PDT is een goed bestudeerde therapie voor de behandeling van kanker sinds het werd meer dan 100 jaar geleden uitgevonden18. Het laatste decennium, PDT is toegepast als een antimicrobiële strategie en effectiviteit tegen sommige antibiotica-resistente ziekteverwekkende bacteriën12heeft aangetoond. Ten opzichte van de planktonische staat, bacteriële biofilms lijken te zijn meer resistent tegen antibiotica behandeling3, terwijl het effect van ALA-PDT...

S. aureus is een belangrijke Gram-positive pathogenen die de huid en mucosa van menselijke gastheren koloniseert. Haar vermogen om vorm biofilms wordt beschouwd als een belangrijk aspect van de pathogenese1. Bacteriële biofilms zijn een Gemeenschap van bacteriën die zijn ingesloten in een zelf geproduceerde matrix, die is samengesteld uit de extracellulaire polymere stoffen, zoals polysacharide, DNA en proteïne. Deze matrix speelt een belangrijke rol in de persistentie van bacteriële infecties, bijgedragen tot een hoge mate van weerstand tegen het menselijke immuunsysteem en de huidige anti-microbiële therapieën2. Antibiotica zijn nog steeds de belangrijke behandeling tegen biofilms infecties, hoewel de effecten van antibiotica op biofilms beperkt zijn. Eerder is aangetoond dat cellen in biofilms 10 - 1000 keer meer resistent zijn tegen antibiotica vergeleken met hun tegenhangers van de planktonische3. Dus, alternatieve strategieën nodig zijn om het veroveren van deze kwestie.
PDT, een alternatieve behandeling van bacteriële infecties, wordt het licht van een passende golflengte om te activeren photosensitizers. Dit leidt tot de productie van reactieve zuurstof soorten (ROS), die dodelijk voor de doelcellen zijn door beschadiging van DNA4, verstoren de celwand en enzymen te inactiveren. Dit kenmerk voor meerdere doelen maakt het moeilijk voor bacteriën ontwikkelen resistentie tegen de behandeling met PDT.
De antimicrobiële effect van PDT op bacteriële en schimmelinfecties biofilms, met meerdere photosensitizers, zoals toluïdine blauw, groen Malachiet, methyleenblauw, chloor e6 en porphyrins, is bestudeerd in vorige verslagen5,6, 7,8,9,10,11,12,13. 5-ALA, een voorstadium van de werkelijke fotosensitizer, PpIX, wordt gekenmerkt door zijn kleine moleculaire gewicht en de snelle goedkeuring12,14. Deze voordelen geven ALA-PDT groot potentieel als een therapeutische toepassing. Hoewel het effect van ALA-PDT op planktonische bacteriën is bestudeerd door vele groepen12, heeft het antimicrobiële effect van ALA-PDT op bacteriële biofilms niet toch geweest opgehelderd. Ondertussen is het moeilijk te vergelijken van de resultaten van eerdere studies. Een van de redenen is dat de verschillende protocollen worden gebruikt door diverse groepen. Dus, dit protocol beschrijft een in vitro model van een ALA-PDT-systeem is gebaseerd op onze eerdere werk15. Het effect van dit model werd bevestigd door CFU berekening en levensvatbaarheid kleuring met CLSM.

Staphylococcus aureus (S. aureus) is een gemeenschappelijke menselijke pathogenen, waardoor pyogenic en systemische infecties. S. aureus -infecties zijn moeilijk uit te roeien niet alleen te wijten aan de opkomst van antibiotica-resistente stammen, maar ook zijn vermogen om vorm biofilms. Fotodynamische therapie (PDT) heeft onlangs, is aangegeven als een van de mogelijke behandelingen voor het beheersen van biofilm infecties. Echter zijn verdere studies nodig om onze kennis van de invloed ervan op bacteriële biofilms, evenals de onderliggende mechanismen te verbeteren. Dit manuscript beschrijft een in vitro model van PDT met 5-aminolevulinic acid (5-ALA), een voorloper van de werkelijke fotosensitizer, protoporfyrine IX (PpIX). Kort, oudere S. aureus biofilms werden geïncubeerd met ALA en vervolgens blootgesteld aan het licht. Vervolgens was het antibacteriële effect van ALA-PDT op S. aureus biofilm gekwantificeerd door het berekenen van de kolonievormende eenheden (CFUs) en gevisualiseerd door levensvatbaarheid fluorescerende vlekken via confocale laser scanning microscopie (CLSM). Representatieve resultaten toonde een sterke antibacteriële werking van ALA-PDT op S. aureus biofilms. Dit protocol is eenvoudig en kan worden gebruikt voor het ontwikkelen van een in vitro model voor het bestuderen van de behandeling van S. aureus biofilms met ALA-PDT. Het kan in de toekomst ook in PDT studies met behulp van andere photosensitizers voor verschillende bacteriestammen met minimale aanpassingen worden verwezen.

De levensvatbaarheid van de bacteriën in de biofilms werd verlaagd na de ALA-PDT behandeling in vergelijking met de besturingselementen (ALA - LED-, ALA + LED- en ALA-LED +) in zowel USA300 als de drie klinische stammen (Figuur 1).
