Tendinopatía es una condición músculo-esquelética degenerativa crónica que se desarrolla en varias partes del cuerpo. Recientemente, el número de casos de tendinopathy ha aumentado considerablemente en el mundo desarrollado debido a la creciente participación en los deportes recreativos y mayor esperanza de vida1,2. La causa de la Tendinopatía es considerada multifactorial y estas causas incluyen la isquemia, lesiones de radicales libres de oxígeno, un desequilibrio entre inervaciones vasoconstrictor y vasodilatador, interno micro-se rasga y los cambios neuro-regulación3 ,4,5,6,7,8. Mayoría de los tratamientos para tendinopathy sólo aliviar sus síntomas. Además, tratamientos sin regeneración de los tejidos requieren mucho tiempo para la rehabilitación y logran una respuesta limitada de tendones lesionados, que impone un desafío clínico para los médicos9.
La incompetencia de las opciones actuales de tratamiento junto con la falta de capacidad de tendón degenerativo de self-heal tiene investigadores a tomar interés en explorar estrategias alternativas del tratamiento. Recientemente, nuevos estudios registrados muchos resultados prometedores para la mejora de la eficacia curativa de los tendones Tendinopatía mediante factores de crecimiento, células madre terapia y terapia de gene10,11,12.
A través de una revisión de la literatura, encontramos que los estudios implicados pueden dividirse en dos categorías basadas en sus análisis de materiales: modelos animales como una rata, un ratón o un conejo; y modelos humanos. En relación con el modelo animal, actualmente existen dos técnicas populares para generar tendinopathy: inducción química de lesión o sobrecarga del modelo mecánico. Sin embargo, cada modelo animal fue limitado en la reproducción de la Tendinopatía humana compleja patología13,14.
Mayoría de los papeles con muestras humanas se analizaron histológicamente o realiza el en vitro experimento basado en un tenocyte humano sano en lugar de una tenocyte degenerativa15,16,17,18 , 19 , 20 , 21. sólo unos pocos trabajos registrados que usaron una tenocyte degenerativa humana, pero no describió en detalle el protocolo usado para obtener la tenocyte degenerativa de la humana22,23. En este contexto, cabe señalar que resultados exitosos en el modelo animal o el tejido sano/tenocyte no pueden necesariamente predecir eficacia humana o dosificación eficaz debido a la degeneración del tendón es un proceso complicado y la patogenesia es aún no completamente entendido.
En conjunto, es necesario describir el protocolo estándar para la obtención de la tenocyte degenerativa del tejido humano sin causar efectos adversos al donante. Este artículo describe un protocolo sobre cómo adquirir la tenocyte degenerativa humana. Para validar el protocolo, los tejidos cosechados fueron analizados histológicamente. Entonces, el cultivo de células fue confirmado como la tenocyte degenerativa por usar inmunocitoquímica (ICC), una reacción en cadena de polimerasa de tiempo real cuantitativa (qRT-PCR) y un análisis de viabilidad.

Tendinopatía, una condición dolorosa que se desarrolla en respuesta a la degeneración del tendón, va en aumento en los países desarrollados debido al aumento de la actividad física y mayor esperanza de vida. A pesar de su creciente prevalencia, la patogenia subyacente sigue siendo confusa, y el tratamiento es generalmente sintomático. Recientemente, se investigaron numerosas opciones terapéuticas, incluyendo factores de crecimiento, células madre y terapia génica, con la esperanza de mejorar la potencia curativa del tendón degenerativo. Sin embargo, la mayoría de estos estudios se llevaron a cabo sólo en modelos animales o tenocytes humana saludable. A pesar de algunos estudios utilizando tenocytes patológica, al mejor de nuestro conocimiento no existe ningún protocolo que describe cómo obtener tenocytes degenerativa humana. El objetivo de este estudio es describir un protocolo estándar para la adquisición de tenocytes degenerativa humana. Inicialmente, el tejido del tendón fue cosechado de un paciente con epicondilitis lateral durante la cirugía. Luego se tomaron muestras de biopsia del tendón extensor carpi radialis brevis correspondiente a los cambios estructurales observados en el momento de la cirugía. Todos los tendones cosechados parecen opaco, gris, friable y edematosa, que hacía visualmente distinta de los sanos. Tenocytes eran cultivados y utilizados para experimentos. Mientras tanto, la mitad de los tejidos cosechados fueron analizada histológicamente, y se ha demostrado que compartían las mismas características claves de Tendinopatía (angiofibroblastic displasia o hiperplasia). Un análisis secundario por immunocytochemistry confirmó que las células cultivadas tenocytes con la mayoría de las células que tienen las manchas positivas para mohawk y tenomodulin las proteínas. Luego se determinaron las cualidades de la naturaleza degenerativa de tenocytes comparando las células con el control sano con un ensayo de proliferación o qRT-PCR. La tenocyte degenerativa muestra una mayor tasa de proliferación y similares patrones de expresión de gene de la Tendinopatía que coincidió con los informes anteriores. En general, este nuevo protocolo podría proporcionar una herramienta útil para futuros estudios de Tendinopatía.

Análisis histológicos revelaron que el tejido recolectado de epicondilitis lateral tenía las características de un tendón de tendinopathic. Sección H & E de Tendinopatía degenerativa del tendón reveló un paquete de colágeno desorganizado con pérdida de polaridad y estructuras bien recto, fuertemente lleno de fibra paralela.
