EGCs spille viktige roller i tarmen homeostase, og det er viktig å isolere og studere dem i vitro. I denne protokollen, ble en enkel metode for å isolere EGCs fra lamina propria av voksen mus tarmen introdusert for å studere enteric glial funksjon.

Interesse i funksjonen for enteric gliacellene (EGCs) har stadig økt på grunn av deres anerkjente roller i tarmen integritet og homeostase1,2. I tillegg varierer EGCs plasseringen langs GI spor3,4. Nylig ble det observert at tapet av kjernefysiske protein menin i isolerte kulturer av EGCs fra den proksimale intestinal submucosa var tilstrekkelig til å indusere uttrykk av hormonet gastrin10. Utfordringen i feltet forblir isolere og studere en eller begge EGC populasjoner i vitro. Linjen spore eksperimenter har vist at EGCs i submucosa og lamina propria stammer fra progenitor celler i myenteric plexus7. Selv om det er flere publiserte isolasjon protokoller tilgjengelig å generere kulturer av myenteric EGCs12,13,14,15,16,17, 18,19, ingen mål spesifikt isolasjon av submucosal/lamina propria EGC befolkningen. Eksisterende protokoller EGC isolering bruker spesifikt en kombinasjon av mekanisk separasjon eller microdissection av den glatte muskulaturen kombinert med enzymatisk dissosiasjon, til slutt forkaster mucosal celle laget.
Målet med dette manuskriptet er å vise trinnene for å isolere ikke-enzymatisk primære EGCs fra lamina propria for i vitro studier. Siden det er ingen indikatorer som spesielt skiller myenteric EGCs fra de i submucosa, utnyttes romlige separasjon epithelial mucosa fra glatt muskel for å isolere submucosal EGCs. Innstigning nervesystemet (ENS) består av nevroner og enteric gliacellene (EGCs) som ligger i glatt muskel veggen, submucosa og lamina propria. EGCs spille en viktig rolle i tarmen homeostase gjennom utgivelsen av ulike trophic faktorer og bidra til integriteten til epithelial barrieren. De fleste studier av primære enteric glial kulturer bruke celler isolert fra myenteric plexus etter enzymatisk dissosiasjon. Her, er en ikke-enzymatisk metode for å isolere og kultur EGCs fra intestinal submucosa og lamina propria beskrevet. Etter manuell fjerning av langsgående muskel laget, ble EGCs frigjort fra lamina propria og submucosa med sekvensiell HEPES-bufret EDTA incubations etterfulgt av inkubasjon i kommersielt tilgjengelige ikke-enzymatisk celle gjenopprettingsløsning.
