Mikro-CT bildebehandling kan brukes til å gi svært detaljert og tredimensjonale rekonstruksjoner av vaskulær patologi i dyremodeller. Forekomsten av aorta disseksjon er 3 tilfeller pr. 100 000 per år1. Aorta disseksjon og aneurisme sykdommer står for over 10 000 dødsfall i USA hvert år, sto for 1-2% av alle dødsfall i vestlige land2. Aorta disseksjon startes av en tåre i intimal laget av fartøyet med utbredelsen av blod gjennom lag av aorta veggen under fysiologiske press. Forhøyet pasienten puls presset er assosiert med økt forekomst av disseksjon og komplikasjoner. Økt veggen skjæring stress er forbundet med aorta veggen utvidelse fører til en aneurisme formasjon3,4. Konsekvensene av aorta disseksjon inkluderer okklusjon av blodstrømmen til fjerne organer inkludert hjernen, nyrene, tarmer, og lemmer, dannelsen av kronisk aneurismer, brudd eller død5,6,7.
I dag er biokjemiske og cellulær prosessene involvert i oppstart og progresjon av aortaaneurismer og disseksjoner fremdeles dårlig forstått. Reproduserbar dyremodeller aortaaneurisme og disseksjon er nøkkelen til å forstå deres patofysiologi. Β-aminopropionitrile (BAPN) er en lysyl oksidase, som hindrer cross-linking av elastin og kollagen, og har vist betydelig endre strukturen til fartøyet veggen ekstracellulær matrix og dens biomekaniske integritet6, 8. Gnagere behandlet med BAPN har blitt benyttet som vanlige dyr modell av aortaaneurisme og disseksjon9,10.
Vaskulær tenkelig modaliteter er instrumental i å identifisere vaskulær patologi, bekrefter fartøy patency og evaluere organ perfusjon. Nylig blitt mikro-beregnet tomografi (mikro-CT) benyttet for å studere blodkar mus og tilsvarende størrelse dyr. I motsetning til bein er aksial avbilding av blod fartøy av beregnede tomografi begrenset, som intraluminal blod er iboende relativt radiolucent. Kombinert med intravascular kontrast agenter, men kan mikro-CT detaljert tredimensjonal rekonstruksjoner av dyr vasculatures for studier av makro-anatomisk vaskulær patologi11.
Valgte kontrast agent er (se Tabell for materiale) en kan lett identifiseres silikongummi som inneholder bly chromate og bly sulfat. Aortaaneurisme og disseksjon er knyttet til betydelige sykelighet og dødelighet i befolkningen, og kan være svært dødelige. Mens dyr modeller av aorta sykdom finnes, er i vivo avbilding av er blodkar begrenset. De siste årene, micro-datastyrt tomografi (mikro-CT) har dukket opp som en foretrukket modaliteten for imaging både store og små skip i vivo og ex vivo. I forbindelse med en metode for vaskulær støping, har vi brukt mikro-CT for å karakterisere frekvens og distribusjon av aorta patologi i β-aminopropionitrile-behandlet C57/Bl6 mus. Tekniske begrensninger ved denne metoden inkluderer variasjoner i kvaliteten på perfusjon introdusert av dårlig dyr forberedelse, anvendelsen av riktig metoder for fartøyet størrelse kvantifisering og ikke-survivability av denne prosedyren.
