EGCs spelen een belangrijke rol in de homeostase van de darm, en het is essentieel om te isoleren en bestuderen ze in vitro. In dit protocol, een eenvoudige methode voor het isoleren van EGCs uit de lamina propria van de volwassen muis darm geïntroduceerd om te bestuderen van de darmen gliale functie.

Interesse in de functie van de darmen gliale cellen (EGCs) is gestaag toegenomen als gevolg van hun erkende rollen in de darm integriteit en homeostase1,2. EGCs is bovendien afhankelijk van hun locatie langs de lengte van de GI tract3,4. EGCs release verschillende trofische factoren, met inbegrip van gliale cel-afgeleide neurotrophic factor (GDNF), bijdragen tot de darm motiliteit1,5 en reageren op microbiële bijproducten6,7. Studies hebben aangegeven dat de EGC populatie heterogeen is en dat hun functie is afhankelijk van of ze submucosal zijn of zich bevinden binnen de myenteric plexus1,7. Bijvoorbeeld, bijdragen EGCs binnen de submucosa aan strakke kruispunten8. Differentiële expressie van het algemeen kaderakkoord voor vrede en fosforylatie in EGCs zijn gekoppeld aan de ziekte van Parkinson, suggereren hun mogelijke link naar het fenotype van de darm van deze stoornis9. De uitdaging op het gebied blijft hoe te isoleren en bestuderen een of beide EGC populaties in vitro. Lineage trace experimenten aangetoond dat EGCs in de submucosa en de lamina propria zijn afkomstig uit voorlopercellen in de plexus myenteric7. Hoewel er verschillende gepubliceerde isolatie protocollen die beschikbaar zijn voor het genereren van de culturen van myenteric EGCs12,13,14,15,16,17, 18,19, geen specifiek gericht op isolatie van de submucosal/lamina propria EGC bevolking. Bestaande protocollen voor EGC isolatie meer specifiek gebruiken een combinatie van de mechanische scheiding of de microdissection van de gladde spieren gecombineerd met enzymatische dissociatie, uiteindelijk de mucosal cellaag teruggooi.
Het doel van dit manuscript is om aan te tonen de stappen voor het niet-enzymatisch isoleren primaire EGCs uit de lamina propria voor in vitro studies. Aangezien er geen markeringen die specifiek onderscheid myenteric EGCs in de submucosa, was de ruimtelijke scheiding van de epitheliale mucosa van de gladde spieren benut om submucosal EGCs isoleren. De darmen zenuwstelsel (ENS) bestaat uit neuronen en darmen gliale cellen (EGCs) die zich binnen de gladde spieren muur, de submucosa en de lamina propria bevinden. EGCs spelen een belangrijke rol in de homeostase van de darm door de vrijlating van verschillende trofische factoren en bijdragen tot de integriteit van de epitheliale barrière. De meeste studies van primaire darmen gliale culturen cellen geïsoleerd van de plexus myenteric na enzymatische dissociatie gebruiken. Hier, wordt een niet-enzymatische methode te isoleren en cultuur EGCs van de intestinale submucosa en de lamina propria beschreven. Na handmatige verwijdering van de Musculus longitudinalis laag, werden EGCs bevrijd van de lamina propria en de submucosa, met behulp van opeenvolgende HEPES-buffer EDTA incubations gevolgd door incubatie in de oplossing van de gegevensterugwinning van de verkrijgbare niet-enzymatische cel. Hier beschrijven we de isolatie van darmen-gliale cellen uit de intestinale submucosa-sequentiële EDTA incubations met divalente kationen en vervolgens incubatie in de oplossing van de gegevensterugwinning van de niet-enzymatische cel chelaat. De resulterende celsuspensie op poly-D-lysine en laminin plating resulteert in een sterk verrijkt cultuur van submucosal gliacellen voor functionele analyse.
