Alle experimentele protocollen die hier werden gebruikt werden goedgekeurd door het Comité voor dierverzorging en gebruik van de Musashino Universiteit. Mannelijke ddY muizen (SLC, Shizuoka, Japan) werden bewaard voor ten minste 7 dagen onder een 12-h licht / donker cyclus voor experimenten met water en voedsel ad libitum. De meest kritische stap in deze experimenten is het succes van de i.c.v. injectie. De i.c.v. injectietechniek die hier wordt gebruikt is vrij eenvoudig maar vereist wat oefening. Voorafgaand aan experimenten wordt de praktijk met kleurstoffen (bijvoorbeeld 0,5% trypanblauw in zoutoplossing) aanbevolen. Dieren krijgen verschillende fysieke en chemische stimuli uit hun omgeving via sensoren op de perifere zenuwen. De voorbijgaande receptor potentieel vanilloid type 1 (TRPV1) is een van de thermogevoelige, niet-reflecterende kanalen die fungeren als warmtesensoren1,2, en activering en / of modulatie van TRPV1 staat bekend als een belangrijke stap voor nociception in zowel normale als inflammatoire contexten3. Hoewel het algemene expressiepatroon controversieel is, is ook de expressie van TRPV1 voorgesteld in suprachirurgische regio's, waarbij het betrokken is bij verschillende hersenactiviteiten (waaronder nociception4, thermoregulatie5, angst6, attention deficit hyperactivity disorder7en epilepsie8). Bovendien is onlangs gesuggereerd dat paracetamol, een veelgebruikte pijnstiller, bemiddelt de activering van centrale TRPV1 om zijn pijnstillende werking9,10uit te lokken .
Toediening van overtollige TRPV1 agonist met inbegrip van capsaïcine en harsiferatoxine (RTX) aan dieren leidt tot de dood van TRPV1-positieve neuronen en langdurige desensibilisatie van TRPV1 agonisten11,12. In combinatie met de lokale toepassing (intrathecal13,14, intracisternal15,16,17, en intraganglional18)heeft deze chemische ablatiebenadering een alternatieve manier geboden om de fysiologische functies van TRPV1 te onderzoeken. We hebben onlangs gemeld dat intracerebroventriculaire (i.c.v.) injectie van RTX het pijnstillende effect van paracetamol bij muizen remt, wat suggereert supraspinal-selectieve TRPV1 desensibilisatie19. In dit manuscript presenteren we het precieze protocol voor i.c.v. injectie en daaropvolgende pijntests.
Directe injectie van drugs in de ventrikels van de hersenen maakt het mogelijk om hun centrale effecten te bestuderen terwijl het minimaliseren van eventuele perifere effecten. De i.c.v. injectieprocedure die hier wordt gepresenteerd is een wijziging van de methode die door Haley en McCormick20wordt gerapporteerd. Deze methode is eenvoudig met het inbrengen van een injectienaald in de laterale ventrikels door de coronale hechting en vereist geen speciale apparatuur of chirurgische ingrepen voor cannulation.
Perifere lokale toepassing van TRPV1 agonisten roept een brandend pijngevoel en neurogene ontsteking op. Muizen die systemisch worden behandeld met RTX en TRPV1-KO muizen, zijn ongevoelig voor deze stimulatie13. We hebben intraplantar injectie van RTX (RTX test) uitgevoerd om het behoud van de perifere TRPV1 in RTX-i.c.v. te bevestigen. Muizen. Er is gemeld dat muizen systemisch behandeld met RTX en TRPV1-KO muizen een normale drempel tonen voor mechanische stimuli11,13,22. Hier presenteren we een procedure voor de staartdruktest voor het testen van veranderingen in het pijnstillende effect van paracetamol.
Al deze procedures zijn orthodox en veelzijdig, en kan worden toegepast op studies van andere drugs.

De voorbijgaande receptor potentieel vanilloid type 1 (TRPV1), een thermogevoelig kation kanaal, is bekend dat pijn in de perifere zenuwen trigger. Procedures worden beschreven voor het testen van nociception aan perifere TRPV1 stimulatie (RTX test) en mechanische stimulatie (staart druk test) dan volgen. Hoewel de nociceptieve reacties van muizen die RTX i.c.v. toegediend waren vergelijkbaar waren met die van de controlegroepen, waren RTX-i.c.v.-toegediende muizen ongevoelig voor het pijnstillende effect van paracetamol, wat suggereert dat i.c.v. RTX-behandeling supraspinale selectieve TRPV1-desensibilisatie kan veroorzaken. Dit muismodel kan worden gebruikt als een handig experimenteel systeem voor het bestuderen van de rol van TRPV1 in de hersenen / supraspinale functie. Het algemene doel van deze voorbehandeling is het genereren van muizen en dat TRPV1-kanalen worden gedesensibiliseerd in supraspinale regio's. Voor de controlegroep, injecteer het voertuig op dezelfde manier.Geef een wegwerp naald van 27 meter door een metalen buis om 3 tot 3,5 millimeter punt van de naald bloot te leggen. Houd vervolgens de squamosale botten van de muis stevig vast met de vingers. Beweeg de naald lateraal op de hoofdhuid en vind de sagittale hechting als de naald tip is verslaafd aan de hechting.Beweeg de tip ongeveer een millimeter naar rechts en beweeg de punt rostrally om de coronale hechting te vinden. Plaats de naald daarna langzaam en verticaal. Injecteer de RTX-oplossing meer dan 10 seconden en houd deze daarna nog 10 seconden vast.Trek de naald vervolgens langzaam terug en breng de muis terug naar zijn thuiskooi. Een week na de RTX-injectie, minstens 60 minuten voor de test, breng je de muis naar de testruimte. Plaats de muis ten minste 30 minuten voor de test in een plexiglaskooi om te kunnen wennen aan het milieu.Dan 20 minuten voor de test, toedien paracetamol op 300 milligram per kilogram aan de muis intraperitoneally. Houd de muis losjes in een kleine doek zak en steek een 30 meter naald in de hiel van de rechter achterpoot. Ga de naald onderhuids naar de buurt van het loopkussen en injecteer 20 microliter RTX-oplossing met 0,05 microgram per milliliter.Meet vervolgens de periode van likken en bijten gedrag in het glabrousgebied van de getroffen poot in elk blok van vijf minuten. Een week na de RTX-injectie, breng de muis naar de testruimte. Plaats de muis in een plexiglas kooi.Markeer de vlekken op 1,5 en 2,5 centimeter van de basis van de staart. Houd vervolgens de muis losjes in een kleine doek zak en druk uitoefenen op de vlekken met een stompe sonde. Houd er rekening mee dat de cutoff druk van 250 gram wordt opgelegd om weefselschade te voorkomen.Bepaal de druk die nodig is om ontsnappen gedrag uit te lokken en bereken de nociceptieve drempel door het gemiddelde van de druk bepaald op de twee plekken. Herhaal de procedures elke 15 minuten. Na het verkrijgen van de basislijn, toedien paracetamol met 300 microgram per kilogram aan de muis intraperitoneally, herhaal dan de staartdruk test om de 15 minuten tot het 90 minuten bereikt.Deze twee cijfers tonen de responsiviteit van SC of ICV behandeld muizen op de intraplantar injectie van RTX. Het likken en bijten gedrag van voertuig-behandelde muizen was opmerkelijk in de eerste 10 minuten. Hoewel de sc-voorbehandelde muizen helemaal geen lik- en bijtgedrag vertoonden, reageerden de voorbehandelde muizen van ICV normaal gesproken op de plantaire injectie van RTX.Bovendien verminderde intraperitoneale toediening van paracetamol het lik- en bijtgedrag van voertuig ICV behandelde muizen, maar niet in RTX ICV behandelde muizen. Deze grafiek toont de pijnstillende effecten van paracetamol met 300 microgram per kilogram in de staartdruktest. Paracetamol verminderde de nociceptieve respons van voorbehandelde muizen in beide tests, maar de pijnstillende effecten van paracetamol werden geremd bij muizen die ICV met RTX voorbehandelden.Eenmaal onder de knie, kan deze ICV injectietechniek worden toegepast voor andere geneesmiddelen. Vergeet niet dat het werken met Resiniferatoxine zeer gevaarlijk kan zijn en zorg ervoor dat rubberen handschoenen en glazen worden gebruikt voor bescherming bij het hanteren.
