegung zu kontrollieren. Entwickelnden Rückenmarks sind nach den Signalen aus der Chorda und angrenzenden Somiten MN Stammväter (pMNs) gegründet. Alle differenziert nach dem mitotischen MNs entstehen dann von pMNs, was schließlich zu einer Reihe von MN Subtypen der Rostrocaudal Achse des Rückenmarks1,2. Spinale MNs sind topographisch und anatomisch gut organisiert. Ihre morphologische Anordnung korreliert mit der Position von ihrem jeweiligen Ziel in der Peripherie-3. Bei ihren Muskel-Ziele zu erreichen, Axone erhalten zellularen und exogene neurotrophen Faktoren, die bewegen sie zum Erweitern und verzweigen weiter in die Muskeln. Innervation und verzweigte Mängel können zum Versagen, neuromuskuläre Kreuzungen (NMJ) bilden beitragen. Z. B. Glia-derived neurotrophen Faktoren (GDNF)-induzierte Pea3 ist unverzichtbar für Axon Arborization in kutane Maximus (CM) und Latissimus Dorsi (LD) Muskeln4,5. Darüber hinaus zeigen DINE -Knockout-Maus-Embryonen defekt Arborization phrenicus Nerven des Zwerchfells, wodurch respiratorische Insuffizienz und Sterblichkeit unmittelbar nach Geburt6,7. Daher ist dieser letzte Schritt der MN Reifung (d.h., axonale Projektion und Verzweigung) entscheidend für die Kommunikation zwischen Neuronen und Zielzellen zu gewährleisten.
Um Arborization Muster zu sehen, führen die Forscher normalerweise konfokale Bildgebung oder zwei-Photonen-Fluoreszenz Lichtmikroskopie geschnitten oder ganz-Mount Proben8,9,10. Beide dieser Mikroskopie-Techniken erzeugen akzeptable Auflösung und Tiefe Penetration. Zwei-Photonen-Fluoreszenz Lichtmikroskopie beinhaltet Erregung des Fluorophore durch gleichzeitige Aufnahme von zwei energieärmeren Photonen11. Da zwei-Photon Erregung Nah-Infrarot-Strahlung verwendet, trägt die verminderte Erregerfrequenz reduzierte Streuung und bessere Gewebe eindringen bis zu 1 mm im Gewebe, wodurch die Bildgebung mit größerer Tiefe. Konfokalen Mikroskopie wird durch Filter, die der Out-of-Focus-Signale und sammelt nur Licht in die Brennebene12,entfernt. Mit diesem Ansatz können Bilder von Proben aus verschiedenen fokalen Ebenen kombiniert werden, um ein dreidimensionales (3D) Bild über eine Z-Stapel-Funktion zu produzieren. Dennoch wird Signalintensität reduziert, da die meisten des Lichtes ist blockiert und hohe numerische Öffnungen die Depth of Field verdecken. Noch wichtiger ist, beitragen beide Techniken zur schweren lichtbedingten und Phototoxizität, da die gesamte Probe Beleuchtung erhält, selbst wenn nur in einer Ebene zu einem bestimmten Zeitpunkt abgebildet wird.
Um diese Unzulänglichkeiten zu umgehen, ist LSFM eine bevorzugte Alternative, mit den Vorteilen des Seins schnelle, effiziente Licht- und weniger phototoxische13,14geworden. Darüber hinaus ermöglicht LSFM MV Bildgebung. Dieser Ansatz eignet sich besonders zur motorischen Axone und ihre Dispergier-Terminals zu visualisieren, wie sie durch den 3D Raum zu verbreiten. LSFM übertrifft die anderen beiden Optionen, weil Proben auf einer Bühne, die Drehung um eine vertikale Achse und Bewegungen entlang der x-, y- und Z-Achsen ermöglicht, montiert sind. Dieses Set-up ermöglicht nicht nur minimal blockierte Blick auf die Probe, sondern auch die Wahl eines Pfades wünschenswert Beleuchtung, ein Manko der zwei-Photonen und konfokale Mikroskopie, beide in der Montage von Proben auf einer flachen Folie benötigen. LSFM ist daher das am besten geeignete Werkzeug für die 3D Darstellung von Axon Arborization und zur Quantifizierung der motorische Nerv Terminals in mausembryonen.

Spinalen motorischen Neuronen (MNs) verlängern ihre Axone zur Kommunikation mit ihren innervating Ziele, wodurch die Steuerung Bewegung und komplexe Aufgaben in Wirbeltieren. Daher ist es wichtig, die molekularen Mechanismen der wie Motor Axone zu navigieren, arborize und die peripheren Muskeln innervieren richtet sich während der Entwicklung und Degeneration zu entdecken. Obwohl transgene Mauslinien Hb9::GFP lange gedient haben, motorischen Axon Trajektorien während der Embryonalentwicklung zu visualisieren, detaillierte Beschreibungen der das gesamte Spektrum der Axon terminal Arborization blieben unvollendet aufgrund der Komplexität der Muster und Einschränkungen der aktuellen optische Mikroskopie. Hier beschreiben wir eine verbesserte Protokoll, die leichte Blech-Fluoreszenz-Mikroskopie (LSFM) und robuste Bildanalyse, qualitativ und quantitativ zu visualisieren, Entwicklung von motorischen Axone verbindet. Dieses System kann leicht angenommen werden, um genetische Mutanten oder MN Krankheitsmodelle mit Hb9::GFP Linien, enthüllt neuere Molekulare Mechanismen, die zu defekten motor Axon Navigations-und Arborization führen zu überqueren.

LSFM bietet detaillierte 3D Visualisierung von MN Flugbahn und Axon Arborization Maus Embryonen. Unter Hellfeld erscheinen nach dem Eintauchen in das kommerzielle Clearing-Reagenz Gewebe vollständig transparent. Keine Schrumpfung oder Schwellung der Probe wurde nach der Lagerung bei der Aufklärung von Reagenz vor Bildgebung bis zu einer Woche bemerkt.
