S. Aureus ist eine wichtige grampositive Erreger, die der Haut und der Schleimhaut des menschlichen Wirte kolonisiert. Seine Fähigkeit, Form Biofilme gilt als einen wichtigen Aspekt ihrer Pathogenese-1. Bakterielle Biofilme sind eine Gemeinschaft von Bakterien, die eingebettet in eine selbst erstellte Matrix, extrazelluläre Polymere Substanzen, einschließlich Polysaccharid, DNA und Proteinen besteht. Diese Matrix spielt eine bedeutende Rolle in der Persistenz von bakteriellen Infektionen, einen Beitrag zu einem hohen Grad des Widerstands gegen das menschliche Immunsystem und aktuellen antimikrobiellen Therapien2. Antibiotika sind noch die großen Behandlung für Biofilm-Infektionen, obwohl die Wirkung von Antibiotika auf Biofilme begrenzt sind. Bisher hat sich gezeigt, dass Zellen in Biofilmen 10 - 1.000 Mal resistenter gegen Antibiotika, die im Vergleich zu ihrer planktonischen Pendants3. Daher sind alternative Strategien erforderlich, um dieses Problem zu erobern.
PDT, eine alternative Behandlung für bakterielle Infektionen, nutzt das Licht einer geeigneten Wellenlänge Photosensibilisatoren aktivieren. Dies führt zu die Produktion von reaktiven Sauerstoffspezies (ROS), die tödlich für Ziel-Zellen sind durch die Zellwand stören, Inaktivierung von Enzymen und Beschädigung DNA4. Diese Multi-Target-Eigenschaft macht es schwierig für Bakterien Resistenz gegen die PDT-Behandlung entwickeln.
Die antimikrobielle Wirkung der PDT auf bakterielle und Pilzinfektionen Biofilme mit mehreren Photosensibilisatoren wie Toluidin blau, Malachitgrün, Methylenblau, Chlor e6 und Porphyrine, wurde in vorherigen Berichten5,6untersucht, 7,8,9,10,11,12,13. 5-ALA, zeichnet sich ein Prodrug von der tatsächlichen Photosensitizer PpIX, durch seine kleinen Molekulargewicht und schnelle Abfertigung12,14. Diese Vorteile geben ALA-PDT großes Potenzial als eine therapeutische Anwendung. Obwohl die Wirkung von ALA-PDT auf planktonischen Bakterien durch viele Gruppen12untersucht wurden, hat die antimikrobielle Wirkung der ALA-PDT auf bakterielle Biofilme noch nicht aufgeklärt. Unterdessen ist es schwierig, die Ergebnisse von früheren Studien zu vergleichen. Einer der Gründe ist, dass die verschiedenen Protokolle von verschiedenen Gruppen genutzt werden. Dieses Protokoll beschreibt also, eine in-vitro- Modell eines ALA-PDT-Systems basierend auf unserer bisherigen Arbeit15. Die Wirkung dieses Modells wurde von KBE Berechnung und Lebensfähigkeit Färbung mit CLSM bestätigt.

Staphylococcus aureus (S. Aureus) ist eine gemeinsame menschliche Erreger, wodurch pyogener und systemische Infektionen. S. Aureus Infektionen sind schwer, nicht nur durch die Entstehung von Antibiotika-resistenten Stämme, sondern auch seine Fähigkeit, Form Biofilme zu beseitigen. Vor kurzem hat die photodynamische Therapie (PDT) als eines der möglichen Behandlungen zur Steuerung von Biofilm-Infektionen angegeben wurde. Allerdings sind weitere Studien erforderlich, um unser Wissen über ihre Wirkung auf bakterielle Biofilme sowie die zugrunde liegenden Mechanismen zu verbessern. Dieses Manuskript beschreibt eine in Vitro -Modell der PDT mit 5-Aminolävulinsäure (5-ALA), ein Vorläufer der eigentlichen Photosensitizer Protoporphyrin IX (PpIX). Kurz, Reifen S. Aureus Biofilme mit ALA inkubiert und dann Licht ausgesetzt waren. Anschließend war die antibakterielle Wirkung der ALA-PDT auf S. Aureus Biofilm durch Berechnung der koloniebildenden Einheiten (KBE) quantifiziert und visualisiert durch Lebensfähigkeit fluoreszierende Färbung durch konfokale Laser-scanning-Mikroskopie (CLSM). Repräsentative Ergebnisse zeigten eine starke antibakterielle Wirkung der ALA-PDT auf S. Aureus Biofilmen. Dieses Protokoll ist einfach und kann zur Entwicklung eines in-vitro- Modells um die Behandlung von S. Aureus Biofilme mit ALA-PDT zu studieren. In Zukunft könnte es auch in PDT Studien unter Verwendung anderer Photosensibilisatoren für verschiedene Bakterienstämme mit minimalen Anpassungen verwiesen werden.

Die Lebensfähigkeit der Bakterien in den Biofilmen war zurückgegangen, nachdem ALA-PDT-Behandlung im Vergleich zu den Kontrollen (ALA - LED, ALA + LED- und ALA-LED +) in USA300 und die drei klinische Stämme (Abbildung 1).
