La maladie d’Alzheimer (ma) est l’une des plus importantes maladies neurodégénératives touchant les personnes âgées dans le monde1. Il est largement admis que l’agrégation anormale de la protéine β-amyloïde (Aß) peut être le principal facteur pathologique d’AD. Agrégats de bêta-amyloïdes sont les principaux composants des plaques séniles, un des marqueurs biologiques dans le cerveau des patients atteints de ma. En outre, agrégats de bêta-amyloïdes, notamment les oligomères provoquent une neurotoxicité puissante, qui pourrait être la cause de la mort neuronale au fur et AD. Par conséquent, l’inhibition de la formation d’oligomères bêta-amyloïdes pourrait empêcher la neurodégénérescence et inhibiteurs d’oligomères bêta-amyloïdes pourraient être développés comme modificateurs traitements d’AD. De nombreuses études ont utilisé un peptide Aβ synthétique pour former des oligomères in vitro, explorer des morphologies et des structures artificielles oligomères bêta-amyloïdes et enquêter sur les inhibiteurs de l’oligomère Aβ utilisant in vitro modèles2,3 , 4. Toutefois, différentes en vitro protocoles de formation d’oligomères bêta-amyloïdes pourraient entraîner oligomère présentant des caractéristiques morphologiques différentes, qui peut entraîner des résultats incomparables entre les différents groupes de recherche. Par conséquent, un protocole standard de formation d’oligomères bêta-amyloïdes est absolument nécessaire.
Jusqu'à présent, pas beaucoup de méthodes ont été signalés à détecter directement les oligomères bêta-amyloïdes. Microscopie électronique à transmission (TEM), non-dénaturisation électrophorèse sur gel, immuno enzymatique (ELISA) et dot blot analyse permet d’examiner la quantité et/ou la morphologie des oligomères bêta-amyloïdes in vitro5, 6. par exemple, la morphologie et la structure de l’oligomère Aβ peuvent être observées en TEM. Les quantités relatives et la taille moléculaire des agrégations d’Aβ pouvaient être mesurés par électrophorèse sur gel non dénaturant. ELISA peut servir à déterminer des oligomères bêta-amyloïdes dans le sérum, le plasma et des extraits de tissus cérébraux. Enfin, dot blot analyse, une technique utilisée pour la détection, analyse et identification des protéines, pourrait servir à évaluer la concentration relative des oligomères bêta-amyloïdes dans différents échantillons à l’aide d’anticorps spécifiques Aβ et oligomère. En outre, une dot blot assay offre gain de temps considérable, comme l’électrophorèse sur gel et les procédures de transfert pour les gels ne sont pas nécessaires. Par conséquent, cette analyse permet normalement d’écran Aβ oligomère des inhibiteurs potentiels. L’objectif général du présent protocole est de décrire une méthode relativement simple, fiable et reproductible pour préparer un échantillon de la riche en oligomères bêta-amyloïdes1-42 , pour analyser les montants de l’oligomère1-42 Aβ par dot blot analyse et oligomère Aβ écran inhibiteurs à l’aide des résultats expérimentaux semi-quantitative.

Β-amyloïde (Aß) est un peptide hydrophobe avec une tendance intrinsèque de s’auto-assembler en agrégats. Parmi les divers agrégats, oligomères bêta-amyloïdes est largement reconnue comme la neurotoxine de premier plan dans la progression de la maladie d’Alzheimer (ma) et est considéré comme l’événement crucial dans la pathogenèse de la ma. Par conséquent, Aβ oligomère inhibiteurs pourraient empêcher la neurodégénérescence et ont le potentiel d’être développé comme modificateurs traitements d’AD. Cependant, protocoles différents formation d’oligomères bêta-amyloïdes pourraient conduire à des oligomères présentant des caractéristiques différentes. En outre, il n’y a pas beaucoup de méthodes pour efficacement écran Aβ1-42 oligomère inhibiteurs. Un anticorps A11 peut réagir avec un sous-ensemble de l’oligomère de1-42 Aß toxique avec des structures de feuillet β antiparallèle. Dans ce protocole, nous décrivons comment préparer un échantillon de riche en oligomères Aβ A11-positif1-42 d’un synthétique Aβ1-42 peptide de vitro et d’évaluer les quantités relatives de l’oligomère1-42 Aβ A11-positives dans les échantillons en une dot-blot analyse à l’aide de l’A11 et Aβ1-42-anticorps 6E10 spécifiques. Utilisant ce protocole, inhibiteurs de l’oligomère1-42 Aβ A11-positif peuvent également projetés de résultats expérimentaux semi-quantitative.

Afin de vérifier si un monomère1-42 de bêta-amyloïdes peut former un oligomère de1-42 Aβ après préparation, analyse TEM a été utilisée.
