Er werd gemeld dat geactiveerde Ras en TGF-β behandeling gedurende 24 uur EndMT in cellen van de MS-1, geïnduceerde terwijl TGF-β alleen mislukte voor het opwekken van EndMT in deze korte periode21. Endotheliale-mesenchymale overgang (EndMT) is het proces waarmee een gedifferentieerde endothelial cel een verscheidenheid van moleculaire veranderingen ondergaat, wat resulteert in een fibroblast-achtige mesenchymale cel1. EndMT werd in eerste instantie beschreven als een Celtransformatie endothelial tijdens de ontwikkeling van het hart2,3. In de vroege ontwikkeling van het hart bestaat de hart-buis uit een binnenste endocard en een buitenste myocard. Deze twee lagen zijn gescheiden door een laag van extracellulaire matrix genaamd de cardiale gelei. De embryonale endocardial cellen, die endothelial cel markeringen verwerven, doorvoer in mesenchymale cellen, de onderliggende cardiale gelei binnenvallen en bevordering van de vorming van de cardiale kussens, de Stichting voorziet in de atrioventriculaire kleppen en tussenschot en de halvemaanvormige kleppen. EndMT is bovendien gesuggereerd als bronnen van pericytes en vasculaire zachte spiercellen in andere embryonale vasculaire systemen met inbegrip van de coronaire bloedvaten, abdominale aorta en longslagader4,5,6. Daarnaast is EndMT betrokken bij de fysiologische angiogenic7kiemen.
Vergaren van bewijs heeft voorgesteld dat de EndMT ook bij meerdere cardiovasculaire aandoeningen en andere ziekten1,8 betrokken is. EndMT-geassocieerde voorwaarden omvatten vasculaire verkalking, atherosclerose, pulmonale arteriële hypertensie, cavernous malformatie, orgel fibrose, ader graft remodelleren, allograft dysfunctie in niertransplantatie en kanker8, 9,10,11,12,13,14,15,16,17, 18. een recent rapport beschreven dat verschillende moleculaire merkers van de EndMT een instrument voor diagnose en prognose-voorspelling van graft renale dysfunctie in nier transplantatie17 zijn kunnen. Modulatie van cellulaire signaalroutes EndMT-gerelateerde is aangetoond dat het verbeteren van de verschillende ziekten met inbegrip van cardiale fibrose en8,15ader graft remodelleren in dierlijke modellen. Daarom is inzicht in de mechanismen onderliggende EndMT belangrijk voor de ontwikkeling van diagnostische en therapeutische strategieën gericht op EndMT.
EndMT wordt gekenmerkt door verlies van cel-cel kruispunten, toename van de trekkende potentieel, Downregulatie van endotheel-specifieke genen zoals VE-cadherine en opregulatie van mesenchymale genen met inbegrip van α-gladde spier actine (α-SMA). Bovendien, EndMT en epitheliale-mesenchymale overgang (EMT), een soortgelijk proces dat epitheliale cellen naar mesenchymale cellen converteert, worden geassocieerd met gewijzigde productie van verschillende componenten van de extracellulaire matrix, die tot de ontwikkeling bijdragen kan van weefsel fibrose8,19.
Onlangs, hebben verscheidene in vitro studies van EndMT details van moleculaire mechanismen van EndMT15,20toegelicht. EndMT wordt veroorzaakt door verschillende signaalroutes inclusief transformeren groeifactor (TGF)-β, Wnt, en Notch1. Onder hen speelt TGF-β cruciaal rollen in de inductie van zowel de EMT en de EndMT. In EndMT, langdurige blootstelling aan TGF-β resultaten in EndMT in verschillende endotheliale cellen, terwijl korte blootstelling lijkt onvoldoende21. We beschreven hier een eenvoudig protocol voor EndMT inductie, waar mijl SVEN 1 (MS-1) muis alvleesklier microvasculaire endotheliale cellen ondergaan EndMT in vitro na langdurige blootstelling aan TGF-β20. In dit model kunnen meerdere stroomafwaartse analyses worden uitgevoerd om te onderzoeken van hallmark kenmerken van EndMT, met inbegrip van morfologische veranderingen, Downregulatie van endothelial markers, opregulatie van mesenchymale merkers en inflammatoire genen, cytoskeletal herschikkingen en collageen gel contractie.
MicroRNAs (miRNAs) zijn ~ 22 nt kleine regelgevende RNAs dat posttranscriptional onderdrukking van verschillende mRNA-doelstellingen-22,23 directe. Door zaad sequentie-gemedieerde doel erkenning, miRNAs onderdrukken van honderden doelgenen en moduleren van diverse cellulaire functies zoals celdifferentiatie, proliferatie en beweeglijkheid. Dit geldt ook voor de verordening van de EMT en EndMT en verschillende miRNAs zijn gemeld als regelgevers van EMT en EndMT24,25. Het model van de EndMT gepresenteerd in dit overzicht kan gemakkelijk worden gecombineerd met miRNA modulatie procedures voor het testen van de rol van miRNAs in EndMT. De huidige herziening bevat een overzicht van onze experimentele procedures om te onderzoeken TGF-β-geïnduceerde EndMT in MS-1 cellen en omvat ook een vergelijking van de voorwaarden van EndMT inductie door TGF-β in andere endotheliale cellen.

molecular analysis of endothelial-mesenchymal transition induced by transforming growth factor-&#946; signaling

Fenotypische plasticiteit van endotheliale cellen ten grondslag ligt aan cardiovasculaire systeemontwikkeling, cardiovasculaire aandoeningen en verschillende voorwaarden in verband met orgel fibrose. In deze voorwaarden verwerven gedifferentieerde endotheliale cellen mesenchymale-achtige fenotypen. Dit proces heet endotheel-mesenchymale overgang (EndMT) en wordt gekenmerkt door Downregulatie van endothelial markers, opregulatie van mesenchymale markeringen en morfologische veranderingen. EndMT wordt veroorzaakt door verschillende signaalroutes, met inbegrip van transformeren groeifactor (TGF)-β, Wnt en Notch, en gereglementeerd door in de moleculaire mechanismen die vergelijkbaar zijn met die van epitheliale-mesenchymale overgang (EMT) belangrijk zijn voor gastrulatie, weefsel fibrose, en uitzaaiing van kanker. Inzicht in de mechanismen van EndMT is belangrijk voor het ontwikkelen van diagnostische en therapeutische methoden gericht op EndMT. Robuuste inductie van EndMT in vitro is nuttig om de gemeenschappelijke gen expressie handtekeningen karakteriseren, druggable moleculaire mechanismen te identificeren en scherm voor modulatoren van EndMT. Hier beschrijven we een in vitro -methode voor de inductie van EndMT. MS-1 muis alvleesklier microvasculaire endotheliale cellen ondergaan EndMT na langdurige blootstelling aan TGF-β en opregulatie van mesenchymale markeringen en morfologische veranderingen als inductie van meerdere inflammatoire chemokines en cytokines tonen. Methoden voor de analyse van microRNA (miRNA) modulatie zijn ook opgenomen. Deze methoden bieden een platform voor het onderzoek naar de mechanismen die ten grondslag liggen aan de EndMT en de bijdrage van miRNAs aan EndMT.

TGF-β is een potente inductor van EndMT in verschillende endotheliale cellen. Na 24 h behandeling met TGF-β in MS-1 cellen toont kleuring voor F-actine reorganisatie van actine stress vezels (figuur 1A)20.
