murine distal colostomy, a novel model of diversion colitis in c57bl/6 mice

تسريب التهاب القولون (DC) حالة سريرية متكررة التي تحدث في المرضى مع قطاعات الأمعاء مستبعدة من مجرى البراز نتيجة تحويل انتيروستومي. مسببات هذا المرض ما زال غير محددة، لكن يبدو أن تختلف عن تلك الأمراض الكلاسيكية التهاب الأمعاء مثل مرض كرون والتهاب القولون التقرحي. البحوث الرامية إلى فك الآليات الفيزيولوجية المرضية التي تؤدي إلى تطور هذا المرض تعطلت بعدم وجود نموذج مورين مناسب. ينشئ هذا البروتوكول نموذج مورين من العاصمة التي تسهل دراسة دور النظام المناعي وتفاعله مع ميكروبيومي في وضع DC. في هذا النموذج باستخدام الحيوانات C57BL/6، تستبعد الأجزاء البعيدة من القولون من تيار البراز بإنشاء كولوستومي القاصي وأحداث تنمية خفيفة إلى معتدلة التهاب في قطاعات الأمعاء المستبعدة واستنساخ الآفات السمة المميزة للإنسان العاصمة مع استجابة التهابات الجهازية معتدلة. على النقيض من نموذج الفئران، تتوفر عدد كبير من نماذج مورين معدلة وراثيا على خلفية C57BL/6. المزيج من هذه الحيوانات مع نموذجنا يسمح الأدوار المحتملة للفرد السيتوكينات أو المستقطبات أو مستقبلات الجزيئات النشطة بيولوجيا (مثلاً، انترلوكين (إيل)-17؛ IL-10، تشيموكيني CXCL13، ومستقبلات تشيموكيني CXCR5 و CCR7، ومستقبلات سفينجوسيني-1-فوسفات 4) إلى تقييم في الآلية المرضية للعاصمة. توافر سلالات الماوس كونجينيك على خلفية C57BL/6 يسهل إلى حد كبير نقل التجارب تحديد دور كل من أنواع الخلايا المتميزة المشاركة في المسببات للعاصمة. وأخيراً، يقدم النموذج الفرصة لتقييم تأثيرات التدخلات المحلية (مثلاً، تعديل ميكروبيومي المحلية أو العلاج للالتهابات المحلية) على حصانة المخاطي في قطاعات الأمعاء المتضررة وغير المتضررة وفي التوازن المناعي الجهازية.
